    Notes: Summary Human α-atrial natriuretic peptide (h-αANP) makes the urine of dehydrated volunteers hypotonic to plasma despite high circulating concentrations of antidiuretic hormone. Urinary dilution with h-αANP also occurs in subjects receiving indomethacin. Therefore, h-αANP antagonises effects of antidiuretic hormone on distal tubular V2-receptors in man, probably without involving prostaglandins.

    Notes: Summary The acute natriuretic effect of human atrial natriuretic peptide (ANP) has been well described in man. We have now studied possible hormonal mediators of this effect. We studied six healthy volunteers on two occasions when they received either an infusion of ANP of 1.5 pmol·kg−1·min−1 for 30 min followed by 15 pmol·kg−1·min−1 for a further 30 min, or matching vehicle infusions in a randomized single-blind fashion. On the placebo day, plasma renin activity (PRA) rose from 1.26±0.08 to 1.57±0.14 ng A1·ml−1·h−1, while on the ANP study day PRA fell from 1.45±0.15 to 1.28±0.05 ng A1·ml−1·h−1 (p〈0.01). No significant changes were found in plasma aldosterone concentrations or in urinary dopamine excretion. These results provide evidence that ANP suppresses renin release in man.

    Notes: Summary Six specific pathogen-free foals shown to be free of equine herpesvirus-1 and 4 (EHV-1 and -4) and lacking in maternally-derived antibodies were used to investigate the pathogenesis of EHV-1 in horses. Following primary intranasal inoculation with EHV-1 all foals showed signs of a mild, self-limiting upper respiratory tract infection. A leucopenia was observed, comprising both a lymphopenia and neutropenia. Virus was isolated from nasal mucus and buffy coat cells over several days during the clinical episode and after the animals became clinically normal. Notwithstanding the mildness of the clinical disease, virus was not eliminated completely and intravenous administration of dexamethasone resulted in reactivation of latent EHV-1 in animals which had received only a single dose of the virus. In a second infection given to four foals, 61 days after the primary inoculation, no clinical signs were observed, haematological changes were minimal and viraemia was absent.

    Notes: Summary EHV-1 was inoculated into specific pathogen-free (SPF) foals in order to study uncomplicated primary responses. Infection resulted in a strong serological response recognizing EHV-1-specific antigens; this contrasts with a previous publication where a weak response was recorded in SPF animals. Antibodies to EHV-1 were readily detected by four techniques (virus neutralization, complement fixation, Western blots and immune precipitation), yet there was comparatively little cross-reaction to EHV-4 target antigen. Re-in-oculation with the same virus strain stimulated antibodies to EHV-1 but no additional antigens were recognized and antibodies cross-reacting with EHV-4 antigens were not enhanced. Having characterized the uncomplicated primary response to EHV-1 in SPF foals, further animals were exposed to either EHV-4 or a thymidine kinase-deficient mutant of EHV-1 prior to challenge with w/t EHV-1 to investigate how these infections might modulate the immune responses to EHV-1 or 4. Primary inoculation with EHV-4 or with a thymidine kinase-deficient mutant of EHV-1 produced productive infections as evidenced by virus shedding and pyrexia. In both these cases, however, in contrast to that with w/t EHV-1, the serological response was very weak. Re-infection of foals primed with either EHV-4 or TK-deficient EHV-1 with w/t EHV-1 resulted in a strong response to EHV-1 antigens detected by all four methods. In addition, in the foals given a primary inoculation with EHV-4, superinfection with EHV-1 resulted in a strong cross-reactive response to EHV-4 target antigens. The relevance of these observations to the interpretation of previously reported serological responses to EHVs in SPF and naturally reared animals is discussed.

    Description: The lack of long-term, highly resolved (annual to sub-annual) and absolutely dated baseline records of marine variability extending beyond the instrumental period (last ~50-100 years) hinders our ability to develop a comprehensive understanding of the role the ocean plays in the climate system. Specifically, without such records, it remains difficult to fully quantify the range of natural climate variability mediated by the ocean, and to robustly attribute recent changes to anthropogenic or natural drivers. Here we present a 211-year (1799-2010 CE; all dates hereafter are common era) seawater temperature (SWT) reconstruction from the northeast Atlantic Ocean derived from absolutely dated, annually resolved, oxygen isotope ratios recorded in the shell carbonate (δ 18 O shell ) of the long-lived marine bivalve mollusc Glycymeris glycymeris . The annual record was calibrated using sub-annually resolved δ 18 O shell values drilled from multiple shells covering the instrumental period. Calibration verification statistics and spatial correlation analyses indicate that the δ 18 O shell record contains significant skill at reconstructing Northeast Atlantic Ocean mean summer SWT variability associated with changes in sub-polar gyre (SPG) dynamics and the North Atlantic Current. Reconciling differences between the δ 18 O shell data and corresponding growth increment width chronology demonstrates that 68% of the variability in G. glycymeris shell growth can be explained by the combined influence of biological productivity and SWT variability. These data suggest G. glycymeris can provide seasonal to multi-centennial absolutely dated baseline records of past marine variability that will lead to the development of a quantitative understanding of the role the marine environment plays in the global climate system.
