    Notes: β-Arrestins are key negative regulators and scaffolds of G protein-coupled receptor (GPCR) signalling. β-Arrestin1 and β-arrestin2 preferentially bind to the phosphorylated GPCRs in response to agonist stimulation, resulting in receptor internalization and desensitization. The critical roles of GPCR kinases (GRKs)-catalyzed receptor phosphorylation and interaction of β-arrestins with the phosphorylated receptor in receptor internalization are well established. However, emerging evidence suggests that an agonist-stimulated internalization mechanism that is independent of receptor phosphorylation may also be employed in some cases, although the molecular mechanism for the phosphorylation-independent GPCR internalization is not clear. The current study investigated the role of receptor phosphorylation and the involvement of different β-arrestin subtypes in agonist-induced δ-opioid receptor (DOR) internalization in HEK293 cells. Results from flow cytometry, fluorescence microscopy, and surface biotin labelling experiments showed that elimination of agonist-induced DOR phosphorylation by mutation GRK binding or phosphorylation sites only partially blocked agonist-induced receptor internalization, indicating the presence of an agonist-induced, GRK-independent mechanism for DOR internalization. Fluorescence and co-immunoprecipitation studies indicated that both the wild-type DOR and the phosphorylation-deficient mutant receptor could bind and recruit β-arrestin1 and β-arrestin2 to the plasma membrane in an agonist-stimulated manner. Furthermore, internalization of both the wild-type and phosphorylation-deficient receptors was increased by overexpression of either type of β-arrestins and blocked by dominant-negative mutants of β-arrestin-mediated internalization, demonstrating that both phosphorylation-dependent and -independent internalization require β-arrestin. Moreover, double-stranded RNA-mediated interference experiments showed that either β-arrestin1 or β-arrestin2 subtype-specific RNAi only partially inhibited agonist-induced internalization of the wild-type DOR. However, agonist-induced internalization of the phosphorylation-deficient DOR was not affected by β-arrestin1-specific RNAi but was blocked by RNAi against β-arrestin2 subtype. These data indicate that endogenous β-arrestin1 functions exclusively in the phosphorylation-dependent receptor internalization, whereas endogenous β-arrestin2, but not β-arrestin1, is required for the phosphorylation-independent receptor internalization. These results thus provide the first evidence of different requirement for β-arrestin isoforms in the agonist induced phosphorylation-dependent and -independent GPCR internalization.

    Description: The foraminifera and geochemical element records from the Core CS2 in the center of the Okinawa Trough (OT) revealed a history of current circulation and bottom redox conditions over the last 19 Ka. During the last glaciation, the few Kuroshio Current (KC) and tropical/subtropical species components, the shoaled depth of thermocline (DOT) and high infaunal foraminifera, together suggest the hypoxia in the isolated and semiclosed OT might be related to an absence/shift of the KC. Furthermore, the hypoxia variations in three episodes coincided with changes in the North Pacific Intermediate Water (NPIW), as stronger hypoxia and enhanced NPIW occurred in the Heinrich Stadia 1 and Younger Dryas periods, and rapidly decreasing hypoxia corresponded with weaker NPIW ventilation in the Bølling/Allerød period. Previously recorded hypoxia in the OT occurred broadly at a water depth of between ~1,300 and 1,800 m. Meanwhile, the lowest productivity precluded the possibility of hypoxia caused by organic matter consumption. We thereby infer the enhanced glacial NPIW might be another reason for the hypoxia. After the postglaciation, the OT transitioned into a fully oceanic environment and was reoccupied by main pathway of the KC. This reoccupation greatly alleviated the hypoxia via weakening stratification and increasing vertical mixing. Evolution research of current circulation and bottom redox conditions can help in understanding the changes in the storage/release of carbon as the paleoclimate changed. Moreover, the variations in bottom redox conditions in the OT offer a possible informative window for understanding the influence of glacial NPIW in the mid-latitude zone.

    Description: Stem characteristics of plants are of great importance to both ecology study and forest management. Terrestrial laser scanning (TLS) may provide an effective way to characterize the fine-scale structures of vegetation. However, clumping plants, dense foliage and thin structure could intensify the shadowing effect and pose a series of problems in identifying stems, distinguishing neighboring stems, and merging disconnected stem parts in point clouds. This paper presents a new method to automatically detect stems in dense and homogeneous forest using single-scan TLS data. Stem points are first identified with a two-scale classification method. Then a clustering approach is used to group the candidate stem points. Finally, a direction-growing algorithm based on a simple stem curve model is applied to merge stem points. Field experiments were carried out in two different bamboo plots with a stem density of about 7500 stems/ha. Overall accuracy of the stem detection is 88% and the quality of detected stems is mainly affected by the shadowing effect. Results indicate that the proposed method is feasible and effective in detection of bamboo stems using TLS data, and can be applied to other species of single-stem plants in dense forests.

    Description: The concept of aromaticity has long played an important role in chemistry and continues to fascinate both experimentalists and theoreticians. Among the archetypal aromatic compounds, heteroaromatics are particularly attractive. Recently, substitution of a transition-metal fragment for a carbon atom in the anti-aromatic hydrocarbon pentalene has led to the new heteroaromatic osmapentalenes. However, construction of the aza-homolog of osmapentalenes cannot be accomplished by a similar synthetic manipulation. Here, we report the synthesis of aza-osmapentalenes by sequential ring contraction/annulation reactions of osmabenzenes via osmapentafulvenes. Nuclear magnetic resonance spectra, X-ray crystallographic analysis, and DFT calculations all suggest that these aza-osmapentalenes exhibit aromatic character. Thus, the stepwise transformation of metallabenzenes to metallapentafulvenes and then aza-metallapentalenes provides an efficient and facile synthetic route to these bicyclic heteroaromatics.     Description: A novel full-field initialization strategy based on the dimension-reduced projection four-dimensional variational data assimilation (DRP-4DVar) is proposed to alleviate the well-known initial shock occurring in the early years of decadal predictions. It generates consistent initial conditions, which best fit the monthly mean oceanic analysis data along the coupled model trajectory in one-month windows. Three indices to measure the initial shock intensity are also proposed. Results indicate that this method does reduce the initial shock in decadal predictions by FGOALS-g2 compared with the 3DVar-based nudging full-field initialization for the same model and is comparable to or even better than the different initialization strategies for other CMIP5 models. Better hindcasts of global mean surface air temperature anomalies can be obtained than in other FGOALS-g2 experiments. Due to the good model response to external forcing and the reduction of initial shock, higher decadal prediction skill is achieved than in other CMIP5 models.

    Description: Dietary restriction (DR) refers to a reduction in food intake to induce undernutrition but not malnutrition, which extends the lifespan of multiple species. Although there are invertebrate aging models, such as the Caenorhabditis elegans and Drosophila melanogaster , aging studies in Lepidoptera are few in number and the underlying life-extending molecular mechanisms are not clear. Research on a broader range of animals is necessary to support generalizations on mechanisms of aging and rates of aging. The aim of this study was to further investigate genes and pathways associated with DR in Bombyx mori . Here, we used mRNA deep sequencing (RNA-seq) to further investigate genes and pathways associated with DR. The transcriptome profiles showed that most of the differentially expressed genes were upregulated following DR, and genes involved in amino acid and protein metabolism, RNA metabolism and translation, energy metabolism, nitrogen metabolism, and juvenile hormone pathway-related proteins were particularly affected. DR also affects the metabolism of uric acid and urea, which accumulated in silkworm following DR. We speculate that this may not be due to activation of uric acid biosynthesis, but rather by downregulating the degradation of uric acid and urea. These results may help us to understand the mechanisms by which DR prolong lifespan in insects and other animals.
