Notes: An-11-day old girl was seen with brownish nodular lesions scattered over (he body with emphasis on the face and seal). Several lesions had started lo involute. Tissue was studied by histopathology, immunohistopathology, routine electron microscopy, and immuno-electron microscopy using cryosubstitution and embedding in K4M. Immunohistopathology revealed that the cells of the dermal infiltrate were Langerhans cells. They expressed Leu 6 and HLA-DR. On routine electron microscopy no Birbeck granules were (bund in the dermal cells. Birbeck granules in epidermal Langerhans cells were deformed and often situated next to laminated dense bodies. The latter expressed Leu 6 and HLA-DR. on immuno-electron microscopy. It was concluded that congenital self-healing reticulohistiocytosis is a benign Langerhans cell disease in which Birbeck granules are transformed to laminated dense bodies and possibly degraded by lysosomal enzymes.

Notes: Normal skin was cryoprotected by submerging it in a mixture of 30% dimethylformamide (DMF) in PBS or RPMI. Subsequently it was frozen in liquid propane gas. Cryosubstitution was carried out at −90°C by using methanol to which uranyl acetate or osmium tetroxide were added. The tissue was embedded in either Lowicryl K4M at −40°C or in Epon at +60°C. The- tissue was evaluated by its overall preservation of ultras-tructural details and by its labeling intensity alter incubation with either anti-clesmoglein or anti-type VII collagen monoclonal antibodies. The mixture of DMF and PBS caused an electron-dense precipitate within the cell. The overall morphology was better in Epon-embedded material than in K4M-embedclecl material. However, the labeling was best in K4M material. Regardless of whether the tissue was embedded in Epon or K4M, the addition of osmium tetroxide markedly reduced the degree of labeling.

Notes: Summary The ultrastructural localization of carbohydrate residues in human melanocytes of normal epidermis and of one compound naevus was studied. The following lectins were used in a post-embedding technique: 1. peanut agglutinin (PNA), which reacts specifically with N-acetyl-galactosamine; 2. Concanavalia ensiformis (Con A) indicating α-d-glucose and α-d-mannose binding sites; 3. Ulex europaeus agglutinin (UEA I) specific for α-l-fucose; 4. Wheat germ agglutinin (WGA), reacting specifically with N-acetyl-glucosamine and neuraminic acid (sialic acid); and 5. Limax flavus agglutinin (LFA), also specific for sialic acid (Neu5Ac-α-2,3-Gal and Neu5Ac-α-2,6-Gal). When incubated with WGA, Con A and LFA strong labelling was seen within the cytoplasm and in the plasma membrane of melanocytes, whereas incubations with PNA and UEA I revealed an occasional gold particle only. The determination of the distribution of carbohydrate residues in normal melanocytes is a prerequisite for future studies of abnormal melanocytes.

Description / Table of Contents: Investigations on Metal Catalysts. I. Structural and Catalytical Investigations on Palladium-Silver PowdersSeveral conditions for preparation of palladium-silver powders are described. By choice of suitable preparation metal crystallites were obtained, in which exists a gradient of concentration with respect to the metals. This gradient determines the behaviour of sorption and catalytical activity, respectively.Characterization of catalysts with regard to their structure takes place by means of X-ray measurements and investigations of sorption (adsorption of N2, chemisorption of CO, sorption of H2), respectively. The catalytical activity of alloys has been measured by means of hydrogenation of benzene.

Description / Table of Contents: Preparation and Properties of Formed Aluminium Oxide. II. Influence of the Mechanical Comminution of Precipitated Boehmite Hydrogel on the Pore Structure and Abrasion Resistance of the Aluminium OxideA report is given on the physical-chemical characteristics of formed aluminium oxide on the basis of boehmite which had been continuously precipitated with nitric acid from sodium aluminate solution, subsequently filtrated, dried at 110°C and ground prior to moulding. In double dependence on the grinding intensity (type of mill, grinding time) and on the precipitation conditions (pH value, temperature) the changes of the morphology and the texture of the boehmite hydrogel are investigated. According to the particles constituting the precipitated boehmite hydrogel is made in two stages. In dependence on the grinding intensity a maximum correlation is observed for the abrasion resistance.
