    Notizen: Summary The inhibitory effect of roquefortine, a secondary metabolite of Penicillium roqueforti, on bacterial protein, RNA and DNA synthesis was studied. Similar results were obtained in colorimetric measurements and in studying the incorporation of radioactive precursors. They show that RNA synthesis was most significantly affected by roquefortine. Inhibition of protein and DNA synthesis was less pronounced and might be a result of primary inhibition of RNA synthesis.

    Notizen: Summary Mycelia of Claviceps purpurea CBS 164.59 which produce clavine alkaloids and ergometrine were immobilized in different matrices. These mycelia exhibited a high sensitivity to extreme physical and chemical treatment, and only a moderate immobilization in calcium alginate proved to be the appropriate method to yield high alkaloid amounts. The alkaloid production is correlated to the amount of biomass. The patterns of alkaloid yields of 4% calcium alginate immobilized cells, compared to free cells, were reduced but not shifted in their maximum. With increasing calcium alginate concentrations (5%–8%) an increase of the alkaloid production even higher than that of free mycelia could be observed. Immobilization of mycelia in 8% calcium alginate resulted in a 35% increase (mg alkaloids/l medium) in overall alkaloid production. Because of reduced O2-diffusion into the beads, high calcium alginate concentrations induced a shift in the clavine alkaloid production: 8% calcium alginate immobilized cells produced agroclavine as main product whereas free cells oxidized agroclavine with molecular oxygen to produce elymoclavine and excreted this alkaloid as main product.

The growth of gram-positive organisms containing hemins was inhibited, whereas Lactobacteria and Clostridia which lack these hemins were only impaired. The growth of gram-negative cells was not affected; therefore, it is supposed that there is no transport of roquefortine into these cells.

    Notizen: Summary Mycelia of Claviceps purpurea CBS 164.59 were immobilized in 2%, 4%, and 8% calcium alginate. Alkaloid production by free cells declined after 60 days, while immobilized cells retained their activity for 200 days. The cumulative alkaloid production for all fermentation cycles using 8% calcium alginate immobilized mycelia was 25 times higher than that from free cells. The best yields of the ergopeptide ergometrine were reached with 4% gel immobilized mycelia, while higher gel concentrations caused a shift in the alkaloid biosynthesis towards high clavine alkaloid production. Beginning with the third cycle of reincubation the immobilized mycelia showed a marked tendency to fragmentize into vacuolated arthrosporoid-like structures and produced violet-black pigments so that the beads recalled sclerotial structures of parasitically living Claviceps.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung MitBacillus stearothermophilus als Testkeim wurde ein Plattendiffusionstest zur quantitativen Roquefortinbestimmung ausgearbeitet, mit dem Konzentrationen von 25 μg/ml noch erfaßt werden können. Im Vergleich zu einer Fluorescenzmeßmethode ist der Test weniger empfindlich, aber ebenso exakt und in seiner Handhabung ungleich einfacher, so daß der biologische Test als zusätzliche Meßmethode angesehen werden kann.
    Notizen: Summary A plate diffusion assay for quantitative determination of roquefortine withBacillus stearothermophilus as test organism was developed. With this assay concentrations of 25 μg/ml could still be measured. In comparison with a fluorescence method this test is less sensitive, but just as exact and much easier to apply, so that the biological assay can be considered as an additional determination.
