    Notes: When peripheral blood lymphocytes (PBL) were stimulated in vitro with the rough form of type 2 Streptococcus pneumoniae R36a, the resulting plaque-forming cells (PFC) did not produce antibodies directed against phosphorylcholine, a major antigenic determinant of the cell wall C-polysaccharide. Instead, R36a stimulated polyclonal PFC in PBL and splenic lymphocytes. We compared the polyclonal responses stimulated by R36a with those induced by two well-characterized polyclonal activators (PA), Staphylococcus aureus Cowan I and pokeweed mitogen (PWM). We found that R36 a was a poor mitogen for PBL, whereas the other two PA were potent mitogens; that the predominant isotype produced in response to all three PA was IgM; that adherent cells strongly inhibited the polyclonal PFC response to both R36a and Staph. aureus but not PWM; and that T cells were necessary for induction of polyclonal antibody-secreting cells by all three stimuli.

    Notes: A new routine has been developed for fitting multiple isomorphous replacement (MIR) maps. The map is first thinned and reduced to a series of `traces'. Each trace consists of a main trace of defined length and a series of branching side traces. Using this trace representation, the entire map can be viewed at one time and the major secondary structural features can be recognized. A number of features have been built into the routine for editing these traces, allowing neighboring traces to be connected and incorrect connections to be broken. After the traces have been edited, a polyserine fit is generated. The traces are used to guide the initial positioning of a short polyserine segment. This segment is then refined using a real-space molecular-dynamics procedure, which uses the gradient of the electron density as a pseudoforce on the polyserine atoms. After the structure of this initial polyserine has been refined, another serine is added at one end, a short period (about 10 s on a personal Silicon Graphics IRIS workstation) of molecular dynamics is applied and the procedure is repeated. The final refined structure depends only on the electron density and is independent of the thinned traces. The routine is illustrated with two examples. One example uses the 3.0 Å map generated directly from a model protein. This allows direct comparison between the polyserine fit and the model that was used to generate the map. The second example uses the experimental MIR map for lipovitellin. This is a very large protein (about 1300 amino acids), which has a poor-quality map that has not yet been refined. The program makes extensive use of Silicon Graphics graphical (GL) routines.

    Notes: Abstract According to hydrodynamical and mode-coupling theories, the angular velocity autocorrelation function decays at long times asν 0(t/10−14 sec)−5/2. For rough spheres under the conditions reported here, the quantityν 0 is predicted to be 262. The molecular dynamics studies presented here yield a long-time tail of the form 230(t/10−14 sec)−2.38. The disagreement between theory and computer results probably arises from statistical error intrinsic to the computations.

    Notes: Summary Surface reflectance factors from bare field soil were measured to determine the relationship between surface soil water content and spectral reflectance. Reflectance in the six reflective Thematic Mapper (TM) bands plus a 1.15 to 1.30 µm bandpass (referred to as MMR 5) was measured using a groundbased radiometer across a soil water gradient provided by a line source sprinkler system. A spectral index of soil brightness (Brightness) derived using the Gram-Schmidt process and utilizing reflectance information was calculated for each band and for combinations of bands. The results of this study show that TM band 7 (2.05–2.30 µm) provided improved estimates of surface soil water content (0–0.5 cm depth) over estimates using reflectance information from all seven bands. Good correlations were also found between band ratio spectral indices of TM 5 (1.55–1.75 µm) / TM 7, MMR 5/TM 7, and MMR 5/TM 5 and surface soil water content. Results indicate that surface reflectance factors within bandpasses that partially overlap water absorption regions, such as TM 7, are most highly correlated with surface soil moisture. Band ratios utilizing a bandpass partially overlapping a water absorption region such as TM 7 and a non-water-absorbing bandpass such as MMR 5 yield close correlations with surface soil water content.
