    Notes: The climatic features of Antarctic waters are more extreme and constant than in the Arctic. The Antarctic has been isolated and cold longer than the Arctic. The polar ichthyofaunas differ in age, endemism, taxonomy, zoogeographic distinctiveness and physiological tolerance to environmental parameters. The Arctic is the connection between the Antarctic and the temperate-tropical systems. Paradigmatic comparisons of the pathways of adaptive evolution of fish from both poles address the oxygen-transport system and the antifreezes of northern and southern species, (i) Haemoglobin evolution has included adaptations at the biochemical, physiological and molecular levels. Within the study of the molecular bases offish cold adaptation, and taking advantage of the information on haemoglobin amino acid sequence, we analysed the evolutionary history of the α and β globins of Antarctic, Arctic and temperate haemoglobins as a basis for reconstructing phylogenetic relationships. In the trees, the constant physico-chemical conditions of the Antarctic waters are matched by clear grouping of globin sequences, whereas the variability typical of the Arctic ecosystem corresponds to high sequence variation, reflected by scattered intermediate positions between the Antarctic and non-Antarctic clades. (ii) Antifreeze (glyco)proteins and peptides allow polar fish to survive at sub-zero temperatures. In Antarctic Notothenioidei the antifreeze gene evolved from a trypsinogen-like serine protease gene. In the Arctic polar cod the genome contains genes which encode nearly identical proteins, but have evolved from a different genomic locus–a case of convergent evolution.

    Notes: Abstract The distribution of metallothionein and metallothionein mRNA was examined in brain, liver and kidney of the red-blooded Antarctic fish Notothenia coriiceps. Basal levels of metallothionein were detected by silver saturation assay; the analysis showed large differences in the metallothionein content, with an increasing amount in kidney, liver and brain, respectively. Reverse transcriptase-mediated polymerase chain reaction was used to amplify two distinct metallothionein cDNA species from RNA extracted from the three tissues examined. Northern blot analysis revealed a high constitutive expression of metallothionein mRNA in kidney, in spite of the low metallothionein content found in this tissue. The analysis of the distribution of the two metallothionein transcripts, termed MT-1 and MT-2, in the tissues showed that the MT-2 transcript was the major form of the metallothionein mRNA in kidney and brain, whilst in liver the expression levels of the two metallothionein transcripts were comparable. This tissue-specific expression of isometallothioneins in the Antarctic fish suggests the presence of an organ-specific regulation of the metallothionein genes.

    Notes: Abstract The presence of an acetyl blocking group at theN-terminus of the coat protein of papaya mosaic virus has been identified by FAB mass spectrometry. Furthermore, we have found that theN-terminal sequence of the protein is four amino-acid residues (AC-Ser-Lys-Ser-Ser-) longer than that previously reported, while Glu instead of Gln is theC-terminal residue. The present paper shows that PMV-protein is made up of 215 amino acid residues, with a molecular mass of 22,960 Da.

    Description: Background: Multidrug-resistant Acinetobacter baumannii, initially considered as having a poor clinical relevance, is frequently isolated from infection cases in intensive care units. We describe the epidemiology of carbapenem resistant A. baumannii (CRAB) in a general ICU in Palermo, Italy, from October 2010 to March 2011.Findings58 of 61 isolates exhibited MICs for meropenem or imipenem [greater than or equal to]16 mg/L. Forty-nine carried blaOXA-23 and two blaOXA-58 genes.Five subtype clusters were detected by rep-PCR. Clusters D and E included 10 isolates that tested negative for the carbapenem resistance genes. MLST attributed all isolates, but two, with sequence type (ST)2, whereas the two remaining isolates with ST78.The respiratory tract was the most common site of infection (26 out of 36 cases. 72.2%). A high infection related mortality rate was observed (18 out of 35 patients, 51.4%). Nineteen patients tested positive for other multidrug resistant organisms in addition to CRAB. In eight cases isolates belonging to distinct subtype clusters and/or with distinct carbapenemase profiles were identified. Conclusions: Carbapenem resistance was prominently driven by the dissemination of CRAB isolates belonging to ST2, carrying the carbapenemase gene blaOXA-23. The colonization/infection of some patients by multiple strains is suggestive of an endemic circulation of CRAB.

    Description: Stresses on Antarctic ecosystems result from environmental change, including extreme events, and from (other) human impacts. Consequently, Antarctic habitats are changing, some at a rapid pace while others are relatively stable. A cascade of responses from molecular through organismic to the community level are expected. The differences in biological complexity and evolutionary histories between both polar regions and the rest of the planet suggest that stresses on polar ecosystem function may have fundamentally different outcomes from those at lower latitudes. Polar ecosystem processes are therefore key to informing wider ecological debate about the nature of stability and potential changes across the biosphere. The main goal of AnT-ERA is to facilitate the science required to examine changes in biological processes in Antarctic and sub-Antarctic marine-, freshwater and terrestrial ecosystems. Tolerance limits, as well as thresholds, resistance and resilience to environmental change will be determined. AnT-ERA is classified into three overlapping themes, which represent three levels of biological organisation: (1) molecular and physiological performance, (2) population processes and species traits, (3) ecosystem function and services.
