    Notizen: Appropriate doping of the electrodes of high-Tc Josephson junctions provides a means to systematically adjust the junctions' electronic properties. This is demonstrated for the exemplary case of grain boundary junctions in bicrystalline Ca-doped YBa2Cu3O7−δ films. It is found that the critical current density is strongly increased and the normal state resistivity significantly reduced in comparison with the values obtained for equivalent junctions in undoped films. © 1999 American Institute of Physics.

    Notizen: Abstract: Nicotinic acetylcholine (ACh) receptors (nAChRs) are important excitatory neurotransmitter receptors in the insect CNS. We have isolated and characterized the gene and the cDNA of a new nAChR subunit from Drosophila. The predicted mature nAChR protein consists of 773 amino acid residues and has the structural features of an ACh-binding α subunit. It was therefore named Dα3, for Drosophilaα-subunit 3. The dα3 gene maps to the X chromosome at position 7E. The properties of the Dα3 protein were assessed by expression in Xenopus oocytes. Dα3 did not form functional receptors on its own or in combination with any Drosophilaβ-type nAChR subunit. Nondesensitizing ACh-evoked inward currents were observed when Dα3 was coexpressed with the chick β2 subunit. Half-maximal responses were at ∼0.15 µM ACh with a Hill coefficient of ∼1.5. The snake venom component α-bungarotoxin (100 nM) efficiently but reversibly blocked Dα3/β2 receptors, suggesting that Dα3 may be a component of one of the previously described two classes of toxin binding sites in the Drosophila CNS.

    Notizen: Summary Structural hair changes may be the expression of a genetic disorder affecting hair growth, part of a congenital syndrome with accompanying hair malformations, or a marker for an underlying metabolic disorder. We report a 22-month-old Turkish girl and her 10-month-old brother, whose scalp hair became fragile and sparse at about 6-7 months of age. Glucosuria. without diabetes or kidney disease, was detected 3-4 months later. Clinical examination revealed normal physical and mental development, and an analysis of plucked hairs showed dysplastic and broken hair shafts. Polarizing microscopy and scanning electron microscopic studies revealed torsion, and irregularities and impressions of the hair shaft, as seen in pilitorti. trichorrhexis nodosa and pseudomonilethrix. Analysis of the amino-acid composition of the hair demonstrated a significant reduction of sulphonic cysteic acid and an elevated cysteine and lanthionine content in the girl, and elevated lanthionine levels in her brother. Electrophoretic analysis of the girl's hair proteins revealed a normal composition but a high extractability of hair proteins.The triad of hypotrichosis structural hair-shaft defects and abnormal amino-acid composition, accompanied by glucosuria without diabetes, may represent a new genetic syndrome.

    Notizen: Abstract  The Drosophila mef2 gene encodes a MADS domain transcription factor required for the differentiation of cardiac, somatic, and visceral muscles during embryogenesis and the patterning of adult indirect flight muscles assembled during metamorphosis. A prerequisite for D-MEF2 function in myogenesis is its precise expression in multiple cell types during development. Novel enhancers for D-mef2 transcription in cardiac and adult muscle precursor cells have been identified and their regulation by the Tinman and Twist myogenic factors have been demonstrated. However, these results suggested the existence of additional regulators and provided limited information on the specification of progenitor cells for different muscle lineages. We have further characterized the heart enhancer and show it is part of a complex regulatory region controlling the activation and repression of D-mef2 transcription in several cell types. The mutation of a GATA sequence in the enhancer changes its specificity from cardial to pericardial cells. Also, the addition of flanking sequences to the heart enhancer results in expression in a new cell type, that being the founder cells of a subset of body wall muscles. As tinman function is required for D-mef2 expression in both the cardial and founder cells, these results define a shared regulatory DNA that functions in distinct lineages due to the combinatorial activity of Tinman and other factors that work through adjacent sequences. The analysis of D-mef2-lacZ fusion genes in mutant embryos revealed that the specification of the muscle precursor cells involved the wingless gene and the activation of a receptor tyrosine kinase signaling pathway.

    Notizen: Abstract The high energy ball-milling technique has been used to produce metastable Ni(Ru) alloys supported on alumina. This catalyst has potential application for the hydrogenation of the C=O bond. The active species are first formed by mechanical alloying pure Ni and Ru powders. The alumina support is ground independently until no structural transformation occurs. Finally, the supersaturated Ni(Ru) powder is milled with the alumina support to produce highly dispersed individual Ni(Ru) nanocrystals on the surface of Al2O3. This supported catalyst has been characterized by X-ray diffraction, transmission electron microscopy, X-ray photoelectron spectroscopy and thermal desorption spectroscopy.

    Notizen: Abstract The International Rosetta Mission, approved by the Science Programme Committee of the European Space Agency as the Planetary Cornerstone Mission in ESA's long-term programme Horizon 2000, will rendezvous in 2011 with Comet 46P/Wirtanen close to its aphelion and will study the nucleus and the evolution of the coma for almost two years until it reaches perihelion. In addition to the investigations performed by the scientific instruments on board the orbiter, a Surface Science Package (Rosetta Lander) will be deployed onto the surface of the nucleus early during the near-nucleus study phase. On its way to Comet 46P/Wirtanen, Rosetta will fly by and study the two asteroids 4979 Otawara and 140 Siwa.
