    Notes: Abstract The examination of oogenesis and reproductive pattern in three deep-sea demosponge species from the foot of the Barents Sea continental slope (Norwegian Sea, ∼ 2300 m, ∼ 75°18′N; 9°55′E), Thenea abyssorum, Trichostemma sol and Tentorium semisuberites, suggests that sexual reproduction is triggered by the vertical flux of particulate organic carbon at least in one species. All three species examined reproduce sexually and proved to be oviparous and gonochoristic. A considerable number of asexually produced reproductive units was found in Thenea abyssorum, but owing to morphological details the origin of these buds is equivocal, and asexual reproduction is therefore judged to be of minor importance. Oogenesis in the three species can be divided into two developmental periods and does not differ greatly from processes known from shallow water species, except for the almost complete absence of nurse cells. For Thenea abyssorum a distinct seasonal reproductive cycle was found. The onset of yolk accumulation coincides precisely with a maximum in vertical particle flux as detected in sediment trap studies (von Bodungen et al. 1994). In addition, prior to yolk accumulation the sponge tissue appears almost “empty”, without storage units. Once yolk accumulation (i.e. the second growth period) has started, gamete development and release are completed within a few weeks. Thus sexual reproduction is an important reproductive mode in deep-sea sponges, which may be triggered by the pulsed input of POC to this food-limited environment, the energy-dependent process of yolk accumulation being the most sensitive process.

    Notes: Abstract About 70% of patients with Prader-Willi syndrome (PWS) and Angelman syndrome (AS) have a common interstitial de novo microdeletion encompassing paternal (PWS) or maternal (AS) loci D15S9 to D15S12. Most of the non-deletion PWS patients and a small number of non-deletion AS patients have a maternal or paternal uniparental disomy (UPD)15, respectively. Other chromosome 15 rearrangements and a few smaller atypical deletions, some of the latter being associated with an abnormal methylation pattern, are rarely found. Molecular and fluorescence in situ hybridization (FISH) analysis have both been used to diagnose PWS and AS. Here, we have evaluated, in a typical routine cytogenetic laboratory setting, the efficiency of a diagnostic strategy that starts with a FISH deletion assay using Alu-PCR (polymerase chain reaction)-amplified 1315S10-positive yeast artificial chromosome (YAC) 273A2. We performed FISH in 77 patients suspected of having PWS (n = 66) or AS (n = 11) and compared the results with those from classical cytogenetics and wherever possible with those from DNA analysis. A FISH deletion was found in 16/66 patients from the PWS group and in 3/11 patients from the AS group. One example of a centromere 15 co-hybridization performed in order to exclude cryptic translocations or inversions is given. Of the PWS patients, 14 fulfilled Holm's criteria, but two did not. DNA analysis confirmed the commmon deletion in all patients screened by the D15S63 methylation test and in restriction fragment length polymorphism dosage blots. In 3/58 non-deletion patients, other chromosomal aberrations were found. Of the non-deleted group, 27 subjects (24 PWS, 3 AS) were tested molecularly, and three patients with an uniparental methylation pattern were found in the PWS group. The other 24/27 subjects had neither a FISH deletion nor uniparental methylation, but two had other cytogenetic aberrations. Given that cytogenetic analysis is indispensable in most patients, we find that the FISH deletion assay with YAC 273A2 is an efficient first step for stepwise diagnostic testing and mutation-type analysis of patients suspected of having PWS or AS.

    Description: Due to the scarcity of information concerning the role that large foraminifera play in deep-sea carbon cycling, the response of a foraminiferal community (>250 pm) to a simulated phytodetritus sedimentation event was assessed over two weeks using sediment cores collected from a deep-fjord environment. Sediment cores were collected from similar to 700 in water depth in the Korsfjorden, western Norway, and incubated ex situ with 1 9 C-org m(2) of labile C-13-labeled Skeletonema costatum for 2, 7, and 14 days. We selectively picked (without prior staining) cytoplasm-containing foraminifera and found the foraminiferal community to be largely dominated (91%) by the deep-dwelling species Globobulimina turgida and Melonis barleeanum, as well as the shallow infaunal species 1 Hyalinea balthica. None of the >250 mu m, cytoplasm-containing fraction was involved in carbon uptake during the first 7 days. After 14 days, 3% of the foraminiferal samples possessed delta C-13 signatures indicative of carbon uptake, but the uptake was confined to the surface-living (0-2 cm) G. turgida. Foraminifera contributed 2.4% to faunal carbon uptake (foraminifera plus macrofauna) after 14 days, despite making up 24% of the combined biomass. Both the dominance of deep-infaunal species, such as G. turgida and M. barleeanum (68%), which are known to prefer degraded over more labile material, together with their large size, which often makes foraminifera respond slower to phytodetritus deposition than the more abundant, smaller-size foraminifera, are possible reasons for the retarded response observed. Overall, results from this investigation highlight that the response of large-size foraminifera to phytodetritus deposition is very slow. In addition, the results presented provide evidence that the foraminiferal response is most likely driven by differences in foraminiferal community composition and structure, with large, deep-infaunal species showing slower reactions to phytodetritus deposition compared to smaller foraminifera, metazoan meiofauna, and macrofauna.

Most of this major reservoir in the global carbon cycle and final repository for anthropogenic wastes is characterized by severe food limitation. Phytodetritus is the major food source for abyssal benthic communities, and a large fraction of the annual food load can arrive in pulses within a few days1, 2. Owing to logistical constraints, the available data concerning the fate of such a pulse are scattered3, 4 and often contradictory5, 6, 7, 8, 9, 10, hampering global carbon modelling and anthropogenic impact assessments. We quantified (over a period of 2.5 to 23 days) the response of an abyssal benthic community to a phytodetritus pulse, on the basis of 11 in situ experiments. Here we report that, in contrast to previous hypotheses5, 6, 7, 8, 9, 10, 11, the sediment community oxygen consumption doubled immediately, and that macrofauna were very important for initial carbon degradation. The retarded response of bacteria and Foraminifera, the restriction of microbial carbon degradation to the sediment surface, and the low total carbon turnover distinguish abyssal from continental-slope ‘deep-sea’ sediments.

    Description: A large fraction of globally produced methane is converted to CO2 by anaerobic oxidation in marine sediments. Strong geochemical evidence for net methane consumption in anoxic sediments is based on methane profiles, radiotracer experiments and stable carbon isotope data. But the elusive microorganisms mediating this reaction have not yet been isolated, and the pathway of anaerobic oxidation of methane is insufficiently understood. Recent data suggest that certain archaea reverse the process of methanogenesis by interaction with sulphate-reducing bacteria. Here we provide microscopic evidence for a structured consortium of archaea and sulphate-reducing bacteria, which we identified by fluorescence in situ hybridization using specific 16S rRNA-targeted oligonucleotide probes. In this example of a structured archaeal-bacterial symbiosis, the archaea grow in dense aggregates of about 100 cells and are surrounded by sulphate-reducing bacteria. These aggregates were abundant in gas-hydrate-rich sediments with extremely high rates of methane-based sulphate reduction, and apparently mediate anaerobic oxidation of methane.

    Description: We have compared 14 different sediment incubation chambers, most of them were used on bottom landers. Measurements of mixing time, pressure gradients at the bottom and Diffusive Boundary Layer thickness (DBL) were used to describe the hydrodynamic properties of the chambers and sediment–water solute fluxes of silicate (34 replicates) and oxygen (23 replicates) during three subsequently repeated incubation experiments on a homogenized, macrofauna-free sediment. The silicate fluxes ranged from 0.24 to 1.01 mmol m−2 day−1 and the oxygen fluxes from 9.3 to 22.6 mmol m−2 day−1. There was no statistically significant correlation between measured fluxes and the chamber design or between measured fluxes and hydrodynamic settings suggesting that type of chamber was not important in these flux measurements. For verification of sediment homogeneity, 61 samples of meiofauna were taken and identified to major taxa. In addition, 13 sediment cores were collected, sectioned into 5–10-mm slices and separated into pore water and solid phase. The pore water profiles of dissolved silicate were used to calculate diffusive fluxes of silicate. These fluxes ranged from 0.63 to 0.87 mmol m−2 day−1. All of the collected sediment parameters indicated that the sediment homogenization process had been satisfactorily accomplished. Hydrodynamic variations inside and between chambers are a reflection of the chamber design and the stirring device. In general, pump stirrers with diffusers give a more even distribution of bottom currents and DBL thicknesses than paddle wheel-type stirrers. Most chambers display no or low static differential pressures when the water is mixed at rates of normal use. Consequently, there is a low risk of creating stirrer induced pressure effects on the measured fluxes. Centrally placed stirrers are preferable to off-center placed stirrers which are more difficult to map and do not seem to give any hydrodynamic advantages. A vertically rotating stirrer gives about five times lower static differential pressures at the same stirring speed as the same stirrer mounted horizontally. If the aim is to simulate or mimic resuspension at high flow velocities, it cannot be satisfactorily done in a chamber using a horizontal (standing) rotating impeller (as is the case for most chambers in use) due to the creation of unnatural conditions, i.e. large static differential pressures and pre-mature resuspension at certain locations in the chamber.
