    Beschreibung: Increasing temperatures and glacier-melting at the Western Antarctic Peninsula (WAP) are causing rapid changes in shallow coastal and shelf systems. Climate change related rising water temperatures, enhanced ice scouring as well as coastal sediment run-off in combination with changing feeding conditions and microbial community composition will affect all elements of the nearshore benthic ecosystem, a major component of which is the Antarctic soft shell clam Laternula elliptica. 454 RNA sequencing was carried out on tissues and hemocytes of L. elliptica, resulting in 42.525 contigs of which 48% were assigned putative functions. Changes in the expression of putative stress response genes were then investigated in hemocytes and siphon tissue of young and old animals subjected to starvation- and injury-experiments in order to investigate their response to sedimentation (food dilution, starvation) and iceberg scouring (injury). Analysis of antioxidant defense (Le-SOD, Le-catalase), wound repair (Le-TIMP, Le-chitinase), stress and immune response genes (Le-HSP70, Le-actin, Le-theromacin) revealed that most transcript were more clearly affected by injury rather than starvation. The up-regulation of these genes was particularly high in the hemocytes of young, fed individuals after acute injury. Only minor changes in expression were detected in young starved and old animals. The stress response of L. elliptica thus depends on the nature of the environmental cue and on age. This has consequences for future population predictions as the environmental changes at the WAP will differentially impact L. elliptica age classes and is bound to alter population structure.

    Notizen: Summary The aim of our echocardiographic study was to characterize cardiac function and anatomy of 14 acromegalics (A: 9 women, 5 men; mean age: 42.4 yrs) more closely. The duration of acromegaly in 4 of these patients was between 3 and 12 years; the disease was diagnosed for the first time in the other patients. Double M-mode echocardiography was performed in all patients and the results compared with data obtained from a control group of 24 healthy volunteers (N: 22 men, 2 women; mean age: 23 yrs). The mean left ventricular diameter at end-diastole was greater in the acromegalics than in the controls (A: 55±6 mm, N: 50±4 mm;p〈0.005, $$\bar x \pm SD$$ ). After correction for age and body surface area, it, however, was outside the 95% confidence interval in 5 patients. Left ventricular hypertrophy was present in 3 patients, one of whom had coexistent arterial hypertension. A total of 3 patients were hypertensive. Significantly higher values for the maximal velocity of systolic wall thickening (A: 6.1±0.6 cm/s, N: 4.2±0.6 cm/s,p〈0.001) and diameter change (A: 12.4±2.0 cm/s, N: 10.6±1.0 cm/s,p〈0.005) indicate increased contractility with concurrently increased relaxation; fractional shortening did not differ significantly (A: 38±5%, N: 37±5%, ns). The isovolumetric relaxation period at diastole was slightly longer in the acromegalics (A: 70±17 ms, N:61±13 ms,p〈0.05). Whereas the values for maximal diastolic velocity of wall thinning (A: 14.1±4.9 cm/s, N: 11.0±3.4 cm/s,p〈0.02) and diameter increase (A: 21.6±4.7 cm/s, N: 17.8±4.1 cm/s,p〈0.02) were significantly higher, those for the rapid filling period did not differ significantly (A: 115±24 ms, N: 102±30 ms, ns). We concluded that systolic and diastolic left ventricular function did not deviate from the norm in our patients, only a relatively small percentage of whom had coexistent arterial hypertension and left ventricular hypertrophy.

    Notizen: Summary In 45 patients with carotid sinus syndrome, the heart rate-independent vasodepressor component under ventricular and av-sequential pacing of the heart was investigated both in supine and in standing patients. Under both forms of pacing, the carotid pressure determination has led to a marked lowering of systolic and diastolic arterial blood pressure which is significantly more pronounced under orthostasis. The blood pressure values reached both in supine and in standing patients are significantly higher under av-sequential pacing than under ventricular pacing. Accordingly, the proportion of symptomatic patients is less under av-sequential pacing in the supine position (17% versus 29%) and in standing (65% versus 83%). The heart rate-independent vasodepressor reaction is maximal on average between 11 s and 16 s after the beginning of the carotid pressure test and persists for 3 s to 7 s. The blood pressure reaches initial values again after 14 s to 20 s. To summarize, these data document that a clinically relevant heart rate-independent vasodepressor reaction is to be reckoned with in the majority of patients with carotid sinus syndrome. This shows a different time course than the vagally determined effect of the carotid sinus reflex on heart rate. Orthostasis intensifies the vasodepressor reaction so that the hemodynamically more favorable avsequential pacing which is actually more favorable in hemodynamic terms cannot guarantee freedom from symptoms in the majority of patients in the upright standing position.

    Beschreibung / Inhaltsverzeichnis: By mechanical, calorimetric and dielectric methods the relaxation processes in the glass transition region of poly-(ethylene terephthalate) were investigated. Based on the results obtained the WLF parameters of the amorphous initial material and the apparent activation energy at different temperatures were determined. The effect of different degrees of crystallization, established by termination of the isothermal crystallization at 105°C, on the glass transition temperature are discussed.
    Notizen: Mit mechanischen, kalorimetrischen und dielektrischen Meßmethoden werden Relaxationsprozesse beim Glasübergang des Polyethylenterephthalats untersucht. Anhand der Meßergebnisse werden die WLF-Parameter für das amorphe Ausgangsmaterial ermittelt und scheinbare Aktivierungsenergien für verschiedene Temperaturen angegeben. Der Einfluß unterschiedlicher Kristallinitätsgrade, die durch Abbruch isothermer Kristallisation bei 105°C erzielt wurden, auf die Glastemperatur wird diskutiert.

    Beschreibung / Inhaltsverzeichnis: Undrawn poly(ethylene terephthalate) was annealed at different constant temperatures in the range 368 to 518 K for different periods of time (4 s to 90 h). Both the thermic (DSC) and dynamic (torsion pendulum) glass transition of the samples were investigated. The results can be discussed by assuming superposition of glass transitions in two different amorphous regions, one occurring inside the other outside the spherulites. The properties of the amorphous regions within the spherulites were studied with respect to the crystallization temperature.

An den Proben wurden der thermische (DSC) und dynamische (Torsionspendel) Glasübergang untersucht. Die Ergebnisse lassen sich mit der Überlagerung von Glasübergängen in zwei verschiedenen amorphen Bereichen, die sich zum einen innerhalb, zum anderen außerhalb der Sphärolithe befinden, interpretieren. Die Eigenschaften der amorphen Bereiche innerhalb der Sphärolithe wurden in Abhängigkeit von der Kristallisationstemperatur diskutiert.

    Beschreibung: Marine organisms have to cope with increasing CO2 partial pressures and decreasing pH in the oceans. We elucidated the impacts of an 8-week acclimation period to four seawater pCO2 treatments (39, 113, 243 and 405 Pa/385, 1,120, 2,400 and 4,000 µatm) on mantle gene expression patterns in the blue mussel Mytilus edulis from the Baltic Sea. Based on the M. edulis mantle tissue transcriptome, the expression of several genes involved in metabolism, calcification and stress responses was assessed in the outer (marginal and pallial zone) and the inner mantle tissues (central zone) using quantitative real-time PCR. The expression of genes involved in energy and protein metabolism (F-ATPase, hexokinase and elongation factor alpha) was strongly affected by acclimation to moderately elevated CO2 partial pressures. Expression of a chitinase, potentially important for the calcification process, was strongly depressed (maximum ninefold), correlating with a linear decrease in shell growth observed in the experimental animals. Interestingly, shell matrix protein candidate genes were less affected by CO2 in both tissues. A compensatory process toward enhanced shell protection is indicated by a massive increase in the expression of tyrosinase, a gene involved in periostracum formation (maximum 220-fold). Using correlation matrices and a force-directed layout network graph, we were able to uncover possible underlying regulatory networks and the connections between different pathways, thereby providing a molecular basis of observed changes in animal physiology in response to ocean acidification.

    Beschreibung: Motivation: Next generation sequencing (NGS) technologies allow a rapid and cost-effective compilation of large RNA sequence datasets in model and non-model organisms. However, the storage and analysis of transcriptome information from different NGS platforms is still a significant bottleneck, leading to a delay in data dissemination and subsequent biological understanding. Especially database interfaces with transcriptome analysis modules going beyond mere read counts are missing. Here, we present the Transcriptome Analysis and Comparison Explorer (T-ACE), a tool designed for the organization and analysis of large sequence datasets, and especially suited for transcriptome projects of non-model organisms with little or no a priori sequence information. T-ACE offers a TCL-based interface, which accesses a PostgreSQL database via a php-script. Within T-ACE, information belonging to single sequences or contigs, such as annotation or read coverage, is linked to the respective sequence and immediately accessible. Sequences and assigned information can be searched via keyword- or BLAST-search. Additionally, T-ACE provides within and between transcriptome analysis modules on the level of expression, GO terms, KEGG pathways and protein domains. Results are visualized and can be easily exported for external analysis. We developed T-ACE for laboratory environments, which have only a limited amount of bioinformatics support, and for collaborative projects in which different partners work on the same dataset from different locations or platforms (Windows/Linux/MacOS). For laboratories with some experience in bioinformatics and programming, the low complexity of the database structure and open-source code provides a framework that can be customized according to the different needs of the user and transcriptome project.
