Abstract:
WNK kinases are intracellular protein kinases associated with familial hypertension. This association caused us to ask whether WNKs are directly activated by hydrostatic pressure. We show the kinase domain of WNK3 is activated by hydrostatic pressure both in vivo and in vitro. Hydrostatic pressure induced modest ~30%-50% enhanced autophosphorylation or substrate phosphorylation. Biophysical characterization exposed a dimer to monomer equilibrium promoted by pressure. This reveals that an intracellular protein kinase is activated by hydrostatic pressure.
