    Notizen: Biphytanyl membrane lipids and 16S rRNA sequences derived from marine Crenarchaeota were detected in shallow North Sea surface water in February 2002. To investigate the carbon fixation mechanism of these uncultivated archaea in situ 13C bicarbonate tracer experiments were performed with this water in the absence of light. About 70% of the detected 13C incorporation into lipids (including fatty acids and sterols) is accounted for by the crenarchaeotal biphytanyl membrane lipids. This finding indicates that marine Crenarchaeota can utilize bicarbonate or CO2 derived thereof in the absence of light and are chemoautotrophic organisms.

    Notizen: The presence and distribution of isoprenoid glycerol dialkyl glycerol tetraethers (GDGTs), lipids that constitute the membranes of Archaea, have been investigated in a 50-cm long core from a Swedish peat bog. In the acrotelm, the periodically water saturated and thus oxic upper layer of the peat bog, only minor amounts of GDGTs were found. These amounts increase considerably in the catotelm, the continuously water saturated and consequently anoxic lower layer of the peat bog. Based on earlier analyses of GDGTs in different settings and on 16S rDNA results from literature, these lipids are likely derived from methanogenic Archaea. Crenarchaeol, previously only found in marine settings and in fresh water lakes, has also been found in this peat bog. Contrary to the other GDGTs, crenarchaeol concentrations remain relatively constant throughout the peat core, suggesting that they are produced by Crenarchaeota thriving in the oxic part of the peat bog and possibly also in the anoxic part.

    Notizen: The stable carbon isotopic compositions of the inorganic carbon source, bulk cell material, and isoprenoid lipids of the hyperthermophilic crenarchaeon Metallosphaera sedula, which uses a 3-hydroxypropionate-like pathway for autotrophic carbon fixation, have been measured. Bulk cell material was approximately 3‰ enriched in 13C relative to the dissolved inorganic carbon, and 2‰ depleted in 13C relative to isoprenoid membrane lipids. The isotope data suggested that M. sedula uses mainly bicarbonate rather than CO2 as inorganic carbon source, which is in accordance with a 3-hydroxypropionate-like carbon fixation pathway. To the best of our knowledge this is the first report of 13C fractionation effects of such a hyperthermophilic crenarchaeon.

    Notizen: A suite of six sterols, lanosterol, lanost-8(9)-en-3β-ol, 4,4-dimethylcholesta-8(14),24-dien-3β-ol, 4,4-dimethylcholest-8(14)-en-3β-ol, 4-methylcholesta-8(14),24-dien-3β-ol and 4-methylcholest-8(14)-en-3β-ol, were identified in the psychrophilic methanotrophic bacterium, Methylosphaera hansonii. Their presence suggests that the capacity for sterol biosynthesis in methanotrophic bacteria is limited to the family Methylococcaceae but which have widely different optimal growth temperatures.

    Notizen: A white, filamentous microbial mat at the Milano mud volcano in the Eastern Mediterranean Sea was sampled during the Medinaut cruise of the R/V Nadir in 1998. The composition of the mat community was characterized using a combination of phylogenetic and lipid biomarker methods. The mat sample was filtered through 0.2 and 5-μm filters to coarsely separate unicellular and filamentous bacteria. Analyses of 16S rRNA gene sequences amplified from the total community DNA from these fractions showed that similar archaeal populations were present in both fractions. However, the bacterial populations in the fractions differed from one another, and were more diverse than the archaeal ones. Lipid analysis showed that bacteria were the dominant members of the mat microbial community and the relatively low δ13C carbon isotope values of bulk bacterial lipids suggested the occurrence of methane- and sulfide-based chemo(auto)trophy. Consistent with this, the bacterial populations in the fractions were related to Alpha-, Gamma- and Epsilonproteobacteria, most of which were chemoautotrophic bacteria that utilize hydrogen sulfide (or reduced sulfur compounds) and/or methane. The most common archaeal 16S rRNA gene sequences were related to those of previously identified Archaea capable of anaerobic methane oxidation. Although the filamentous organisms observed in the mat were not conclusively identified, our results indicated that the Eastern Mediterranean deep-sea microbial mat community might be sustained on a combination of methane- and sulfide-driven chemotrophy.

    Notizen: Salt adaptation in chemolithotrophic alkaliphilic sulfur-oxidizing strains belonging to genera Thioalkalimicrobium and Thioalkalivibrio has been studied by determination of salt-dependent changes in fatty acid and compatible solute composition. In both alkaliphilic groups, represented by the low salt-tolerant Thioalkalimicrobium aerophilum strain AL 3T and the extremely salt-tolerant Thioalkalivibrio versutus strain ALJ 15, unsaturated fatty acids predominate over saturated fatty acids. In strain AL 3T, C18:1, C16:0 and C16:1 were the dominant fatty acids. In strain ALJ 15, the concentrations of C18:1 and C19cyclo were salt-regulated in an inverse proportional relationship, suggesting the stimulation of cyclopropyl-synthetase activity. Squalene has been found in substantial amounts only in strain ALJ 15. Ectoine and glycine betaine were found to be the main osmolytes in Thioalkalimicrobium aerophilum and Thioalkalivibrio versutus, respectively. The production of ectoine and glycine betaine was positively correlated with the salt concentration in the growth medium. A novel type of membrane-bound yellow pigments was uniformly detected in the extremely salt-tolerant strains of Thioalkalivibrio with a backbone consisting of C15-polyene, whose specific concentration correlated with increasing salinity of the growth medium. The results suggest that the mechanisms of haloalkaliphilic adaptation in Thioalkalimicrobium sp. and Thioalkalivibrio sp. involve the production of cyclopropane fatty acids, organic compatible solutes and, possibly specific pigments.

    Beschreibung: The Cretaceous period (similar to 145-65 m.y. ago) was characterized by intervals of enhanced organic carbon burial associated with increased primary production under greenhouse conditions. The global consequences of these perturbations, oceanic anoxic events (OAEs), lasted up to 1 m.y., but short-term nutrient and climatic controls on widespread anoxia are poorly understood. Here, we present a high-resolution reconstruction of oceanic redox and nutrient cycling as recorded in subtropical shelf sediments from Tarfaya, Morocco, spanning the initiation of OAE2. Iron-sulfur systematics and biomarker evidence demonstrate previously undescribed redox cyclicity on orbital time scales, from sulfidic to anoxic ferruginous (Fe-rich) water-column conditions. Bulk geochemical data and sulfur isotope modeling suggest that ferruginous conditions were not a consequence of nutrient or sulfate limitation, despite overall low sulfate concentrations in the proto-North Atlantic. Instead, fluctuations in the weathering influxes of sulfur and reactive iron, linked to a dynamic hydrological cycle, likely drove the redox cyclicity. Despite the potential for elevated phosphorus burial in association with Fe oxides under ferruginous conditions on the Tarfaya shelf, porewater sulfide generation drove extensive phosphorus recycling back to the water column, thus maintaining widespread open-ocean anoxia.
