    Notes: The system of single bunch injection (SBI) for the storage ring NIJI-IV was completed. The system was used for the injection of the short pulse beams from an electron gun into only one of the 16 rf buckets in the ring. On a compact storage ring, the multipulse beams in one pulse train can be stored into only one of many bunches by one injection. The optimum number of the grid pulse in one pulse train was determined to improve the storage efficiency and the storage current by one injection. The keeping time of the bump orbit was calculated by using the transfer matrices. The optimum pulse number obtained by the calculation is 2. In the SBI system, the number of grid pulses in one pulse train can be selected by the gate circuit. A snap off diode and a clip line are used to generate 2 ns grid pulses. The measured grid pulse was shorter than the longitudinal space 5.5 ns of the rf bucket with voltage of −80 V. The EIMAC Y-646B cathode-grid assembly is used as the electron source. It is expected that the injection system provides a higher peak current and easier free electron laser use operation in comparison with the rf-knockout system. © 1995 American Institute of Physics.

    Notes: Micromachining Teflon was achieved by direct exposure to synchrotron radiation and the microstructures made had the smallest surface detail down to 20 μm with structural height of more than 200 μm, that is, aspect ratio on the order of 10. The quality of micromachining Teflon by this process was found to be critically dependent on photon flux of the synchrotron radiation. Analysis of the mass distribution of gaseous species formed upon this process suggested that photochemical processes rather than pyrolytic processes may still dominate. © 1995 American Institute of Physics.

    Notes: Using a lipopolysaccharide (LPS)-responsive murine B-cell line, CH12.LX, we assessed the possible role of CD14 in LPS-induced activation of B cells. Flow cytometric analysis indicated that CH12.LX cells expressed the CD14 molecule with a lower intensity than did the macrophage cell line J774.1. A reverse transcription-polymerase chain reaction and Northern blot analysis revealed low, but significant, levels of CD14 mRNA in CH12.LX cells, whose cDNA was identical to that of the mouse macrophage CD14 gene. After stimulation with LPS, CH12.LX cells proliferated, accompanied by up-regulations of CD14, transforming growth factor (TGF)-β and interleukin (IL)-6 mRNA, and increased IgM and IgA secretion. In the absence of serum or with the addition of anti-CD14 monoclonal antibodies, however, LPS-stimulation induced neither the up-regulation of CD14 and TGF-β mRNA nor an increase in IgA secretion. These findings indicate that CD14 expression is not restricted to myeloid cells, but is involved in some cellular activation events of murine B cells elicited by LPS. Furthermore, a CD14-independent pathway may also exist in the LPS-induced activation of B cells that leads to proliferation, IL-6 production and the enhancement of IgM (but not IgA) secretion.

    Notes: The complete nucleotide sequence of the gene for a cell-surface protein antigen (PAc) of Streptococcus mutans MT8148 (serotype c) was determined. The pac gene consisted of 4695 bp and coded for a 170 773 D protein. The pac gene product contained a putative 38 amino acid signal peptide, resulting in a 166817D mature protein. A potential promoter sequence and a putative Shine-Dalgarno sequence preceded the open reading frame. Two internal repeating amino acid sequences were present in the PAc. One repeating region located in the N-terminal region was rich in alanine, and the other located in the central region was rich in proline. Southern blot analysis under the less stringent condition (allowing up to 35% base mismatch) revealed that the probe covering the prolinerich region hybridized to DNA preparations from strains of Streptococcus cricetus, Streptococcus sobrinus and Streptococcus downei as well as Streptococcus mutans.

    Notes: The structural gene for a 190kD protein antigen (PAc) of Streptococcus mutans MT8148 (serotype c) was cloned into the plasmid vector pUC118. SDS-polyacrylamide gel electrophoresis and Western immunoblotting showed that the Escherichia coli harbouring the chimaeric plasmid produced multiple polypeptides of 190-210kD. Immunodiffusion analysis revealed that the cloned PAc had the same specific determinants as S. mutans PAc. The cloned pac gene was mapped, and its transcriptional orientation was determined by characterizing deletion mutants of the chimaeric plasmid. Southern blot analysis with the cloned gene sequence as a probe revealed the presence of a homologous sequence in DNAs from sero-types e and f S. mutans. PAc-defective mutants were constructed by inserting an erythromycin-resistance gene into the pac gene. The cell-surface hydrophobicity of the mutants was lower than that of the parent strain.

    Notes: Porphyromonas gingivalis fimbriae are critical for the promotion of bacterial infection. The fimA gene encoding fimbrillin, a subunit of fimbriae, has been classified into five genotypes (types I to V) based on their nucleotide sequences. Using a fimA type-specific PCR assay, our previous study demonstrated a close relationship between P. gingivalis possessing type II and type IV fimA genes and adult periodontitis. In that study, some clinical specimens were found to be positive for both types I- and II- fimA specific primers, likely due to the coexistence of two clonal types or a single clone of an unknown genotype in the samples. In the present study, we cloned a new variant of the fimA gene, designated as type Ib fimA, from P. gingivalis HG1691. The nucleotide sequence of the cloned fimA gene showed a 97.1% homology with that of type I fimA, indicating it as a clonal variant of type I fimA. Organisms with type Ib fimA were detected in 13.5% of periodontitis patients and in 2.9% of periodontal healthy adults. Statistical analysis revealed a strong relationship between periodontitis and specific fimA types such as type Ib [odds ratio (OR) 6.51], type II (OR 77.8), and type IV (OR 7.54). Moreover, type Ib fimA-organisms were also found to be related to periodontitis in Down's syndrome (OR 1.91) and mentally disabled populations (OR 4.00). These findings suggest that P. gingivalis with type Ib fimA is closely associated with the progression of periodontitis, similar to organisms with type II and IV fimA.

    Notes: The relative roles of αβ or γδ T cells in protection against Actinobacillus actinomycetemcomitans were analysed by using mutant mice deficient in αβ or γδ T cell receptor (TCR). The dose of A. actinomycetemcomitans producing lethality for the TCR-α deficient mice was 25% of that for control C57BL/6 mice, while that of TCR-δ deficient mice was similar to that of C57BL/6 mice. Reverse transcription (RT)-PCR revealed that γδ T cells from the A. actinomycetemcomitans-infected αβ TCR deficient mice expressed significant levels of interferon (IFN)-γ, interleukin (IL)-5 and IL-6 mRNA, whereas αβ T cells from A. actinomycetemcomitans-infected γδ TCR deficient mice exhibited significant levels of IFN-γ and IL-2 mRNA. Delayed-type hypersensitivity (DTH) reactions against heat shock protein (HSP) 60 were prominently observed in A. actinomycetemcomitans-infected TCR-δ mutant and C57BL/6 mice, but were absent in TCR-α mutant mice, suggesting that the DTH response is exclusively dependent on HSP60-specific αβ T cells producing IFN-γ and IL-2 mRNA. It was found that all the mice used exhibited similar levels of serum IgG and IgM responses against A. actinomycetemcomitans-specific antigens (whole cells and HSP60), suggesting that αβ as well as γδ T cells participate in the serum immune responses. In addition, the humoral antibody responses displayed in the TCR-α deficient mice implies resistance to A. actinomycetemcomitans infection. Thus, the resistance to A. actinomycetemcomitans infection may be ascribed mainly to αβ T cells, while γδ T cells can partially compensate for the αβ T cell defect.

    Notes: The induction of immune responses to orally administered trinitrophenyl (TNP)-haptenated Streptococcus mutans and its enhancement with muramyldipeptide (MDP), peplidoglycan (PG), and concanavalin A (Con A) were investigated in lipopolysaccharide (LPS)-non-responsive C3H/HeJ mice and the syngencic, UPS-responsive C3H/HeN strain. Both mouse strains manifested similar immune responses, primarily of the IgM isotype, after a single gastric intubation (GI) with TNP-S. mutans. However, when groups of animals were first carrier-primed by GI with S. mutans for 2 consecutive days, followed by a single GI with TNP-S. mutans I week later, C3H/HeJ mice gave a significantly higher (P 0.01) splenic IgA anti-TNP plaque-forming cell (PFC) response than identically treated C3H/HeN mice. Furthermore, saliva, urine and serum from these C3H/HeJ mice possessed high levels of IgA anti-TNP antibodies as determined by the enzyme-linked immunosorbent assay, whereas C3H/HeN mice exhibited low antibody levels. Oral administration of Con A (either 250 μg or 500 μg/mouse) or purified PG (1 mg/mouse) at the time of TNp-S. mutans immunization resulted in significantly (P≤ 0.01) enhanced splenic IgA anti-TNP PFC responses, especially in C3H/HeJ mice. On the other hand, MDP promoted IgA anti-TNP PFC responses in LPS-responsive C3H/HeN mice but did not augment responses in C3H/HeJ animals. A similar immune response pattern was seen when antibody levels were measured in serum, saliva, and urine of both mouse strains. These results demonstrate that haptenated S. mutans is a good antigen for the induction of high IgA responses in orally immunized C3H/HeJ mice and that this high response can be enhanced with the adjuvants Con A and PG. However, MDP is ineffective in C3H/HeJ mice but enhances IgA responses in normal LPS-responsive C3H/HeN animals.

    Notes: Reproducible and quantitative evaluation of patient satisfaction with their complete dentures is of great importance for preoperative diagnosis, treatment planning and assessment of complete denture treatment outcome. This study attempted (1) to clarify the degree of contribution of various factors to overall satisfaction, and (2) to develop a method for quantitative assessment of overall satisfaction with complete dentures. Twelve satisfaction factors and a three-grade scale were used to assess 302 complete denture patients. The contribution of each grade of the 12 factors to the overall satisfaction level was determined by multiple regression analysis. Seven factors were highly correlated to the overall satisfaction. Based on the level of contribution by these seven significant factors, a scoring method for satisfaction was established. Category scores for these factors were calculated and the sum of the category scores was converted to an integer between 0 and 100. The resulting quantification score was closely correlated with overall satisfaction. A protocol for scoring overall satisfaction was developed based on the scientifically analysed contribution of each satisfaction factor.

    Notes: Background : The polymorphic enzyme cytochrome P450 2C19 affects omeprazole metabolism. This influence on metabolism might affect serum gastrin levels, and safety, during long-term treatment of reflux oesophagitis.Aim : To examine the relationship between cytochrome P450 2C19 genotype and the safety profile of long-term omeprazole treatment.Methods : A total of 119 Japanese patients with recurrent reflux oesophagitis underwent cytochrome P450 2C19 genotyping prior to receiving daily omeprazole 10 mg or 20 mg for 6–12 months, during which adverse event frequency, serum gastrin levels and endoscopic findings were monitored.Results : The incidences of adverse events, serious adverse events and adverse events leading to withdrawal did not differ between homozygous extensive metabolizer (n = 46), heterozygous extensive metabolizer (n = 53) or poor metabolizer (n = 20) groups. In all genotype groups, serum gastrin increased during the first 3 months of dosing but stabilized thereafter. No significant differences were seen either in the rate of reflux oesophagitis healing or symptom improvement among genotype groups.Conclusions : Long-term treatment with omeprazole was well-tolerated in Japanese patients, irrespective of their cytochrome P450 2C19 metabolic genotype, indicating that dose adjustment depending on metabolic genotype is not required during treatment with omeprazole.
