    Notes: Abstract PURPOSE: This study was designed to determine whether the ability of interferon-γ to upregulate the expression of a human tumor antigen improved the therapeutic efficacy of a radionuclide-conjugated monoclonal antibody. METHODS: Tumor xenografts of the moderately differentiated human colon tumor cell line HT-29 were grown in athymic mice. Constitutive levels of the human tumor antigen, tumor-associated glycoprotein-72, were measured before and after treatment with interferon-γ. Antitumor effects of an131I-labeled antitumor-associated glycoprotein-72 monoclonal antibody, CC49, were determined by measuring changes in tumor volumes in the respective groups of athymic mice. RESULTS: Interferon-γ induced a time-dependent and dose-dependent increase in tumor-associated glycoprotein-72 expression in the HT-29 tumors. Immunohistochemical staining revealed a more homogeneous tumor-associated glycoprotein-72-positive tumor cell population in tumors isolated from mice treated for eight days with interferon-γ, which accounted for the enhanced tumor localization of131I-CC49 in mice. That experimental model was used to examine the antitumor effects of combining interferon-γ with131I-CC49. Administration of 300 μCi of131I-CC49 to mice bearing HT-29 tumors induced a transient suppression of tumor growth. Conversely, a long-term, sustained HT-29 tumor growth suppression was achieved in mice given 300μCi of131I-CC49 and interferon-γ. In fact, the cytokine/radioimmunoconjugate combination eradicated any evidence of tumor in approximately 30 percent of the mice. CONCLUSION: The ability of interferon-γ to enhance tumor-associated glycoprotein-72 expression substantially augmented the antitumor effects of the radioimmunoconjugate. Those observations provide additional argument for use of a radioimmunoconjugate in combination with a cytokine to improve tumor diagnosis and therapy.

    Description / Table of Contents: Zusammenfassung Die terminale Herzinsuffizienz kann nach Ausschöpfung medikamentöser Therapie oft nur mit mechanischer Kreislaufunterstützung beherrscht werden. Besonders die Therapieergebnisse des rechtzeitigen elektiven Einsatzes von pulsatilen kardialen Assist-Systemen als Überbrückung zur Herztransplantation unterstreichen den Erfolg der Behandlungsmethode. Durch die Aufrechterhaltung bzw. die Wiederherstellung einer ausreichenden Zirkulation mittels mechanischer Kreislaufunterstützung kommt es bei vielen Patienten auch zur Erhaltung oder Wiederherstellung einer ausreichenden Organfunktion. Bei Patienten der NYHA-Klasse IV mit Kontraindikation gegen eine Herztransplantation bietet sich die Implantation eines Langzeitsystems als allerdings bisher noch zeitlich limitierte Alternative zur Transplantation. Eine postkardiotomiebedingte Kreislaufunterstützung bietet bei ansonsten infauster Prognose vielen Patienten die einzige Möglichkeit des Überlebens. Wenn Hauptkomplikationen wie Thrombembolie, Blutung und Infektion nicht auftreten, lohnen gute Ergebnisse den hohen personellen und technischen Aufwand. Das postoperative intensivmedizinische und pflegerische Management erfordert eine spezielle Ausbildung der Mitarbeiter einer Intensivstation.
    Notes: Summary Despite medical therapy, chronic heart failure can often only be treated using a mechanical assist system. The results, when using such devices electively, underline that it is an effective therapy. In many patients, “dependent” organs, such as liver or kidneys etc., can regain their usual function soon after adequate circulatory function is reestablished by the mechanical assist system. In patients with NYHA class IV heart failure and contraindications for heart transplantation, implantation of a long-lasting system is an alternative approach, even though available systems do not yet last forever. Post-cardiosurgical implantation of an assist device is the only option in some patients. The good results are rewarding the high technical and personal efforts if no major-complications, such as thromboembolism, bleedings or infections, occur. Postoperative treatment of these patients requires special training of all those involved.

    Description: The Jurassic Period (ca. 201–145 Ma) is marked by fundamental reorganizations of paleogeography, paleoceanography, ecosystems, and the progressive shift from aragonite to calcite as the favored marine biogenic carbonate polymorph. Sr/Ca ratios of well-preserved Jurassic oysters and belemnites from sections in the UK and Poland demonstrate that the Sr/Ca ratio of seawater varied systematically throughout the Early and Middle Jurassic in parallel with already documented seawater 87 Sr/ 86 Sr. The Sr flux from increased mid-ocean ridge activity in the Early Jurassic outbalanced the input of riverine Sr, leading to gradually lower seawater 87 Sr/ 86 Sr associated with the parallel and strong decrease in seawater Sr/Ca ratios. This downward trend was reversed by enhanced continental input in the Toarcian and Aalenian, but resumed in the Bajocian and continued to the Callovian. Parallel changes of seawater 87 Sr/ 86 Sr and Sr/Ca suggest a common cause for these long-term variations and are best explained by changes in the balance of continental weathering and hydrothermal fluxes. These findings underline the strong control of global tectonic processes on the evolution of biomineralization and downplay the role of biomineral evolution in influencing strontium chemistry of seawater in the mid-Mesozoic.

    Description: The age distribution of lithium-cesium-tantalum (LCT) pegmatites largely overlaps with major phases of collisional orogenic events and assembly of supercontinents. Some of the largest known LCT pegmatite deposits formed in very short intervals, 2.7–2.5 Ga and 1.9–1.8 Ga, corresponding to two major pulses of continental crust growth. However, the exact process of generation and segregation of large volumes of Li-bearing pegmatite liquids, perhaps involving disequilibrium fractional crystallization and leaving residual melts enriched in fluxing elements such as B, F, H 2 O, Li, and P, remains obscure. The new data on Li contents and isotope compositions in major mineral phases from temporally and geographically separated pegmatite bodies document extreme variations in 7 Li values among individual large LCT pegmatite bodies, in particular Archean occurrences. The observed 〉10 variations in 7 Li values for the same mineral phases from different localities (i.e., beryl, petalite, spodumene, lepidolite, amblygonite, muscovite) contrast with globally homogeneous Li isotope systematics of major mineral phases from unmodified mantle rocks. Consistent Li isotope offsets between coexisting mineral phases are best explained by Li isotope fractionation as a function of the bond length between Li and neighboring ions (O, OH, F). We suggest that spatially distinct Li isotope patterns act as fingerprints for different pegmatites and can be explained by the preexisting Li isotope differences of their crustal sources at the time of pegmatite formation owing to differences in crustal age and evolution. This would imply secular evolution of the continental crust over Earth history toward present-day globally broadly uniform crustal 7 Li/ 6 Li ratios ( 7 Li ~0). The differences among Archean occurrences could reflect possible Archean paleogeography and could be linked with different thermal regimes of individual cratons as a consequence of variations in crustal thickness. One possible application of the new data set may be in source verification of gem-quality stones by using Li isotopes.
