Hand.-Mazz. and S. minor Scop. subsp. minor, of which S. minor is a new record for India. A key to the Indian taxa is provided, nomenclature and typification discussed; cytological, palynological, ecological data and notes on economic uses, wherever available, are furnished, range of distribution indicated and specimens examined, cited.

    Beschreibung / Inhaltsverzeichnis: Abstract Окисление сульфаниловой кислоты до соответствующего азокси-производного с помощью пероксомонофосфорной кислоты (PMPA) было исследовано в водном растворе. Наблюдаемая зависимость pH—скорость с профилем колокола интерпретировалась на основе протонирования аминогруппы, ионизации PMPA в различные частицы и подбирая соответствующий закон скорости. Механизм окисления заключает в себе нуклеофильную атаку азота на электрофильный пероксокислород.
    Notizen: Abstract Oxidation of sulfanilic acid to the corresponding azoxy derivative by peroxomonophosphoric acid (PMPA) has been studied in aqueous medium. The observed bell-shaped pH-rate profile has been rationalized on the basis of protonation of the amino group and ionization into different PMPA species and a suitable rate law has been proposed. The mechanism of oxidation involves the nucleophilic attack of nitrogen on the electrophilic peroxo oxygen.

    Notizen: The kinetics of dimethyl sulfoxide (DMSO) oxidation by peroxomonophosphoric acid (PMPA) in aqueous medium at 308 K and I = 0.4 mol/dm3 follow the rate expressions In the pH range from 0 to 2, where k1 and k2 are 5.092 × 10-1 dm3/mol sec and ≃ 0, respectively; in the pH range from 4 to 7, where k2 = 8.127 × 10-3 and k3 = 2.90 × 10-3 dm3/mol sec; and in the pH range from 10 to 13.6, where k4 ≃ 0, and k5 = 3.08 × 10-2 dm3/mol sec.The reaction is interpreted in terms of mechanisms involving an electrophilic and a nucleophilic attack of the peroxomonophosphoric acid species, respectively, in acid and alkaline regions, on the sulfur atom of the sulfoxide molecule giving rise to SN2-type transition states followed by oxygen-oxygen bond fission to form the products.

    Notizen: Pyridiniumchlorochromate (PCC) oxidizes aniline and substituted anilines except nitro anilines smoothly in chlorobenzene-nitrobenzene mixtures in the presence of dichloroacetic acid. The reaction has unit dependence on each of the aniline, PCC, and dichloroacetic acid concentrations. Electron-releasing substituents accelerate the reaction, whereas electronwithdrawing groups retard the reaction, and the rate data obey Hammett's relationship. The reaction constant ρ is -3.75. Azobenzene and p-benzoquinone have been obtained as products. The observed experimental data have been rationalized in terms of the formation of an intermediate complex involving PCC-amine undergoing a rapid decomposition to products.

    Beschreibung: Mismatch repair (MMR) is required for proper maintenance of the genome by protecting against mutations. The mismatch repair system has also been implicated as a driver of certain mutations, including disease-associated trinucleotide repeat instability. We recently revealed a requirement of hMutSβ in the repair of short slip-outs containing a single CTG repeat unit (1). The involvement of other MMR proteins in short trinucleotide repeat slip-out repair is unknown. Here we show that hMutLα is required for the highly efficient in vitro repair of single CTG repeat slip-outs, to the same degree as hMutSβ. HEK293T cell extracts, deficient in hMLH1, are unable to process single-repeat slip-outs, but are functional when complemented with hMutLα. The MMR-deficient hMLH1 mutant, T117M, which has a point mutation proximal to the ATP-binding domain, is defective in slip-out repair, further supporting a requirement for hMLH1 in the processing of short slip-outs and possibly the involvement of hMHL1 ATPase activity. Extracts of hPMS2-deficient HEC-1-A cells, which express hMLH1, hMLH3, and hPMS1, are only functional when complemented with hMutLα, indicating that neither hMutLβ nor hMutLγ is sufficient to repair short slip-outs. The resolution of clustered short slip-outs, which are poorly repaired, was partially dependent upon a functional hMutLα. The joint involvement of hMutSβ and hMutLα suggests that repeat instability may be the result of aberrant outcomes of repair attempts.

    Beschreibung: R-loops, transcriptionally-induced RNA:DNA hybrids, occurring at repeat tracts (CTG) n , (CAG) n , (CGG) n , (CCG) n and (GAA) n , are associated with diseases including myotonic dystrophy, Huntington's disease, fragile X and Friedreich's ataxia. Many of these repeats are bidirectionally transcribed, allowing for single- and double-R-loop configurations, where either or both DNA strands may be RNA-bound. R-loops can trigger repeat instability at (CTG)·(CAG) repeats, but the mechanism of this is unclear. We demonstrate R-loop-mediated instability through processing of R-loops by HeLa and human neuron-like cell extracts. Double-R-loops induced greater instability than single-R-loops. Pre-treatment with RNase H only partially suppressed instability, supporting a model in which R-loops directly generate instability by aberrant processing, or via slipped-DNA formation upon RNA removal and its subsequent aberrant processing. Slipped-DNAs were observed to form following removal of the RNA from R-loops. Since transcriptionally-induced R-loops can occur in the absence of DNA replication, R-loop processing may be a source of repeat instability in the brain. Double-R-loop formation and processing to instability was extended to the expanded C9orf72 (GGGGCC)·(GGCCCC) repeats, known to cause amyotrophic lateral sclerosis and frontotemporal dementia, providing the first suggestion through which these repeats may become unstable. These findings provide a mechanistic basis for R-loop-mediated instability at disease-associated repeats.
