    Notizen: Abstract Deoxyribonucleic-acid sequences expressed at high levels in meristematic tissues of barley (Hordeum vulgare L.) have been cloned by differential hybridization. Five out of the seven cDNA clones studied showed homologies to histone genes H2a (two clones), H2b, H3 and H4. Their patterns of expression, as studied by RNA and in-situ hybridization, were typical for genes transcribed during cell division. A sixth cDNA clone, Sab2, had a 65.7% identity (on a protein basis) to L2-like ribosomal proteins of Escherichia coli and other lower prokaryotes. In a domain of 50 amino acids, the seventh clone, Sab35, showed 69.0% sequence identity to the ribosomal protein L21 of Rattus norvegicus. The Sab35 mRNA contained in its 5′-untranslated leader sequence small open reading frames, a feature pointing to a possible translational control. The Sab35 in-situ hybridization pattern was to a certain degree different from that of the histone-like clone Sab11: it detected transcripts not only in tissues that are associated with vegetative and reproductive apices but also in sub-apical regions. The visualization in situ of transcripts coded by Sab11, 35 and 44 is discussed as a possible technique for studying differential gene expression in barley meristematic tissues.

    Notizen: Abstract A full-length double-stranded DNA copy of the single-stranded circular DNA associated with coconut foliar decay virus (CFDV) was constructed. Full-length CFDV DNA and smaller fragments were transcriptionally fused to the β-glucuronidase reporter gene and examined for promoter activity in vivo. In stably transformed tobacco plants, the CFDV DNA promoter confered a tissue-specific expression pattern in that the reporter gene was specifically expressed in the phloem tissue of the vascular system in stem, leaves and flower. These results are in agreement with the previously reported association of CFDV DNA with the phloem of its coconut host plant.

    Notizen: Summary cDNA clones were isolated from tissue specific cDNA libraries of barley and maize using as a probe the cDNA of the maize gene C1, a regulator of anthocyanin gene expression. C1-related homology for all of the four cDNAs characterized by sequence analysis is restricted to the N-terminal 120 amino acids of the putative proteins. This region shows striking homology to the N-proximal domain of the myb oncoproteins from vertebrates and invertebrates. Within the myb proto-oncogene family this part of the respective gene products functions as a DNA binding domain. Acidic domains are present in the C-proximal protein segments. Conservation of these sequences, together with the genetically defined regulator function of the C1 gene product, suggest that myb-related plant genes code for trans-acting factors which regulate gene expression in a given biosynthetic pathway.

    Notizen: Bacteria of the genera Acetobacter, Bifidobacterium, Brevibacterium, Lactobacillus, Micrococcus, Propionibacterium, and Streptococcus, which are used as so-called starter cultures for the large-scale production of fermented foods and beverages in food biotechnology, have been investigated for the chirality of their amino acids (AA) by gas chromatography (GC). Bacteria were grown in complex media, centrifuged, and washed with 0.85% aqueous NaCl. Aliquots were totally hydrolyzed (6 M HCl, 110°C, 18 h), or extracted with 70% aqueous ethanol in order to isolated free AA. The AA were adsorbed on Dowex WX 8 cation-exchanger, eluted with 4 M ammonia and converted into their N(O)-trifluoroacetyl(TFA) 2-propyl esters or TFA methyl esters. The AA derivatives were investigated by capillary GC using the chiral stationary phases Chirasil-L-Val, Chirasil-D-Val, and Lipodex E. Besides L-AA, in all bacteria D-amino acids (D-AA) were detected; those in the highest relative amounts were D-Ala and D-Asp (occurring in all bacteria) and, in several cases, D-Glu. Lower, but significant amounts of other D-AA such as D-Ser, D-Pro, D-Val, D-Thr, D-Ile, D-Leu, D-Met, D-Phe, D-Tyr, D-Orn, and D-Lys were also detected in certain bacteria. These findings explain the origin of D-AA found in all fermented foods and drinks produced with the aid of bacterial starter cultures. © 1993 Wiley-Liss, Inc.

    Notizen: From a submerse culture of the fungus Hypocrea muroiana Hino et Katsumoto (Ascomycetes: Hypocreales) two major groups of peptides, designated hypomurocin (HM) A and B, belonging to the peptaibol family, could be characterized. Both groups showed antibiotic activity (Bacillus subtilis) and caused hemolysis of rat erythrocytes, HM A being less active than HM B. Peptides were isolated from the culture broth by chromatography on XAD-2 adsorber resin, octyl silica, and Sephadex LH 20. HM A and B were separated by preparative TLC, and individual peptides from each microheterogeneous group were isolated by preparative HPLC. Amino acid analysis and sequence determination by fast atom bombardment and electrospray tandem mass spectrometry revealed the composition and structures of six 11-mer peptides of the HM A group and of six 18-mer peptides of the HM B group. Positions of isomeric amino acids Leu/Ile and Val/Iva (present in some of the peptides) were determined by methanolytic cleavage of the pure peptides, followed by trifluoroacetylation of the dipeptide methyl esters released and assignment of their structures by gas chromatography-selected ion monitoring mass spectrometry. As examples, two sequences of HM A and HM B are presented (exchange positions in parentheses). - HM A-1(2): Ac-Aib1(D-Iva1)-Gln-Val-Val-Aib-Pro-Leu-Leu-Aib-Pro-Leuol11; HMB-1(2): Ac-Aib1-Ser-Ala-Leu-Aib-Gln-Aib-Val-Aib-Gly-Aib-Pro-Leu-Aib-Aib-Gln-Valol18 (Leuol18), (Ac = acetyl, Aib = α-aminoisobutyric acid, Iva = D-isovaline, Leuol = L-leucinol, Valol = L-valinol).

    Notizen: Hydrogen Cyanide N-Methylide: Preparation, Spectroscopic Identification, and Its Relationship to Other C2H3N IsomersWith the intention of isolating 1H-azirene (8) we studied several precursors which should offer an entry into the C2H3N potential energy hypersurface. It was found that in addition to the known isomers 1, 2, 3, and 6 hydrogen cyanide N-methylide (5)  -  and probably aminoacetylene (4)  -  can be detected in an argon matrix at 10 K. No spectroscopic evidence for the formation of 1H-azirene (8) was found. In other words, the first six members 1 - 6 are now all identified. The elucidation of the structure of the new C2H3N species is based on the comparison of the observed IR spectra with those calculated by ab initio methods.

    Notizen: Scale biogenesis in algae represents a unique model system to study the transport of secretory macromolecules through the Golgi apparatus (GA) and their exocytosis. The larger scales can be visualized in the light microscope, and thus the kinetics of scale assembly, transport, and secretion can be studied in vivo. In addition, scales are osmiophilic and readily visible in conventional transmission electron microscopy; thus, details of scale assembly and sorting can be studied without invoking immunolabeling techniques. The following are distinctive features of scale biogenesis in algae: (1) transport of scales through the GA-stack occurs by cisternal progression; (2) scale secretion may be very rapid (in some cases a single GA-cisterna leaves the stack every 15-20 s); (3) sorting of different scale types does not occur in the GA, but in a post-GA-compartment. Recent progress in the analysis of scale formation in the green flagellates Tetraselmis and Scherffelia is reviewed.
