    Beschreibung / Inhaltsverzeichnis: Aufgrund der hohen Konzentration von Bevölkerung, ökonomischen Werten und Infrastrukturen können Städte stark von extremen Wetterereignissen getroffen werden. Insbesondere Hitzewellen und Überflutungen in Folge von Starkregen verursachen in Städten immense gesundheitliche und finanzielle Schäden. Um Schäden zu verringern oder gar zu vermeiden, ist es notwendig, entsprechende Vorsorge- und Klimaanpassungsmaßnahmen zu implementieren. Im Projekt „Urbane Resilienz gegenüber extremen Wetterereignissen – Typologien und Transfer von Anpassungsstrategien in kleinen Großstädten und Mittelstädten” (ExTrass) lag der Fokus auf den beiden extremen Wetterereignissen Hitze und Starkregen sowie auf kleineren Großstädten (100.000 bis 500.000 Einwohner:innen) und kreisfreien Mittelstädten mit mehr als 50.000 Einwohner:innen. Im Projekt wurde die Stärkung der Klimaresilienz als Verbesserung der Fähigkeiten von Städten, aus vergangenen Ereignissen zu lernen sowie sich an antizipierte Gefahren anzupassen, verstanden.     Notizen: Abstract Local irradiation of the alga Vaucheria sessilis (Vauch.) D.C. with blue light, which stimulates cortical fiber reticulation and chloroplast aggregation (M.R. Blatt and W.R. Briggs, 1980, Planta 147, 355–362), also induces an outward-directed current from the irradiated region of the cell. This current appears in conjunction with cortical fiber reticulation and precedes chloroplast aggregation. The current is not photosynthetic in origin, as indicated by experiments with 3(3,4-dichlorophenyl)-1,1-dimethyl urea and carbonyl-cyanide-m-chlorophenylhydrazone (CCCP). It shows a wavelength-dependence similar to that of chloroplast aggregation and reaches a maximum of 500 nA cm-2 with saturating light intensities. The current is not dependent upon the presence of Na+, K+, or Cl- in a test medium containing only Na+, K+, Ca2+, and Cl-, but is inhibited, apparently nonspecifically, in the absence of external calcium. Both the light-induced current and chloroplast aggregation are stimulated by increases in the external KCl concentration and are inhibited by sub-micromolar concentrations of CCCP or by external pHs below approximately 5.5. We suggest that blue light stimulates the local extrusion of cations, possibly of protons, at the plasma membrane, an event which may act to destabilize the cortical fibers in Vaucheria, disrupt cytoplasmic streaming, and eventually lead to organelle aggregation in the light.

    Notizen: Abstract Spores of the ferns, Dryopteris filix-mas, D. paleacea and Polystichum minutum, sown on plain agar in quartz-distilled water, required several hours of red light in order to germinate. When, however, water agar was replaced by agar made up with a mineral nutrition medium, a single pulse of red light (about 1 min) was able fully to induce germination. Under these conditions spores became light-sensitive a few minutes after sowing. Thus, zero germination in dark controls was obtained only when all light was excluded immediately after sowing or when saturating far-red was given thereafter. The effect of the mineral medium was also obtained using low ion concentrations with an osmolality of less than 100 μmol l−1. Thus, a specific ion effect appears more probable than an unspecific osmotic effect. Species differences in light sensitivity and in dark-germination levels, as reported in the literature, might partly be the consequence of different culture media and of light acting at a very early stage after sowing, which hitherto was assumed to be still insensitive to light. On water agar as well as on mineral agar, the inducing effect of a single red pulse could be increased by the appropriate pretreatment, i.e. by preirradiation with red light for several hours, followed by a saturating pulse of far-red, the latter abolishing the direct inducing effect of the red preirradiation. The nature of both the ion-phytochrome interaction and the phytochrome-phytochrome interaction has not yet been analysed.

Observations on the orientation of the chloroplast following such irradiation give evidence that during photoconversion the axis of dichroism of the phytochrome shifts: PR absorbs parallel and PFR normal to the surface of the cell.

    Notizen: Abstract Chloroplast orientation in the green alga Mougeotia has been induced by unidirectional red or blue light, given continuously during one hour. In addition, part of the preparations obtained scattered strong far-red light simultaneously with the orienting light. This far-red light completely abolished the response to red light, consistent with phytochrome as the sensor pigment for orientation in Mougeotia. In blue light, however, the response was completely insensitive to far-red light, thus pointing to a different sensor pigment in the shortwavelength region.

    Notizen: Summary Mougeotia cells with chloroplasts oriented in profile have been irradiated with small spots of monochromatic red polarized light in order to induce chloroplast movement. In these experiments, four factors have been varied: 1. the orientation of the vibration plane of the light in relation to the cell axis, 2. the localization of the spot, i. e. irradiation of the chloroplast or the cytoplasm, 3. the spot size, and 4. the duration of the irradiation. As a result of our experiments, we conclude that the photoreceptor molecules responsible for the light-induced chloroplast movement are localized in the cytoplasm. As the photoreceptor of this reaction is the well known phytochromesystem, we may assume that also in other plants the phytochrome is localized in the cytoplasm rather than in the chloroplast.

    Notizen: Summary By means of partial illumination with small spots of monochromatic light, polarized obliquely to the cell axis, we could demonstrate that the phytochrome molecules inMougeotia are oriented in a spiral pattern around the cell surface. The lines of this spiral form an angle of about 45o with the cell axis. The degree of exactness of this orientation has been discussed.
