    Beschreibung: T-antigen (Galβ1-3GalNAcα-1-Ser/Thr), also known as Thomsen–Friedenreich antigen (TF antigen), is an oncofetal antigen commonly found in cancerous tissues. Availability of anti-T-antigen human antibodies could lead to the development of cancer diagnostics and therapeutics. Four groups of single-chain variable fragment (scFv) genes were previously isolated from a phage library (Matsumoto-Takasaki et al. (2009) Isolation and characterization of anti-T-antigen single chain antibodies from a phage library. BioSci Trends 3:87–95.). Here, four anti-T-antigen scFv genes belonging to Group 1–4 were expressed and produced in a Drosophila S2 cell expression system. ELISA and surface plasmon resonance (SPR) analyses confirmed the binding activity of 1E8 scFv protein to various T-antigen presenting conjugates. NMR experiments provided evidence of the folded nature of the 1E8 scFv protein. ScFv-ligand contact was identified by STD NMR, indicating that the galactose unit of T-antigen at the non-reducing end was primarily recognized by 1E8 scFv. This thus provides direct evidence of T-antigen specificity.

    Notizen: Background Several extracellular matrix genes, most notably α1(I) and α1(III) procollagen, are reported to be co-ordinately expressed in cultures of dermal fibroblasts. However, it remains unclear whether the expression of these genes is truly co-ordinate or whether it may be the result of averaging the phenotypic expression of different fibroblast subpopulations present within each culture. Objectives To determine by Northern analysis the correlation between α1(I) and α1(III) procollagen mRNA levels in clonal populations of human dermal fibroblasts. Methods As previously described, clonal cultures were derived from parent strains of human dermal fibroblasts by a microscopically controlled dilution technique and by stimulation of single cells with low oxygen tension in the early phases of clonal growth. Results In agreement with previous reports, we found that baseline steady-state levels of α1(I) procollagen mRNA were co-ordinately regulated with the α1(III) procollagen mRNA in 26 parent strains (r = 0·9003; P 〈 0·0001). However, this close correlation between the expression of these two procollagen chains was absent in a total of 40 unselected clonal strains derived from four of the parent cultures (r = 0·5745; P 〈 0·0001). Moreover, this intrachain heterogeneity in α1(I) and α1(III) procollagen mRNA levels in clonal cultures was statistically significant from that measured in parent strains (P = 0·0016). Conclusions α1(I) and α1(III) procollagen mRNA levels in clonal cultures do not show the tight co-ordinate regulation observed in non-clonal cultures, suggesting that these two genes operate under different sets of regulatory controls. This clonal heterogeneity may provide additional flexibility to the process of tissue repair and fibroblast clonal expansion.

    Notizen: In order to determine whether adrenal and gonadal weak androgens are utilized to form active androgens in human hair, we studied the expression of mRNA for androgen receptor (AR), 5α-reductase and 17β-hydroxysteroid dehydrogenase (17β-HSD) in cultured dermal papilla cells (DPCs) from various body sites. AR mRNA was expressed in beard (Be) and axillary hair (Ax) DPCs from both sexes, but only at a low level in DPCs from occipital scalp hair (OS). Type I 5α-reductase mRNA was expressed by all DPCs. Type II 5α-reductase mRNA was identified in beard DPCs, but was absent from OS and Ax DPCs. Type II 17β-HSD mRNA was strongly expressed in outer root sheath cells (ORSCs), while DPCs except for male Ax expressed no type II 17β-HSD mRNA. In contrast, type III 17β-HSD mRNA was strongly expressed in Be DPCs and Ax DPCs from both sexes; ORSCs showed a low level of expression. Expression of type III 17β-HSD mRNA was not regulated by androgen in DPCs. These results suggest that the sensitivity of hairs to androgen is partially controlled by the site-specific expression of AR, 5α-reductase and 17β-HSD in DPCs.

    Notizen: Cubic Laves phase alloy YFe2 forms four types of hydrides, YFe2Hx: β1, β2 (hexagonal) of 1.2〈x〈1.6, γ (hexagonal) of 2.8〈x〈3.45, and δ (orthorhombic) of 3.7〈x. The β1 and β2 hydrides are formed in the hydrogen absorption below and above 500 K. The deuterides YFe2Dx have been prepared by deuterium absorption at various temperatures for YFe2 powders of diameters less than 0.3 mm. The formed samples are mixtures of main and extra deuteride phases. The x-ray and magnetization measurements in a magnetic field of 10.6 kOe showed five types of ferromagnetic deuterides with Curie temperatures of about 100 K (δ1), 300 K (δ2), 380 K (γ), 300–380 K (β1), and 540 K (amorphous). The extrapolated values of magnetization at 0 K are 3.5μB/YFe2Dx. The crystal structures are as follows: δ1, hexagonal, a=0.57 nm, c=2.74 nm; δ2, cubic, a=0.79 nm; γ, hexagonal, a=0.56 nm, c=2.63 nm; and β, hexagonal (a=0.53 nm, c=0.87 nm)–(a=0.51 nm, c=0.82 nm). © 1996 American Institute of Physics.

    Notizen: Resonant tunneling bipolar transistors (RBTs) using InAlAs/InGaAs heterostructures were fabricated. These devices are bipolar transistors which use a resonant tunneling barrier as a minority-carrier injector. The RBT exhibits a collector current peak as a function of the base-emitter voltage at room temperature. The peak-to-valley ratio of the collector current is 3.5, and the peak collector current density is 5.7×104 A/cm2. The common-emitter current gain reaches a value of 24. These InAlAs/InGaAs RBTs characteristics are much better than those of AlGaAs/GaAs RBTs. We measured the microwave characteristics of the InAlAs/InGaAs RBT at room temperature, and obtained a cutoff frequency of 12.4 GHz. An equivalent circuit analysis and device simulation yielded an estimated resonant tunneling barrier response time of 1.4 ps.

    Notizen: Inelastic scattering of neutrons has been used to study the magnetic excitations in Fe-doped Mn3Si at 7 K. Mn3Si is an incommensurate antiferromagnet with magnetic Bragg points of the form (h,k,l)±(δ,δ,δ) with δ=0.425. Its magnetic excitations are qualitatively similar to those of the γ-Mn alloys, or of the itinerant antiferromagnet, chromium. The addition of 5 at. % of iron causes the value of δ to change to 1/2, so that the material becomes a commensurate antiferromagnet, greatly simplifying the shape of the excitation spectrum. In addition the low Néel temperature (23 K) permits measurements to be extended to energies of several times kTN. Constant energy scans through a magnetic Bragg point are qualitatively consistent with the spin-wave dispersion relation forming a "cone'' around the magnetic Bragg point. Low-energy scans (1 meV≤ΔE≤4 meV) are centered on the Bragg point and flat-topped, while at higher energies (5 meV≤ΔE≤15 meV) the peak is clearly resolved as double. A fit using a damped-harmonic oscillator cross section suggests that the slope of the dispersion relation increases with energy, and that the excitations are highly damped.

    Notizen: 1. We compared the serum and aortic lipid levels in spontaneously hypertensive rats (SHR) and Dahl salt-sensitive rats (DSR) fed a high-cholesterol (HC) diet.2. In SHR fed the HC diet, the serum cholesterol level significantly increased, but no aortic cholesterol deposition was observed.3. The serum cholesterol level in DSR fed the HC diet markedly increased compared to that in DSR fed the basal diet, and this change was greater with the diet containing 8% NaCl than 0.4% NaCl. A significant increase in the content of aortic cholesterol, notably cholesteryl ester, was observed in only DSR fed the HC diet containing 8% NaCl.4. These results suggest that the combination of hyper-cholesterolaemia with salt-induced hypertension acts as a greater risk factor for atherosclerosis than that with genetic hypertension.
