    Description: Macrobenthos of the deep, northern Gulf of Mexico (GoM) was sampled with box cores (0.2 m2) along multiple cross-depth transects extending from depths of 200 m to the maximum depth of the basin at 3700 m. Bathymetric (depth) zonation of the macrofaunal community was documented for 6 major taxa (a total of 957 species) on the basis of shared species among geographic locations; 4 major depth zones were identified, with the 2 intermediate-depth zones being divided into east and west subzones. Change of faunal composition with depth reflects an underlying continuum of species replacements without distinct boundaries. The zonal patterns correlated with depth and detrital particulate organic carbon (POC) export flux estimated from remotely-sensed phytoplankton pigment concentrations in the surface water. The Mississippi River and its associated mesoscale eddies, submarine canyon, and deep sediment fan appear to influence the horizontal zonation pattern through export of organic carbon from the ocean surface and the adjacent continental margin. On the local scale, near-bottom currents may shape the zonation pattern by altering sediment grain size, food availability, and larval dispersal. This study suggests a macroecological relationship between depth, export POC flux, and zonation; parsimonious zonal thresholds need to be tested independently for other continental margin ecosystems.

    Description: The TetraEther indeX of 86 carbon atoms (TEX86) is widely used as a proxy to reconstruct past sea surface temperatures. Most current applications of TEX86 are primarily based on analyzing the composition of isoprenoid glycerol dialkyl glycerol tetraethers (isoGDGTs) that comprise TEX86 in sediments, with the assumption that the sedimentary isoGDGTs are mainly derived from the surface mixed layer. Here we report on the variations in the isoGDGT distribution, archaeal abundance and community through the water column of the Western Pacific Ocean, directly testing the export depth of isoGDGTs and constraining the temperature records of TEX86. Our data show that maximum isoGDGT concentrations occurred in subsurface waters (150–200 m) with maximum archaeal abundances. The ratio between isoGDGTs bearing 2 vs. 3 cyclopentane moieties, i.e. [2/3] ratio, increased with depth, which is likely related to the shift of the archaeal community from Ca. Nitrosopelagicus-dominance to norank_f__Nitrosopumilaceae-dominance. Models based on the [2/3] ratios in the water column predicted an average export depth of isoGDGTs to sediments of around 150–200 m, consistent with the robust relationship between the compiled sedimentary TEX86 and the annual mean subsurface temperature. Taken together, our findings support that TEX86 records subsurface rather than surface temperatures in the open ocean.

    Description: Purpose The regularly incremented phase encoding–magnetic resonance fingerprinting (RIPE-MRF) method is introduced to limit the sensitivity of preclinical MRF assessments to pulsatile and respiratory motion artifacts. Methods As compared to previously reported standard Cartesian–MRF methods (SC-MRF), the proposed RIPE-MRF method uses a modified Cartesian trajectory that varies the acquired phase-encoding line within each dynamic MRF dataset. Phantoms and mice were scanned without gating or triggering on a 7T preclinical MRI scanner using the RIPE-MRF and SC-MRF methods. In vitro phantom longitudinal relaxation time (T 1 ) and transverse relaxation time (T 2 ) measurements, as well as in vivo liver assessments of artifact-to-noise ratio (ANR) and MRF-based T 1 and T 2 mean and standard deviation, were compared between the two methods (n = 5). Results RIPE-MRF showed significant ANR reductions in regions of pulsatility ( P  〈 0.005) and respiratory motion ( P  〈 0.0005). RIPE-MRF also exhibited improved precision in T 1 and T 2 measurements in comparison to the SC-MRF method ( P 〈  0.05). The RIPE-MRF and SC-MRF methods displayed similar mean T 1 and T 2 estimates (difference in mean values 〈 10%). Conclusion These results show that the RIPE-MRF method can provide effective motion artifact suppression with minimal impact on T 1 and T 2 accuracy for in vivo small animal MRI studies.     Description: Activation of the Wnt/β-catenin signaling pathway drives colorectal cancer growth by deregulating expression of downstream target genes, including the c-myc proto-oncogene. The critical targets that mediate the functions of oncogenic c-Myc in colorectal cancer have yet to be fully elucidated. Previously, we showed that activation of PI3K/Akt/mTOR contributes to colorectal cancer growth and metastasis. Here, we show that Deptor, a suppressor of mTOR, is a direct target of Wnt/β-catenin/c-Myc signaling in colorectal cancer cells. Inhibition of Wnt/β-catenin or knockdown of c-Myc decreased, while activation of Wnt/β-catenin or overexpression of c-Myc increased the expression of Deptor. c-Myc bound the promoter of Deptor and transcriptionally regulated Deptor expression. Inhibition of Wnt/β-catenin/c-Myc signaling increased mTOR activation, and the combination of Wnt and Akt/mTOR inhibitors enhanced inhibition of colorectal cancer cell growth in vitro and in vivo. Deptor expression was increased in colorectal cancer cells; knockdown of Deptor induced differentiation, decreased expression of B lymphoma Mo-MLV insertion region 1 (Bmi1), and decreased proliferation in colorectal cancer cell lines and primary human colorectal cancer cells. Importantly, our work identifies Deptor as a downstream target of the Wnt/β-catenin/c-Myc signaling pathway, acting as a tumor promoter in colorectal cancer cells. Moreover, we provide a molecular basis for the synergistic combination of Wnt and mTOR inhibitors in treating colorectal cancer with elevated c-Myc.Significance: The mTOR inhibitor DEPTOR acts as a tumor promoter and could be a potential therapeutic target in colorectal cancer.
