    Notes: Abstract. The present work establishes a correlation between cell length and patterns of mitotic microtubular assemblies in Allium cepa L. root meristems. Binucleate cells were formed by a short caffeine treatment which aborted the formation of the phragmoplast during telophase. The largest binucleate cells (about 50 μm in length) behaved as two contiguous mononucleate cells in their next mitosis: they developed two preprophase bands (PPBs), one around each nucleus, where two spindles and two phragmoplasts were subsequently formed. On the other hand, the shortest binucleate cells (about 36 μm in length) formed a single PPB at the site of the aborted phragmoplast and, in the medium-sized cells (about 44 μm) in which the single PPB formed around the nucleus possessing the largest cytoplasmic environment, the two mitotic spindles and the new phragmoplasts moved to, or were assembled in the position of the phragmoplast that had been aborted one cycle earlier. Some rules derive from these observations. First of all, the aborted phragmoplast left a signal for microtubule positioning which was still operative one cycle later, in two-thirds of the bimitoses. Also, that formation of the PPB is dispensable. Moreover, its development does not always predict the future division plane, because of the presence of competing old signals which are stronger than those shed by the PPB in the same mitosis, but which fade away with distance. Finally, the positional signals were reinforced when the ratio of monomeric to fibrillar actin was increased by cytochalasin D during their shedding. When this drug was given simultaneously with caffeine, the frequency of bimitoses which, one cycle later, developed spindles and phragmoplasts in the positions of the old phragmoplast increased. On the other hand, those frequencies dropped in relation to control when the cytochalasin D treatment took place during bimitosis, indicating that at this time the treatment reinforced the signals produced in bimitosis itself.

    Notes: Abstract This paper analyses the effect of acid hydrolysis on the differential Giemsa staining of 5-bromo-2′deoxyuridine (BrdU) substituted chromatids in human and plant chromosomes, after treatment with a fluorochrome and light. Human lymphocytes and Allium cepa L. root tips were grown in BrdU for two or three cell cycles. Lymphocyte spreadings and meristem squashes were treated with fluorochrome Hoechst 33258, exposed to sunlight, hydrolysed with 5N HCl and stained with Giemsa. This acid hydrolysis improves the differential staining of BrdU substituted and non-substituted chromatin. It also allows the differentiation of sister chromatids with the DNA specific dye azure-A.

    Notes: Abstract The structural organization of the dense chromatin in G1, S and G2 of onion meristem cells has been evaluated. A naturally synchronous subpopulation of caffeineinduced binucleate cells was employed. — Fibre size is positively correlated with cell stage in interphase. Fibre size distribution is unimodal in G1 nuclei with the peak at 12.5 nm in diameter, while in G2 the distribution is bimodal due to a new population of thicker fibres (22.5 nm in diameter). Separation between fibre centres takes place between mid G1 and mid S. — Using stereological principles, the length of chromatin fibres integrated in the chromatin patches could be estimated. This length remains constant from mid G1 to mid S, when a 1.5 fold increase in total DNA takes place. On the other hand, 40 mm/hour of chromatin fibre is integrated in the chromatin patches between mid S and mid G2. Comparable data of the relative proportion of the different nuclear components have been obtained in each interphase period. — The reported changes provide evidence of a cyclic pattern of chromatin condensation, which may be the structural support for a model of chromatin function in these cycling cells.

    Notes: Abstract The method described in a previous work to separate trace amounts of selenium in organic samples without using a carrier, based on the adsorption on active carbon filters of the complex formed with ammonium pyrrolidindithiocarbamate (APDC) at pH 1.5–2, has been applied to urine samples from 15 females patients suffering from cervical uterine cancer. With this type of sample the method reaches a maximum sensitivity (few ppb) with a good statistical variation (±12%). Since the highest concentration of selenium in human tissues is found in the kidney, and the elimination of this element is mainly by the urine, the method seems to be a powerful tool in the research about the human metabolism of selenium. This paper shows a possible relation of selenium concentration in human urine and the evolution time of cervical uterine cancer, in spite of limits imposed by the statistical error plus the inhomogeneity of the sample.

    Notes: Abstract Gamma exposure rate and radon levels were measured in 75 single-family dwellings in Mexico City in order to correlate them with local environment. Radon monitoring was performed both indoors and outdoors using a continuous working level monitor for short-lived radon decay products; the gamma exposure rate was measured using CaSO4: Dy+PTFE. The results obtained show a log-normal distribution. The mean indoor radon concentration is lower than 45 Bq/m3 and the mean indoor gamma exposure rate was 11.29 μR/h.

    Notes: Abstract A method is described to separate trace amounts of selenium in organic samples without using a carrier, based on the adsorption on active carbon filters of the complex formed with APDC at pH 1.5. The separation is made prior to the neutron irradiation and the correction for selenium in the reagents performed by the irradiation of a blank. The method has been successfully used in fodder samples as well as some other organic reference materials, using a fast pneumatic transfersystem, which takes the samples from the irradiation position to the counting position in 3s, with the only limitation of the thermal neutron flux available, 4.7·1011 cm−2·s−1. Detection limits of 0.1 μg are obtained under these experimental conditions.

    Notes: Abstract Neutron activation analysis of Se in urine reaches an optimum sensitivity (few ppb) and precision (±12%) when the traces are complexed without using a carrier by ammonium pryrrolidindithiocarbamate (APDC) at pH 1.5–2 and adsorbed on activated carbon filters. In this way the selenium traces analysis have been carried out through77mSe in 45 urine samples on a pre-separation basis by cyclic activation ofthe carbon filters. The selenium concentration in our blanks is virtually zero, because APDC proved to be selnium free and selenium mass in 50 mg of activated carbon used as a filter is 20 times below our qualitative detection limit and 144 times below our quantitative detection limit. The samples were first of day urine from healthy and ill women suffering cervical uterine cancer, at different evolution stages: incipient, intemediate and advanced, with no treatment, and surgery, radiotherapy, chemotherapy, or a combined treatment. The results show a consistent tred to increase the selenium trace concentration during the intemediate stage, whereas it is the same than nomal for incipient cases, and it decreases to the lowest concentrations for advanced cases.
