Notes: Abstract  The topographic relations of complex structures and the morphogenesis of organ systems can only be fully understood in their three-dimensional context. Three-dimensional (3-D) reconstruction of physically sectioned specimens has become an indispensable tool in modern anatomical and embryological research. Teaching also makes increasingly use of 3-D representations, in particular in the case of embryonic systems that undergo complicated transformations of form and shape. At present no cheap and simple technique is available that generates accurate 3-D models of sectioned objects. In this study we describe a novel technique that rapidly provides faithful 3-D models of sectioned specimens. The images are captured directly from the cutting surface of the embedding block after each sectioning and ”on block” staining step. Automatic image processing generates a stack of binary images of the specimen contour. Binary images of internal structures are obtained both by automatic segmentation and manual tracing. Since these image series are inherently aligned, they can be reconstructed three-dimensionally without time-consuming alignment procedures. The quality and the flexibility of the method are demonstrated by reconstructing three kinds of specimens of different histological composition and staining contrast: a 4 mm mouse embryo together with several of its inner organs, a cavernous sinus region of a human infant, and a segment of a human carotid artery. Very short processing times and the faithful representation of complex structural arrangements recommend this technique for routine use in morphological research and for creating embryologic teaching models or 3-D embryonic staging series.

Description: Two loess sections from the Upper Palaeolithic site of Kammern-Grubgraben (Lower Austria) were analysed to test geochemical proxies, as well as radiocarbon data of different components for their reliability and consistency in an archaeological context. Only a reliable basal age (28.9  27.8 ka cal BP) was obtained from charcoal fragments derived from a tundra gley underlying the archaeological horizons and assigned to Greenland Interstadials 3 or 4. Grain size, organic and inorganic geochemistry, and stable isotopes of the fine organic fraction (13Corg) and of rhizoconcretions (13C, 18O) were analysed to provide information on palaeoenvironmental conditions. Low-resolution geochemical and sedimentological analyses document a humidity-related variability, while 13Corg values indicate a predominant C3 vegetation. High-resolution elemental variations derived from X-ray fluorescence scanning exhibit increasing Ca and decreasing Fe and Ti values indicating drier conditions towards the top. Secondary pedogenic carbonate concretions provide post-sedimentary (Holocene) ages and are not suitable for assessing climate and environmental changes for the Palaeolithic.

Description: The paper presents results of multiproxy-investigations of a 3 m long sediment section from the Glowe Palaeolake, covering the period Pre-Bølling to the middle of the Preboreal. The Lateglacial and early Holocene landscape development comprising climate fluctuations, lake evolution, lake-level variations and vegetation history is reconstructed using pollen, diatom, macrofossil, molluscs as well as sedimentological and geochemical data based on 14C-dating. The palaeolake appeared due to the decay of the permafrost during the Bølling and developed in the Allerød into a 3–4 m deep, species-poor and macrophyte-rich stillwater. The submerse vegetation and fauna decreased during the Younger Dryas, but returned fast and with higher density in the Preboreal. Phases of cooler climate can be parallelized with the Gerzensee oscillation, the Younger Dryas and the Rammelbeek oscillation, which each are palynologically bipartite. In contrast, indications for the Older Dryas were only scarce. The cooler phases were characterized by intensified allochthonous clastic input into the lake. During the Younger Dryas the input was dominated by solifluction processes, while during the Allerød and the Preboreal predominantly fluvial processes occurred. The most significant changes in the palaeoecology of the lake were caused by the rapid warming at the onset of the Preboreal. During the phases of warmer climate the vegetation development was influenced by the vicinity to the Baltic Ice Lake, which caused – compared to more southerly regions – a delayed spread of Pinus. Also, the long term climate changes determined the alterations in the chemical sediment composition, the diatom flora and the macrophyte vegetation. Short term variations, which caused the closely spaced sediment layering mainly in the older part of the sediment section cannot be explained so far. The course of the outcropping stratigraphic units was used to construct a lake-level curve. It shows a rapid rise in the early Allerød and a subsequent slower rise until the highstand in the Younger Dryas. In the early Preboreal, a fast lake-level fall occurred, the palaeolake silted up and dried out in the middle of the Preboreal.
