    Description: Increased natural and anthropogenic stresses have threatened the Earth’s ability to meet growing human demands of food, energy and water (FEW) in a sustainable way. Although much progress has been made in the provision of individual component of FEW, it remains unknown whether there is an optimized strategy to balance the FEW nexus as a whole, reduce air and water pollution, and mitigate climate change on national and global scales. Increasing FEW conflicts in the agroecosystems make it an urgent need to improve our understanding and quantification of how to balance resource investment and enhance resource use efficiencies in the FEW nexus. Therefore, we propose an integrated modeling system of the FEW nexus by coupling an ecosystem model, an economic model, and a regional climate model, aiming to mimic the interactions and feedbacks within the ecosystem-human-climate systems. The trade-offs between FEW benefit and economic cost in excess resource usage, environmental degradation, and climate consequences will be quantitatively assessed, which will serve as sustainability indicators for agricultural systems (including crop production, livestock and aquaculture). We anticipate that the development and implementation of such an integrated modeling platform across world’s regions could build capabilities in understanding the agriculture-centered FEW nexus and guiding policy and land management decision making for a sustainable future.

    Description: Background: To evaluate the effects of contrast agents containing increasing concentrations of iodine on the renal oxygenation level determined by blood oxygenation level dependent (BOLD) magnetic resonance imaging (MRI) in a rabbit model of diabetic nephropathy. Methods: BOLD-MRI was performed using saline or iodinated (I) contrast agents (200, 240, 300, 350 and 400 mg I/mL) at 1, 24, 48, and 72 h after experimentally inducing type 2 diabetic nephropathy in rabbits. Differences in renal oxygenation levels between type 1 and type 2 diabetic nephropathy were also assessed by BOLD-MRI after injecting 400 mg I/mL of contrast agent. Results: Contrast agents increased the R2* values of the renal cortex, outer medulla, and inner medulla to the maximum levels at 24 h. The R2* values then decreased to their lowest levels at 72 h. The R2* was highest following injection of 400 mg I/mL, especially in the outer medulla. The R2* values were not significantly different between types 1 and 2 diabetic nephropathy. Conclusions: Iodinated contrast agents had the greatest influence on renal outer medulla oxygenation level at 24 h in type 2 diabetic nephropathy, with the greatest effects observed at the 400 mg I/mL dose level. There were no differences in BOLD-MRI values between type 1 and type 2 diabetic nephropathy after administering the contrast agent at 400 mg I/mL.

One of the pressing requirements from the user community is an improved ability to detect small fires (less than 50 ha), whose impact on terrestrial environments is empirically known but poorly quantified, and is often excluded from global earth system models. The newest generation of BA algorithms combines the capabilities of both the BA and AF detection approaches, resulting in a general improvement of detection compared to their predecessors. Accuracy assessments of these products have been done in several ecosystems; but more complex ones, such as regions that are characterized by frequent small fires and steep terrain has never been assessed. This study contributes to the understanding of the performance of global BA and AF products with a first assessment of four selected datasets: MODIS-based MCD45A1; MCD64A1; MCD14ML; and, ESA’s Fire_CCI in a mountainous region of northwest Yunnan; P.R. China. Due to the medium to coarse resolution of the tested products and the reduced sizes of fires (often smaller than 50 ha) we used a polygon intersection assessment method where the number and locations of fire events extracted from each dataset were compared against a reference dataset that was compiled using Landsat scenes. The results for the two sample years (2006 and 2009) show that the older, non-hybrid products MCD45A1 and, MCD14ML were the best performers with Sørensen index (F1 score) reaching 0.42 and 0.26 in 2006, and 0.24 and 0.24 in 2009, respectively, while producer’s accuracies (PA) were 30% and 43% in 2006, and 16% and 47% in 2009, respectively. All of the four tested products obtained higher probabilities of detection when smaller fires were excluded from the assessment, with PAs for fires bigger than 50 ha being equal to 53% and 61% in 2006, 41% and 66% in 2009 for MCD45A1 and MCD14ML, respectively. Due to the technical limitations of the satellites’ sensors, a relatively low performance of the four products was expected. Surprisingly, the new hybrid algorithms produced worse results than the former two. Fires smaller than 50 ha were poorly detected by the products except for the only AF product. These findings are significant for the future design of improved algorithms aiming for increased detection of small fires in a greater diversity of ecosystems.

    Description: Previous studies have shown that promoter hypermethylation plays a key role in DLC-1 inactivation in nasopharyngeal carcinoma (NPC). However, DLC-1 mutation in NPC has not been reported, and there remain some discrepancies in methods and results between different groups. Here, we examined the mRNA and protein expression of DLC-1 in chronic nasopharyngitis (CN) and NPC tissues by reverse transcription-polymerase chain reaction/qPCR and immunohistochemistry, respectively. DLC-1 mRNA was undetectable in all the seven widely used NPC cell lines and absent or significantly down-regulated in 70% of NPC tissues. DLC-1 protein level was reduced in 74.3% of NPCs when compared with CN tissues, and significantly lower in NPC samples at advanced clinical stages than that at early stages. Then, we purified the same batch of specimens by microdissection and analyzed the possible mechanisms of DLC-1 downregulation with mutation and allelic loss analysis, methylation-specific PCR and bisulfite genomic sequencing. Only one mutation was detected at codon 693 of exon 8 in 3.3% of NPCs and five single nucleotide polymorphisms (SNPs) were identified. Loss of DLC-1 was detected in 23.3% of NPC tissues. The 100% of NPC cell lines, 80% of primary NPC and 22.2% of CN tissues showed methylation in DLC-1 promoter, while DLC-1 expression was recovered in seven NPC cell lines after 5-aza-dC treatment. Patched methylation assay confirmed that promoter methylation could repress DLC-1 expression. This report demonstrates that DLC-1 is negatively associated with NPC carcinogenesis, and promoter hypermethylation along with loss of heterozygosity, but not mutation, contributes to inactivation of DLC-1 in NPC. © 2013 Wiley Periodicals, Inc.

    Description: Background: The European (EU) genotype of porcine reproductive and respiratory syndrome virus (Genotype-I PRRSV) has recently emerged in China. The coexistence of Genotype-I and -II PRRSV strains could cause seriously affect PRRSV diagnosis and management. Current vaccines are not able to protect against PRRSV infection completely and have inherent drawbacks. Thus, genetically engineered vaccines, including DNA vaccine and live vector engineered vaccines, have been developed. This study aimed to determine the enhanced immune responses of mice inoculated with a DNA vaccine coexpressing GP3 and GP5 of a Genotype-I PRRSV. Results: To evaluate the immunogenicity of GP3 and GP5 proteins from European-type PRRSV, three DNA vaccines, pVAX1-EU-ORF3-ORF5, pVAX1-EU-ORF3 and pVAX1-EU-ORF5, were constructed, which were based on a Genotype-I LV strain (GenBank ID: M96262). BALB/c mice were immunized with the DNA vaccines; delivered in the form of chitosan-DNA nanoparticles. To increase the efficiency of the vaccine, Quil A (Quillaja) was used as an adjuvant. GP3 and GP5-specific antibodies, neutralizing antibodies and cytokines (IL-2, IL-4, IL-10 and IFN gamma) from the immunized mice sera, and other immune parameters, were examined, including T-cell proliferation responses and subgroups of spleen T-lymphocytes. The results showed that ORF3 and ORF5 proteins of Genotype-I PRRSV induced GP3 and GP5-specific antibodies that could neutralize the virus. The levels of Cytokines IL-2, IL-4, IL-10, and IFN-gamma of the experimental groups were significantly higher than those of control groups after booster vaccination (P 〈 0.05). The production of CD3+CD4+ and CD3+CD8+ T lymphocyte was also induced. T lymphocyte proliferation assays showed that the PRRSV LV strain virus could stimulate the proliferation of T lymphocytes in mice in the experimental group. Conclusions: Using Quil A as adjuvant, Genotype-I PRRSV GP3 and GP5 proteins produced good immunogenicity and reactivity. More importantly, better PRRSV-specific neutralizing antibody titers and cell-mediated immune responses were observed in mice immunized with the DNA vaccine co-expressing GP3 and GP5 proteins than in mice immunized with a DNA vaccine expressing either protein singly. The results of this study demonstrated that co-immunization with GP3 and GP5 produced a better immune response in mice.

    Description: Background: IgG4-related kidney disease is a comprehensive term for renal lesions associated with IgG4-related disease, which mainly manifests as plasma cell-rich tubulointerstitial nephritis with increased IgG4+ plasma cells and fibrosis. IgG4-related kidney disease in the renal pelvis is rare.Case presentationWe describe a 53-year-old Asian woman who was referred to our hospital with a space-occupying renal lesion discovered by medical examination. A physical examination and laboratory evaluation revealed no significant abnormalities. Computed tomography scans showed a soft-tissue mass with an irregular border and mild homogeneous enhancement in the right renal pelvis and calyces. A positron emission tomography/computed tomography scan revealed soft-tissue density shadows with increased radionuclide uptake. To investigate a suspected pelvic carcinoma, a right ureteronephrectomy was performed. A pathologic examination of the renal sections showed a dense lymphoplasmacytic infiltrate rich in IgG4+ plasma cells, with fibrosis beneath the urothelial epithelium of the renal pelvis. Postoperatively, the serum IgG4 level was significantly elevated. The patient was diagnosed with IgG4-related kidney disease. Conclusion: We present a case of IgG4-related kidney disease mimicking urothelial carcinoma in the renal pelvis. When a buried and solitary hypovascular tumor is detected in the kidney, we must consider IgG4-related kidney disease as a differential diagnosis. Accordingly, elevated serum IgG4, radiologic findings, and pathologic examination may improve the diagnosis.
