    Description: Model simulations of the last glacial maximum (LGM) and RCP8.5 projections suggest that factors responsible for past and future changes in the Mediterranean region are different. The wet LGM conditions were determined mainly by low evaporation, with some increase of precipitation in the western areas, while dry rcp8.5 conditions will be driven by a reduction of precipitation over the whole region. These changes were caused by atmospheric dynamics (changes of mean atmospheric circulation ) in LGM and it will be caused by the atmospheric thermodynamics (reduction of mean moisture content ) in the future rcp8.5. In both cases, the Mediterranean region appears to be more sensitive to climate change than the rest of areas within the same latitudinal range, particularly considering the hydrological cycle, whose characteristics in winter exhibit large changes between these two different climates. These conclusions emerge from the substantial consensus among six PMIP3 and CMIP5 models, simulating LGM, pre-Industrial and rcp8.5 climate conditions.

    Notes: Conclusions In summary, the LOS of B. pertussis lacks the long-chain polysaccharide O antigen, and is resolved electrophoretically into two distinct bands designated LOS A and LOS B. Mice administered with anti-LOS A mAbs can be protected from fatal B. pertussis infection. These observations may indicate potential for oligosaccharides as a component of new acellular pertussis vaccines. Studies performed thus far also suggest that idiotypic profiles influence the protective value of antibodies and that several antibody-mediated effector functions may participate in the protection. Further research investigating the factors inducing protective immunity is needed for a better definition of new vaccine formulations.

    Notes: Abstract  The present study uses increased atmospheric pressure as an ethanol antagonist to test the hypothesis that allosteric coupling pathways in the GABAA receptor complex represent initial sites of action for ethanol. This was accomplished using behavioral and in vitro measures to determine the effects of pressure on ethanol and other GABAergic drugs in C57BL/6 and LS mice. Behaviorally, exposure to 12 times normal atmospheric pressure (ATA) of a helium-oxygen gas mixture (heliox) antagonized loss of righting reflex (LORR) induced by the allosteric modulators ethanol and pentobarbital, but did not antagonize LORR induced by the direct GABA agonist 4,5,6,7-tetrahydroisoxazolo-pyridin-3-ol (THIP). Similarly, exposure to 12 ATA heliox antagonized the anticonvulsant effects verses isoniazid of ethanol, diazepam and pentobarbital. Biochemically, exposure to 12 ATA heliox antagonized potentiation of GABA-activated 36Cl– uptake by ethanol, flunitrazepam and pentobarbital in LS mouse brain preparations, but did not alter GABA-activated 36Cl– uptake per se. In contrast to its antagonist effect versus other allosteric modulators, pressure did not antagonize these behavioral or in vitro effects induced by the neuroactive steroid, 3α-hydroxy-5β-pregnan-20-one (3α,5β-P). These findings add to evidence that pressure directly and selectively antagonizes drug effects mediated through allosteric coupling pathways. The results fit predictions, and thus support the hypothesis that allosteric coupling pathways in GABAA receptors represent initial sites of action for ethanol. Collectively, the results suggest that there may be common physicochemical and underlying structural characteristics that define ethanol sensitive regions of receptor proteins and/or their associated membranes that can be identified by pressure within (e.g., GABAA) and possibly across (e.g., GABAA, NMDA, 5HT3) receptors.

    Notes: Abstract This paper discusses research designed to investigate farmworker interpretation of a serious incident of pesticide poisoning, specifically, whether this incident was considered to precipitate the Mexican folk illness “susto.” The study revealed that a total of 23% of the population interviewed felt that they definitelyor possibly had developed susto as a result of the pesticide exposure. These patients complained of more residual symptoms from the pesticide exposure than did those who had been affected by the poisoning but who did not feel they suffered from susto. The findings have implications for medical providers working with these populations. A self diagnosis of “susto” may indicate that a patient is indeed sicker than other indicators suggest, and point to a need for increased follow-up.

    Description: Background: Drug metabolism via the cytochrome P450 (CYP450) system has emerged as an important determinant in the occurrence of several drug interactions (adverse drug reactions, reduced pharmacological effect, drug toxicities). In particular, CYP3A4 and CYP3A5 (interacting with more than 60% of licensed drugs) exhibit the most individual variations of gene expression, mostly caused by single nucleotide polymorphisms (SNPs) within the regulatory region of the CYP3A4 and CYP3A5 genes which might affect the level of enzyme production.In this study, we sought to improve the performance of sensitive screening for CYP3A polymorphism detection in twenty HIV-1 infected patients undergoing lopinavir/ritonavir (LPV/r) monotherapy. Methods: The study was performed by an effective, easy and inexpensive home-made Polymerase Chain Reaction Direct Sequencing approach for analyzing CYP3A4 and CYP3A5 genes which can detect both reported and unreported genetic variants potentially associated with altered or decreased functions of CYP3A4 and CYP3A5 proteins. Proportions and tests of association were used. Results: Among the genetic variants considered, CYP3A4*1B (expression of altered function) was only found in 3 patients (15%) and CYP3A5*3 (expression of splicing defect) in 3 other patients (15%). CYP3A5*3 did not appear to be associated with decreased efficacy of LPV/r in any patient, since none of the patients carrying this variant showed virological rebound during LPV/r treatment or low levels of TDM. In contrast, low-level virological rebound was observed in one patient and a low TDM level was found in another; both were carrying CYP3A4*1B. Conclusions: Our method exhibited an overall efficiency of 100% (DNA amplification and sequencing in our group of patients). This may contribute to producing innovative results for better understanding the inter-genotypic variability in gene coding for CYP3A, and investigating SNPs as biological markers of individual response to drugs requiring metabolism via the cytochrome P450 system.
