    Description: The rate of discovery of reefs of the cold-water coral Lophelia pertusa (Linnaeus, 1758) has been remarkable, and attributable to the increased use of underwater video. These reefs form a major three-dimensional habitat in deeper waters where little other ‘cover’ for fish is available. They are common in the eastern North Atlantic, and occur at least in the western North Atlantic and off central Africa. There are also other non-reef records of Lophelia in the Atlantic, and in Indian and Pacific oceans. Thus, not only are these reefs a significant habitat on a local scale, but they may also provide an important habitat over a very wide geographic scale. The present study examined the association of fish species with Lophelia in the Northeast Atlantic, including the Trondheimsfjord and Sula Ridge in Norway, Kosterfjord in Sweden, Darwin Mounds west of Scotland, and Rockall Bank, Rockall Trough and Porcupine Seabight off Ireland. The fish fauna associated with a shipwreck west of Shetland was also studied. Data were collected from 11 study sites at 8 locations, using 52 hours of video and 15 reels of still photographs. Video and still photographs were collected from (1) manned submersible, (2) surface controlled remotely operated vehicle (ROV), (3) a towed “hopper” camera, (4) wide angle survey photography (WASP), (5) seabed high resolution imaging platform (SHRIMP), and (6) an in situ time-lapse camera “Bathysnap”. It was possible to identify 90 % of fish observed to species level and 6.5 % to genus or family level. Only 3.5 % of the fish were not identifiable. Twenty-five species of fishes from 17 families were recorded over all the sites, of which 17 were of commercial importance and comprised 82 % of fish individuals observed. These commercial fish species contribute 90 % of commercial fish tonnage in the North Atlantic. The habitats sampled were comprised of 19 % reef, 20 % transitional zone (i.e. between living coral and debris zone), 25 % coral debris and 36 % off-reef seabed. Depth was the most significant parameter in influencing the fish associated with the reefs, both at the species and family level. There was a complete separation of sites above and below 400–600 m depth by multi-dimensional scaling (MDS) analysis. Less distinct assemblages of fish species were associated with each habitat. Fish species richness and abundance was greater on the reef than surrounding seabed. In fact, 92 % of species, and 80 % of individual fish were associated with the reef. The present data indicates that these reefs have a very important functional role in deep-water ecosystems as fish habitat.

    Description: ABSTRACT Mass spectrometry-based phosphoproteomics provides a unique unbiased approach to evaluate signaling network in cancer cells. The tyrosine kinase inhibitor sunitinib is registered as treatment for patients with renal cell cancer (RCC). We investigated the effect of sunitinib on tyrosine phosphorylation in RCC tumor cells to get more insight in its mechanism of action and thereby to find potential leads for combination treatment strategies. Sunitinib inhibitory concentrations of proliferation (IC50) of 786-O, 769-p and A498 RCC cells were determined by MTT-assays. Global tyrosine phosphorylation was measured by LC-MS/MS after immunoprecipitation with the antiphosphotyrosine antibody p-TYR-100. Phosphoproteomic profiling of 786-O cells yielded 1519 phosphopeptides, corresponding to 675 unique proteins including 57 different phosphorylated protein kinases. Compared to control, incubation with sunitinib at its IC50 of 2µM resulted in downregulation of 86 phosphopeptides including CDK5, DYRK3, DYRK4, G6PD, PKM and LDH-A, while 94 phosphopeptides including Axl, FAK, EPHA2 and p38α were upregulated. Axl- (y702), FAK- (y576) and p38α (y182) upregulation was confirmed by Western Blot in 786-O and A498 cells. Subsequent proliferation assays revealed that inhibition of Axl with a small molecule inhibitor (R428) sensitized 786-O RCC cells and immortalized endothelial cells to sunitinib up to 3 fold. In conclusion, incubation with sunitinib of RCC cells causes significant upregulation of multiple phosphopeptides including Axl. Simultaneous inhibition of Axl improves the antitumor activity of sunitinib. We envision that evaluation of phosphoproteomic changes by TKI treatment enables identification of new targets for combination treatment strategies. This article is protected by copyright.     Description: U-series age patterns obtained on reef framework-forming cold-water corals collected over a nearly 6000-km-long continental margin sector, extending from off Mauritania (17{degrees}N; northwest Africa) to the southwestern Barents Sea (70{degrees}N; northeastern Europe), reveal strong climate influences on the geographical distribution and sustained development of these ecosystems. Over the past three glacial-interglacial cycles, framework-forming cold-water corals (Lophelia pertusa and Madrepora oculata) seem to have predominantly populated reefs, canyons, and patches in the temperate East Atlantic and the Mediterranean Sea. Above 50{degrees}N corals colonize reefs in the northern East Atlantic primarily during warm climate periods with the biogeographic limit advancing from [~]50{degrees}N to [~]70{degrees}N. We propose that north-south oscillations of the biogeographic limit of reef developments are paced by ice ages and may occur synchronously with north-south displacement of cold nutrient-rich intermediate waters and surface productivity related to changes of the polar front.

    Description: We present a comprehensive regional bathymetric data compilation for the southwest Indian Ocean (swIOBC) covering the area from 4°S to 40°S and 20°E to 45°E with a spatial resolution of 250 m. For this, we used multibeam and singlebeam data as well as data from global bathymetric data compilations. We generated the swIOBC using an iterative approach of manual data cleaning and gridding, accounting for different data qualities and seamless integration of all different kinds of data. In comparison to existing bathymetric charts of this region the new swIOBC benefits from nearly four times as many data-constrained grid cells and a higher resolution, and thus reveals formerly unseen seabed features. In the central Mozambique Basin a surprising variety of landscapes were discovered. They document a deep reaching influence of the Mozambique Current eddies. Details of the N-S trending Zambezi Channel could be imaged in the central Mozambique Basin.

    Description: Subpolar regions are key areas to study natural climate variability, due to their high sensitivity to rapid environmental changes, particularly through sea surface temperature (SST) variations. Here, we have tested three independent organic temperature proxies (UK'37, TEX86 and LDI) on their potential applicability for SST reconstruction in the subpolar region around Iceland. UK'37, TEX86 and TEXL86 temperature estimates from suspended particulate matter showed a substantial discrepancy with instrumental data, while long chain alkyl diols were below detection limit in most of the stations. In the northern Iceland Basin, sedimenting particles revealed a seasonality in lipid fluxes i.e. high fluxes of alkenones and GDGTs were measured during late spring-summer, and high fluxes of long chain alkyl diols during late summer. The flux-weighted average temperature estimates had a significant negative (ca. 2.3°C for UK'37) and positive (up to 5°C for TEX86) offset with satellite-derived SSTs and temperature estimates derived from the underlying surface sediment. UK'37 temperature estimates from surface sediments around Iceland correlate well with summer mean sea surface temperatures, while TEX86 derived temperatures correspond with both annual and winter mean 0-200 m temperatures, suggesting a subsurface temperature signal. Anomalous LDI-SST values in surface sediments, and low mass flux of 1,13- and 1,15-diols compared to 1,14-diols, suggest that Proboscia diatoms are the major sources of long chain alkyl diols in this area rather than eustigmatophyte algae, and therefore the LDI cannot be applied in this region.

    Description: Here we present a case study of three cold-water coral mounds in a juvenile growth stage on top of the Pen Duick Escarpment in the Gulf of Cadiz; Alpha, Beta and Gamma mounds. Although cold-water corals are a common feature on the adjacent cliffs, mud volcanoes and open slope, no actual living cold-water coral has been observed. This multidisciplinary and integrated study comprises geophysical, sedimentological and (bio)geochemical data and aims to present a holistic view on the interaction of both environmental and geological drivers in cold-water coral mound development in the Gulf of Cadiz. Coring data evidences (past or present) methane seepage near the Pen Duick Escarpment. Several sources and pathways are proposed, among which a stratigraphic migration through uplifted Miocene series underneath the escarpment. The dominant morphology of the escarpment has influenced the local hydrodynamics within the course of the Pliocene, as documented by the emplacement of a sediment drift. Predominantly during post-Middle Pleistocene glacial episodes, favourable conditions were present for mound growth. An additional advantage for mound formation near the top of Pen Duick Escarpment is presented by seepage-related carbonate crusts which might have offered a suitable substrate for coral settling. The spatially and temporally variable character and burial stage of the observed open reef frameworks, formed by cold-water coral rubble, provides a possible model for the transition from cold-water coral reef patches towards juvenile mound. These rubble "graveyards" not only act as sediment trap but also as micro-habitat for a wide range of organisms. The presence of a fluctuating Sulphate-Methane Transition Zone has an important effect on early diagenetic processes, affecting both geochemical and physical characteristics, transforming the buried reef into a solid mound. Nevertheless, the responsible seepage fluxes seem to be locally variable. As such, the origin and evolution of the cold-water coral mounds on top of the Pen Duick Escarpment is, probably more than any other NE Atlantic cold-water coral mound province, located on the crossroads of environmental (hydrodynamic) and geological (seepage) pathways.
