    Notes: As part of an ongoing examination of microbial diversity associated with hydrothermal vent polychaetes of the family Alvinellidae, we undertook a culture-independent molecular analysis of the bacterial assemblage associated with mucous secretions of the Northeastern Pacific vent polychaete Paralvinella palmiformis. Using a molecular 16S rDNA-based phylogenetic approach, clone libraries were constructed from two samples collected from active sulfide edifices in two hydrothermal vent fields. In both cases, clone libraries were largely dominated by ε-Proteobacteria. Phylotypes belonging to the Cytophaga–Flavobacteria and to the Verrucomicrobia were also largely represented within the libraries. The remaining sequences were related to the taxonomic groups Fusobacteria, Green non-sulfur bacteria, Firmicutes, γ- and δ-Proteobacteria. To our knowledge, this is the first report of the presence of Verrucomicrobia, Fusobacteria and green non-sulfur bacteria on hydrothermal edifices. The potential functions of the detected bacteria are discussed in terms of productivity, recycling of organic matter and detoxification within the P. palmiformis microhabitat.

    Description: Knowledge of the processes shaping deep-sea benthic communities at seasonal scales in cold-seep environments is incomplete. Cold seeps within highly dynamic regions, such as submarine canyons, where variable current regimes may occur, are particularly understudied. Novel Internet Operated Vehicles (IOVs), such as tracked crawlers, provide new techniques for investigating these ecosystems over prolonged periods. In this study a benthic crawler connected to the NEPTUNE cabled infrastructure operated by Ocean Networks Canada was used to monitor community changes across 60 m2 of a cold-seep area of the Barkley Canyon, North East Pacific, at ~890 m depth within an Oxygen Minimum Zone (OMZ). Short video-transects were run at 4-h intervals during the first week of successive calendar months, over a 14 month period (February 14th 2013 to April 14th 2014). Within each recorded transect video megafauna abundances were computed and changes in environmental conditions concurrently measured. The responses of fauna to environmental conditions as a proxy of seasonality were assessed through analysis of abundances in a total of 438 video-transects (over 92 h of total footage). 7698 fauna individuals from 6 phyla (Cnidaria, Ctenophora, Arthropoda, Echinodermata, Mollusca, and Chordata) were logged and patterns in abundances of the 7 most abundant taxa (i.e. rockfish Sebastidae, sablefish Anoplopoma fimbria, hagfish Eptatretus stoutii, buccinids (Buccinoidea), undefined small crabs, ctenophores Bolinopsis infundibulum, and Scyphomedusa Poralia rufescens) were identified. Patterns in the reproductive behaviour of the grooved tanner crab (Chionnecetes tanneri) were also indicated. Temporal variations in biodiversity and abundance in megabenthic fauna was significantly influenced by variabilities in flow velocity flow direction (up or down canyon), dissolved oxygen concentration and month of study. Also reported here for the first time are transient mass aggregations of grooved tanner crabs through these depths of the canyon system, in early spring and likely linked to the crab's reproductive cycle.

    Description: The unique ecosystems and biodiversity associated with mid-ocean ridge (MOR) hydrothermal vent systems contrast sharply with surrounding deep-sea habitats, however both may be increasingly threatened by anthropogenic activity (e.g., mining activities at massive sulphide deposits). Climate change can alter the deep-sea through increased bottom temperatures, loss of oxygen, and modifications to deep water circulation. Despite the potential of these profound impacts, the mechanisms enabling these systems and their ecosystems to persist, function and respond to oceanic, crustal, and anthropogenic forces remain poorly understood. This is due primarily to technological challenges and difficulties in accessing, observing and monitoring the deep-sea. In this context, the development of deep-sea observatories in the 2000s focused on understanding the coupling between sub-surface flow and oceanic and crustal conditions, and how they influence biological processes. Deep-sea observatories provide long-term, multidisciplinary time-series data comprising repeated observations and sampling at temporal resolutions from seconds to decades, through a combination of cabled, wireless, remotely controlled, and autonomous measurement systems. The three existing vent observatories are located on the Juan de Fuca and Mid-Atlantic Ridges (Ocean Observing Initiative, Ocean Networks Canada and the European Multidisciplinary Seafloor and water column Observatory). These observatories promote stewardship by defining effective environmental monitoring including characterizing biological and environmental baseline states, discriminating changes from natural variations versus those from anthropogenic activities, and assessing degradation, resilience and recovery after disturbance. This highlights the potential of observatories as valuable tools for environmental impact assessment (EIA) in the context of climate change and other anthropogenic activities, primarily ocean mining. This paper provides a synthesis on scientific advancements enabled by the three observatories this last decade, and recommendations to support future studies through international collaboration and coordination. The proposed recommendations include: i) establishing common global scientific questions and identification of Essential Ocean Variables (EOVs) specific to MORs, ii) guidance towards the effective use of observatories to support and inform policies that can impact society, iii) strategies for observatory infrastructure development that will help standardize sensors, data formats and capabilities, and iv) future technology needs and common sampling approaches to answer today’s most urgent and timely questions.

    Description: As ocean acidification (OA) sensor technology develops and improves, in situ deployment of such sensors is becoming more widespread. However, the scientific value of these data depends on the development and application of best practices for calibration, validation, and quality assurance as well as on further development and optimization of the measurement technologies themselves. Here, we summarize the results of a 2-day workshop on OA sensor best practices held in February 2018, in Victoria, British Columbia, Canada, drawing on the collective experience and perspectives of the participants. The workshop on in situ Sensors for OA Research was organized around three basic questions: 1) What are the factors limiting the precision, accuracy and reliability of sensor data? 2) What can we do to facilitate the quality assurance/quality control (QA/QC) process and optimize the utility of these data? and 3) What sort of data or metadata are needed for these data to be most useful to future users? A synthesis of the discussion of these questions among workshop participants and conclusions drawn is presented in this paper.

    Description: Nitrous oxide (N2O) is a strong greenhouse gas and an ozone depleting agent. In marine environments, N2O is produced biologically via ammonium oxidation, nitrite, and nitrate reduction. The relative importance of these principle production pathways is strongly influenced by oxygen availability. We conducted 15N tracer experiments of N2O production in parallel with measurements of N2O concentration and natural abundance isotopes/isotopomers in Saanich Inlet, a seasonally anoxic fjord, to investigate how temporal and vertical oxygen gradients regulate N2O production pathways and rates. In April, June, and August 2018, the depth of the oxic‐anoxic interface (dissolved oxygen = 2.5 μmol L−1 isoline) progressively deepened from 110 to 160 m. Within the oxygenated and suboxic water column, N2O supersaturation coincided with peak ammonium oxidation activity. Conditions in the anoxic deep water were potentially favorable to N2O production from nitrate and nitrite reduction, but N2O undersaturation was observed indicating that N2O consumption exceeded rates of production. In October, tidal mixing introduced oxygenated water from outside the inlet, displacing the suboxic and anoxic deep water. This oxygenation event stimulated N2O production from ammonium oxidation and increased water column N2O supersaturation while inhibiting nitrate and nitrite reduction to N2O. Results from 15N tracer incubation experiments and natural abundance isotopomer measurements both implicated ammonium oxidation as the dominant N2O production pathway in Saanich Inlet, fueled by high ammonium fluxes (0.6–3.5 nmol m−2 s−1) from the anoxic depths. Partial denitrification contributed little to water column N2O production because of low availability of nitrate and nitrite.
