    Description / Table of Contents: Two novel cyclodepsipeptides, scopularides A and B, were found in the fungus Scopulariopsis brevicaulis, which was isolated from the marine sponge Tethya aurantium. In addition, the known fungal metabolite paxilline was identified. The structures of the scopularides were elucidated by NMR, MS, and chemical derivatization methods as cyclo-(3-hydroxy-4-methyldecanoyl-Gly-L-Val-D-Leu-L-Ala-L-Phe) and cyclo-(3-hydroxy-4-methyloctanoyl-Gly-L-Val-D-Leu-L-Ala-L-Phe) for scopularide A and B, respectively. Antibiotic activity against Gram-negative bacteria was absent and against Gram-positive bacteria was weak, but activity against several tumor cell lines was significant at 10 [my]g/mL.

    Description: The distribution of methane and hydrogen sulfide concentrations in sediments of various basins of the Baltic Sea was investigated during 4 cruises in 1995 and 1996. Significant differences in the concentrations of both compounds were recorded between the basins and also between different areas within the Gotland Deep. High-methane sediments with distinctly increasing concentrations from the surface to deeper layers were distinguished from low-methane sediments without a clear gradient. Methane concentrations exhibited a fair correlation with the sediment accumulation rate, determined by measuring the total thickness of the post-Ancylus Holocene sequence on echosounding profiles in the Gotland Deep. Only weak correlations were observed with the content of organic matter in the surface layers of the sediments. Hydrogen sulfide concentrations in the sediments showed a positive correlation with methane concentrations, but, in contrast to methane concentrations, were strongly influenced by the transition from oxic to anoxic conditions in the water column between 1995 and 1996. Sediments in the deepest part of the Gotland Basin (〉237 m water depth), covering an area of approximately 35 km**2, were characterized by especially high accumulation rates (〉70 cm/ka) and high methane and hydrogen sulfide contents. Concentrations of these compounds decreased rapidly towards the slope of the basin.

    Description: An ultrastructural investigation was made of the symbiotic bacteria in the gills of the bivalve Thyasira sarsi from the Central Skagerrak. The bacteria were characterized by sulphur globules in the periplasmic space and by peripheral intracellular membrane stacks. Apart from the membrane system they were very similar to previously described sulphur-oxidizing symbionts of the same species from Bergen, Norway. Stable carbon isotope data and lack of C1 -metabolism enzymes gave additional evidence for sulphur-oxidizing symbionts in T. sarsi in the Skagerrak, too.

    Description: A new member of the Flavobacteriales was isolated from the surface of a stone collected on the German North Sea shore. The bacterium, strain ANORD5T, is a mesophilic, chemoheterotrophic aerobic, typical marine bacterium. Optimal growth was observed at 20–30 °C, pH 7.0–8.5 and 1–2 % sea salt. The 16S rRNA gene sequence revealed a distant relationship with the representatives of the Cryomorphaceae , with less than 90 % sequence similarity. Strain ANORD5T forms a cluster together with Owenweeksia hongkongensis UST20020801T (89.9 %), Cryomorpha ignava 1-22T (87.9 %), Luteibaculum oceani CC-AMWY-103BT (88.1 %) and Phaeocystidibacter luteus PG2S01T (87.3 %). Strain ANORD5T has a low DNA G+C content (31 mol%). Based on morphological, physiological and phylogenetic data, strain ANORD5T is considered a type strain of a new species and a new genus of the family Cryomorphaceae for which the name Vicingus serpentipes is proposed. The type strain is ANORD5T (=NCIMB 15042T=DSM 103558T=MTCC 12686T).

    Description: Methane seeps in shallow waters in the northern Kattegat off the Danish coast form spectacular submarine landscapes - the 'bubbling reefs' - due to carbonate-cemented sandstone structures which are colonized by brightly coloured animals and plants. These structures may be 100 m2 in area and consist of pavements, complex formations of overlying slab-type layers, and pillars up to 4 m high. The carbonate cement (high-magnesium calcite, dolomite or aragonite) is 13C-depleted, indicating that it originated as a result of microbial methane oxidation. It is believed that the cementation occurred in the subsurface and that the rocks were exposed by subsequent erosion of the surrounding unconsolidated sediment. The formations are interspersed with gas vents that intermittently release gas, primarily methane, at up to 25 1 h-' The methane most likely originated from the microbial decomposition of plant material eposited during the Eemian and early Weichselian periods, i.e. l00 000 to 125 000 years B.P. Aerobic methane oxidation in the sediment was restricted Lo the upper 4 cm in muddy sand and to the upper 13 cm In coarse sand. Maximum aerobic methane oxidation rates ranged from 4.8 to 45.6 pm01 dm-3 d". The rock surfaces and epifauna around the seeps were also sites of methane-oxidizing activity. Integrated sulphate reduction rates for the upper 10 cm of muddy sand gave 4.2 to 26.6 mm01 m-2 d-' These rates are higher than those previously reported from similar water depths in the Kattegat but did not relate to the sediment methane content. Since gas venting occurs over several km2 of the sea floor in the Kattegat it is likely to make a significant local contribution to the cycling of elements in the sediment and the water column. The rocks support a diverse ecosystem ranging from bacteria to macroalgae and anthozoans. Many animals live within the rocks in holes bored by sponges, polychaetes and bivalves. Stable carbon isotope composition (6'") of tissues of invertebrates from the rocks were in the range -17 to -24 'A, indicating that methane-derived carbon makes little direct contribution to their nutrition. Within the sediments surrounding the seeps there is a poor metazoan fauna, in terms of abundance, diversity and biomass. This may be a result of toxicity due to hydrogen sulphide input from the gas.

    Description: In the frame of studies on secondary metabolites produced by fungi from deep-sea environments we have investigated inhibitors of enzymes playing key roles in signaling cascades of biochemical pathways relevant for the treatment of diseases. Here we report on a new inhibitor of the human protein tyrosine phosphatase 1B (PTP1B), a target in the signaling pathway of insulin. A new asperentin analog is produced by an Aspergillus sydowii strain isolated from the sediment of the deep Mediterranean Sea. Asperentin B (1) contains an additional phenolic hydroxy function at C-6 and exhibits an IC50 value against PTP1B of 2 μM in vitro, which is six times stronger than the positive control, suramin. Interestingly, asperentin (2) did not show any inhibition of this enzymatic activity. Asperentin B (1) is discussed as possible therapeutic agents for type 2 diabetes and sleeping sickness.

    Description: A novel spirocyclic drimane coupled by two drimane fragment building blocks 2 and a new drimane 1 were identified in mycelia and culture broth of Stachybotrys sp. MF347. Their structures were established by spectroscopic means. This is the first example of spirocyclic drimane coupled by a spirodihydrobenzofuranlactam unit and a spirodihydroisobenzofuran unit; and the connecting position being N-C instead of an N and N connecting unit. Strain MF347 produced also the known spirocyclic drimanes stachybocin A (12) and stachybocin B (11) featured by two sesquiterpene-spirobenzofuran structural units connected by a lysine residue; the known spirocyclic drimanes chartarlactam O (5); chartarlactam K (6); F1839A (7); stachybotrylactam (8); stachybotramide (9); and 2α-acetoxystachybotrylactam acetate (10); as well as ilicicolin B (13), a known sesquiterpene. The relative configuration of two known spirobenzofuranlactams (3 and 4) was determined. All compounds were subjected to biological activity tests. The spirocyclic drimane 2, 11, and 12, as well as the sesquiterpene 13, exhibited antibacterial activity against the clinically relevant methicillin-resistant Staphylococcus aureus (MRSA).

    Description: Micromonospora sp. strain DB620 was isolated from a Wadden Sea sediment sample collected near Büsum (Germany) and is closely related (99% 16S-rRNA gene sequence similarity) to Micromonospora coxensis strain MTCC8093. It produced a new polyene dicarboxylic acid named diacidene (1) and in addition a derivative of chorismic acid, the known 3-[(1-carboxyvinyl)oxy]benzoic acid. The structure elucidation of 1 was achieved by applying different 1D and 2D NMR techniques as well as mass spectrometry and UV spectroscopy.
