    Notizen: Endosymbiotic methanogenic bacteria of three species of anaerobic ciliates (Plagiopyla frontata, Metopus conforms, and M. palaeformis) were inactivated with the specific methanogen inhibitor 2-bromoethanesulfonic acid. the absence of endosymbiont methanogens reduced growth rate and growth yield by about 30% in P. frontata and M. contortus, while no significant change in fitness was observed in M. palaeformis. In Plagiopyla the growth rate constant is not affected by an artificially increased pH2 neither in normal nor in methanogen-free ciliates. the energetic advantage conferred by endosymbiont methanogens in Plagiopyla and in Metopus contortus probably is due to excretion of organic material from the bacteria at the expense of bacterial reproduction. It is unlikely that the maintenance of a low pH2 within the cells due to H2-consumption by the bacteria is important to the ciliates.

    Notizen: . Behavioral responses to light at different oxygen tensions were studied in the ciliate Loxodes striatus. In the absence of O2 it does not react to light. In the presence of O2 it reacts to light as if the pO2 had been further increased, with the induction of positive geotaxis, a transient phobic response, and finally with a permanent kinetic response (increased swimming velocity and a decreased rate of tumbling). Cells treated with cyanide behave as cells in an anoxic environment and do not react to light. It is concluded that the light response is due to the photochemical production of oxygen radicals and that the sensory receptors for O2 and for light are identical. The three types of behavioral response (geotaxis, transient, and kinetic responses) are discussed in terms of their adaptive significance for the orientation of Loxodes in the natural environment.

    Notizen: . The Müller vesicle is a characteristic organelle of loxodid ciliates. Its structure and development have been investigated using light microscopy and TEM. The organelle consists of a membrane-covered mineral body (the statolith), a vacuole, and various structures derived from the overlying kinety. There is strong evidence that the vesicle functions as a gravity sensor: a) its structure and relative dimensions fulfil the minimum requirements of a functional statocyst; b) its structure bears a close resemblance to the statocysts of some higher animals; c) re-orientation of the cell with respect to gravity produces a gravity-induced displacement of the mineral body, and d) geotaxis in Loxodes can be demonstrated experimentally. The transduction of the signal probably takes place at the level of the two kinetosomes of the organelle, one of which is in close contact with the cell membrane, while the other is connected to the statolith by a fairly rigid stalk containing a bundle of microtubules.

    Notizen: The life cycle of a methanogenic bacterium, symbiotic within the marine, free-living anaerobic ciliate Plagiopyla frontata, was studied using light microscopy and transmission electron microscopy (TEM). the bacteria are disc-shaped. During the growth phase of the host, bacteria and hydrogenosomes (organelles which ferment pyruvate into acetate and hydrogen) are arranged in conglomerates resembling stacks of coins in which bacteria and hydrogenosomes alternate; hydrogenosomes always cap the ends of the stacks. During the growth phase, numbers of hydrogenosomes and bacteria remain constant (about 5,000 and 3,500 per cell, respectively). Hydrogenosomes increase in volume shortly after cell division. Methanogens increase in volume slowly during the growth phase of the ciliate and rapidly when the ciliate begins to divide. the hydrogenosomes divide mainly during the initial phases of cell division while the methanogens divide synchronously during the last phase of ciliate division. the timing of reproduction of the symbionts is controlled by the host-cell cycle. the ciliate is known to receive an energetic advantage from its symbionts. the suppression of continuous bacterial reproduction may trigger the secretion of excess bacterial production as soluble organic compounds, for use by the ciliate.

    Notizen: 〈list xml:id="l1" style="custom"〉1 The major components of the microbial food web (dissolved organic carbon, bacteria, protozoa, rotifers and algae) of Priest Pot, a small freshwater pond, were investigated over a period of 5 months. Water samples were collected from the epilimnion every 1–3 days.2 Time series analysis helped identify the trophic relationships within the planktonic community. There were strong predator—prey relationships between both ciliates and large rotifers and the total nanoplankton, between rotifers and small ciliates and between the total microzooplankton community and phytoplankton. Small rotifers and small ciliates probably share the same food resources. The major bacterivores in the system could not be identified with our methods. However, our previous results point to a dominating role of nanoplanktonic (2–20 μm) heterotrophic protists as the main grazers of bacteria.3 Rotifers are the major type of metazoan zooplankton in Priest Pot; crustacean zooplankton are absent from the community. Bacterial production probably reaches rotifers via a variety of pathways: there may be a three-step link from bacteria to bacterivorous nanoplankton, to ciliates and then to rotifers. Furthermore, a strong correlation between the nanoplankton and rotifers suggests a direct link between these components, implying a much shorter pathway. Some of the rotifers in the pond can graze directly on bacteria, and many of the larger planktonic organisms (large ciliates and rotifers) are algivores. The latter two predator—prey relationships suggest an efficient transfer of bacterial and primary production to higher trophic levels.

The results of laboratory experiments on ciliate reproductive and respiratory rates have been combined with field data for ciliate numbers, to produce estimates of production and respiratory energy losses by communities of benthic ciliates. Annual production decreased with increasing depth, and it was estimated to be 345, 149 and 40 J cm−2 year−1 at three sites in Airthrey Loch, Scotland. A similar trend with increasing depth was recorded for annual respiratory energy losses, being 16, 11 and 4 J cm−2 year−1 at the three sites. The corresponding annual net production efficiencies (%) were 95.2, 93.1 and 91.5. When net production efficiencies were calculated for each period (the time between successive sampling occasions) as 100 (daily production/daily assimilation), the range extended from 63 to 98. The energetic efficiencies of the ciliate populations are discussed with reference to data in the literature for higher organisms.

    Notizen: . The free-living anaerobic flagellate Hexamita sp. was observed to actively consume O2 with a Km O2 of 13 μM. Oxygen consumption increased lineraly with O2 tension up to a threshold level of 100 μM, above which it was inhibited. Oxygen uptake was supported by a number of substrates but probably not coupled to energy conservation as cytochromes could not be detected spectro-photometrically. In addition, inhibitors specific for respiratory chain components did not significantly affect O2 uptake. Respiration was however, partially inhibited by flavoprotein and iron-sulfur protein inhibitors. NAD(P)H supported O2 consumption was measured in both particulate and soluble fractions; this activity was partially inhibited by quinacrine. A chemosensory response was observed in cells exposed to air, however no response was observed in the presence of superoxide dismutase plus catalase. Catalase and nonspecific peroxidase activity could not be detected, but superoxide dismutase activity was present. Superoxide dismutase was sensitive to NaN3 and H2O2 but not KCN, suggesting a Fe prosthetic group. Flow cytometric analysis revealed that thiol levels in live cells were depleted in the presence of t-butyl H2O2. The observed NADPH-driven glutathione reductase activity is believed to recycle oxidized thiols in order to re-establish reduced thiol levels in the cell. The corresponding thiol cycling enzyme glutathione peroxidase could not be detected. The ability to withstand high O2 tensions (100 μM) would enable Hexamita to spend short periods in a wider range of habitats. Prologed exposure to O2 tensions higher than 100 μM leads to irreversible damage and cell death.

    Notizen: . Psilotricha acuminata was described by Stein in 1859 as the type species of the ciliate genus PsilotrichaStein, 1859. The ciliate has rarely been found since, and its infraciliature has never been described with the aid of silver-impregnation techniques. We have found P. acuminataStein, 1859 in soil samples from upland grassland in Scotland (U.K.). Living and healthy organisms of P. acuminata are oblong in outline, and dorso-ventrally compressed. They closely resemble ciliates of the genus Euplotes. The main morphological features used for identification of P. acuminata are the very long and sparse cirri, and the two macronuclei. When the ciliate crawls, the cirri appear stiff and directed backwards. Specimens observed from the ventral side have a protruding anterior end. a rounded or acuminate posterior pole, and a “beak-like” projection to the left side of the posterior end. The ciliate shares characters with the Euplotidae (body shape and reduced ciliature) and with the Oxytrichidae (marginal rows, macronuclei, reduced number of transverse cirri). Because the arrangement of the silver-impregnated infraciliature was unknown, and as the only description of the ciliate was that of Stein (1859a, b), the genus Psilotricha became confused with other hypotrich genera, especially UrospinulaCorliss, 1960. Here we provide a full redescription of P. acuminata based on living and silver-impregnated specimens, and a revision of the genera PsilotrichaStein, 1859 and UrospinulaCorliss, 1960. We resurrect the genus Urospinula, and give an emended diagnosis for both genera. The species now included within the genus Psilotricha are P. acuminataStein, 1859 (type species); Psilotricha viridis (Penard, 1922) Kahl, 1932; and Psilotricha geleii (Gelei, 1954) Stiller, 1974. Psilotricha viridis sensu Kahl, 1932 and Psilotricha dragescoiGrolière, 1975 are considered incertae sedis.
