    Notes: Abstract We present preliminary data on the performance of a new fast photodetector based on a W–InSb metal-insulator-semiconductor point contact diode operating at room temperature and with no bias voltage. The device can work either as a video detector or as harmonic mixer for radiation from far–infrared (FIR) to visible. In the FIR region, for wavelengths from 200 to 400 μm, the W–InSb point contact diode showed a sensitivity comparable to that of Golay cells. In the visible region the device showed a video and heterodyne detection responsivity much higher with respect to standard M.I.M. point contact diodes. Owing to its ruggedness, low cost and wide band of operation, the W–InSb point contact diode may be very attractive as a general purpose optical sensor.

    Notes: Abstract The technique of optical pumping in polar molecules is the most efficient for Far-Infrared (FIR) laser generation, providing also a versatile and powerful tool for molecular spectroscopy in this spectral region. Methanol (CH3OH) and its isotopic varieties are the best media for optically pumped FIR laser, with over thousand lines observed, and the most widely used for investigations and applications. In this sense, it is important organize and make available catalogues of FIR laser lines as complete as possible. Since the last critical reviews of 1984 [1] on methanol and its isotopic varieties [2,3,4], over hundred papers have been published dealing with hundreds of new FIR laser lines. In 1992 a review of FIR laser lines from CH3OH was presented [5]. In this communication we extend this work to the other methanol isotopes, namely CH3OD, CD3OH, CD3OD,13CH3OH,13CD3OH,13CD3OD, CH3 18OH, CH2DOH, CHD2OH and CH2DOD.

    Notes: Abstract We used a Stark-Optoacoustic cell and hybrid waveguide resonators to perform an Infrared and Far Infrared Stark Spectroscopy study on some transitions of13CD3OH. Different behaviours of the transitions in the presence of a d.c. electric field were observed. The Stark splittings of six FIR laser lines ranging from 34 to 136 MHz/kVcm−1 were determined. The analysis of the behaviour of the IR and FIR transitions in the presence of the external electric fields gives important and exclusive information on the levels involved in the transitions.

    Description: Highlights: • 18 southern European charcoal records document 16,000 years of fire regime changes. • Since the Neolithic, land uses have artificially maintained high fire frequencies. • Human-induced decreases in forest cover led to a reduction in the biomass burned. • Human-modified landscapes affected ecological processes more meaningly than expected. Abstract: Variability in fire regime at the continental scale has primarily been attributed to climate change, often overshadowing the widely potential impact of human activities. However, human ignition modifies the rhythm of fire episodes occurrence (fire frequency), whereas land use alters vegetation composition and fuel load, and thus the amount of biomass burned. It is unclear, however, whether and how humans have exercised a significant influence over fire regimes at continental and millennial scales. Based on sedimentary charcoal records, we use new alternative estimate of fire frequency and biomass burned for the last 16000 years (here after 16 ky) that we evaluate with outputs from climate, vegetation, land use and population models. We find that pronounced regional-scale land use changes in southern Europe at the beginning of the Neolithic (8–6 ky), during the Bronze Age (5–4 ky) and the medieval period (1 ky) caused a doubling of fire frequency compared to the Holocene average (the last 11.5 ky). Despite anthropogenic influences, southern European biomass burned decreased from 7 ky, which is in line both with changes in orbital parameters leading climate cooling and also reductions in biomass availability because of land use. Our study underscores the role of elevation-dependent parameters, and particularly biomass and land management, as major drivers of fire regime variability. Results attest a determinant anthropogenic driving-force on fire regime and a decrease in fire-carbon emissions since 7 ky in Southern Europe.

    Description: The EPR spectrum of Mn 2+ in microcrystalline calcite geomaterials (e.g., marbles, travertines) possesses exceptional diagnostic characteristics, allowing to relate samples to their origin (natural/synthetic, inorganic, organogenic, ...) and to evaluate the details of impurities clustering. This information, beyond their mineralogical and geochemical interest, is of paramount importance for environmental, palaeoclimatic, and cultural heritage studies. Accessing the information hidden in the Mn 2+ EPR spectrum relies on disentangling spurious self-correlation among spin Hamiltonian parameters in the powder spectrum. In the present study, this goal is achieved through a systematic comparison of the temperature dependencies of four different microcrystalline calcite geomaterials. Accordingly, an assessment of the internal correlation structure of the spin Hamiltonian parameters is provided and the most sensitive discriminating parameters, which are able to mark samples, are identified. It has been found that the spin Hamiltonian parameters useful for discrimination purposes are those which are dependent on the ligand field interaction, whereas the Fermi contact interaction, as well as the spin-spin, spin-phonon, and spin-lattice interactions, are not able to "store" information related to formation processes, nor post-depositional events. This characteristic behavior is ascribed to the occurrence of mosaic structure and to the clustering among Mn 2+ and other impurity ions, which are able to induce a strong and variable ligand field interaction. In particular, the proposed method appears fully able to reveal the biogenic origin of microcrystalline calcites and to trace post-depositional events.

    Description: Surfactant protein D (SP-D) modulates the lung's immune system. Its absence leads to NOS2-independent alveolar lipoproteinosis and NOS2-dependent chronic inflammation, which is critical for early emphysematous remodeling. With aging, SP-D knockout mice develop an additional interstitial fibrotic component. We hypothesize that this age-related interstitial septal wall remodeling is mediated by NOS2. Using invasive pulmonary function testing such as the forced oscillation technique and quasistatic pressure-volume perturbation and design-based stereology, we compared 29-wk-old SP-D knockout (Sftpd –/– ) mice, SP-D/NOS2 double-knockout (DiNOS) mice, and wild-type mice (WT). Structural changes, including alveolar epithelial surface area, distribution of septal wall thickness, and volumes of septal wall components (alveolar epithelium, interstitial tissue, and endothelium) were quantified. Twenty-nine-week-old Sftpd –/– mice had preserved lung mechanics at the organ level, whereas elastance was increased in DiNOS. Airspace enlargement and loss of surface area of alveolar epithelium coexist with increased septal wall thickness in Sftpd –/– mice. These changes were reduced in DiNOS, and compared with Sftpd –/– mice a decrease in volumes of interstitial tissue and alveolar epithelium was found. To understand the effects of lung pathology on measured lung mechanics, structural data were used to inform a computational model, simulating lung mechanics as a function of airspace derecruitment, septal wall destruction (loss of surface area), and septal wall thickening. In conclusion, NOS2 mediates remodeling of septal walls, resulting in deposition of interstitial tissue in Sftpd –/– . Forward modeling linking structure and lung mechanics describes the complex mechanical properties by parenchymatous destruction (emphysema), interstitial remodeling (septal wall thickening), and altered recruitability of acinar airspaces.

    Description: The universal second messenger cyclic di-GMP (cdG) is involved in the regulation of a diverse range of cellular processes in bacteria. The intracellular concentration of the dinucleotide is determined by the opposing actions of diguanylate cyclases and cdG-specific phosphodiesterases (PDEs). Whereas most PDEs have accessory domains that are involved in the regulation of their activity, the regulatory mechanism of this class of enzymes has remained unclear. Here, we use biophysical and functional analyses to show that the isolated EAL domain of a PDE from Escherichia coli (YahA) is in a fast thermodynamic monomer-dimer equilibrium, and that the domain is active only in its dimeric state. Furthermore, our data indicate thermodynamic coupling between substrate binding and EAL dimerization with the dimerization affinity being increased about 100-fold upon substrate binding. Crystal structures of the YahA-EAL domain determined under various conditions (apo, Mg2+, cdG·Ca2+ complex) confirm structural coupling between the dimer interface and the catalytic center. The built-in regulatory properties of the EAL domain probably facilitate its modular, functional combination with the diverse repertoire of accessory domains.

    Description: In these studies we determined whether progressive pulmonary inflammation associated with aging in surfactant protein D ( Sftpd ) –/– mice leads to an exacerbated response to ozone. In Sftpd –/– mice, but not wild-type (WT) mice, age-related increases in numbers of enlarged vacuolated macrophages were observed in the lung, along with alveolar wall rupture, type 2 cell hyperplasia, and increased bronchoalveolar lavage protein and cell content. Numbers of heme oxygenase+ macrophages also increased with age in Sftpd –/– mice, together with classically (iNOS+) and alternatively (mannose receptor+, YM-1+, or galectin-3+) activated macrophages. In both WT and Sftpd –/– mice, increasing age from 8 to 27 wk was associated with reduced lung stiffness, as reflected by decreases in resistance and elastance spectra; however, this response was reversed in 80-wk-old Sftpd –/– mice. Ozone exposure (0.8 ppm, 3 h) caused increases in lung pathology, alveolar epithelial barrier dysfunction, and numbers of iNOS+ macrophages in 8- and 27-wk-old Sftpd –/– , but not WT mice at 72 h postexposure. Conversely, increases in alternatively activated macrophages were observed in 8-wk-old WT mice following ozone exposure, but not in Sftpd –/– mice. Ozone also caused alterations in both airway and tissue mechanics in Sftpd –/– mice at 8 and 27 wk, but not at 80 wk. These data demonstrate that mild to moderate pulmonary inflammation results in increased sensitivity to ozone; however, in senescent mice, these responses are overwhelmed by the larger effects of age-related increases in baseline inflammation and lung injury.

    Description: The clinical usage of dendritic cells (DC) for tumor immunotherapy still requires improvements. In this study, three alternative maturation mixtures were compared with the cytokine-based gold standard, and the overall interaction of the resulting DC with effector cells from the innate as well as the adaptive immunity was evaluated in healthy donors. Stimulation with the TLR-4 ligand monophosphoryl lipid A together with IFN- (alt-2 DC) resulted in DC with the highest levels of costimulatory molecule expression and IL-12p70/IL-10 ratio. Whereas all alternative DC were able to induce NK and T cells to acquire cytotoxic properties and secrete type 1 and proinflammatory cytokines, after both short (20-h)- and long (5–8 d)-time coculture, secretion of IFN- by the innate populations was induced in response to alt-2 and alt-1 DC (TNF-α, IFN-α, IFN-, IL-1β, poly IC), but not to alt-3 DC (TNF-α, IFN-, IL-1β, CL097). Regarding CD8 + T cell–mediated Ag-specific immune responses, a heterogeneous pattern of responses was obtained among the healthy donors, suggesting rather a competition than a synergy among the different effector cells. Our data promote further evaluation of alt-2 fast DC for translatability into clinical immunotherapy trials, while also fostering the need to identify biomarkers for immune cell responsiveness and tumor susceptibility to be able to select for each patient the best possible DC-based therapy.
