    Notes: Abstract. The extracellular products (ECP) of Aeromonas salmonicida, prepared by the cellophane overlay method, are lethal to rainbow trout when administered parenterally. Sublethal doses when injected i.p. or i.m. are shown to reproduce all the lesions that have been described in the literature as being associated with furunculosis. In addition, meningitis may be an important feature of furunculosi s and is reproduced by injection of ECP. A serum factor, probably an α-globulin, present in normal serum of rainbow trout, is capable of neutralizing the toxic effects of ECP. The pathology is discussed with reference to the proteolytic and leucocidal properties of the ECP and its effects upon the eosinophilic granular cells which are caused to disperse and degranulate with the possible release of histamine.

    Notes: Abstract. The process of the localization of bovine serum albumin (BSA) within the lymphoid tissues of the plaice is described. The spleen is seen to be of particular importance. Antigen is held, probably as an immune complex, in an extracellular position associated with the metalophil reticular fibres present mainly in the ellipsoid walls and scattered throughout the peri-ellipsoid areas and the melano-macrophage centres. In the kidney tissue, antigen was present mainly intracellularly within the reticulo-endothelial system (RES) and free macrophages. The extracellular antigen within the spleen persisted the longest (at least 37 days). Localization of BSA differed markedly from colloidal carbon in that the metalophil fibres of the ellipsoids and to a small extent those of melano-macrophage centres, are the site of BSA localization and not, as with carbon, macrophages which aggregate within melano-macrophage centres. The thymus and RES of the atrium of the heart played no part in uptake of BSA. The significance of antigen-trapping in vertebrates is discussed and the possible analogous function of the ellipsoid system in fish and germinal centres of higher vertebrates is considered.

    Notes: Forty strains of Flavobacterium psychrophilum were tested for the production of siderophores using the universal Chrome Azurol S (CAS) assay. The majority of the strains (85%) were CAS positive (CAS+) and some (15%) were CAS negative (CAS−). The cryptic plasmid pCP1 was carried by all positive strains and was lacking from negative strains. While a weak catechol reaction was detectable in CAS+ culture supernatants, the CAS reaction was, to some extent, heat sensitive, questioning whether the positive reaction was caused only by siderophores. The ability to grow in vitro under iron-restricted conditions did not correlate with the CAS reactivity, as growth of both CAS+ and CAS− strains was similarly impaired under iron restriction induced by 2,2 dipyridyl. Suppressed growth under these conditions was restored by addition of FeCl3, haemoglobin and transferrin for both CAS+ and CAS− strains.

    Notes: In populations of Atlantic salmon in sea water, infectious pancreatic necrosis virus (IPNV) could be detected by standard virological culture methods in sonicated kidney homogenates and in mucus samples (gill, skin and rectum) from 14 and nine of 25 fish, respectively, but all fish were positive by virus culture from lysates of kidney macrophages and adherent blood leucocytes. In fish which tested negative for IPNV by the standard method of detection, the virus could be detected using adherent blood leucocytes isolated on a Percoll gradient from as little as 10 μL of blood. The blood sample could be stored for at least 3 days in a heparinized tube on ice before preparing the plastic adherent leucocytes. Furthermore, the latter could be prepared without prior fractionation on Percoll simply by incubating whole blood (33 μL) in cell culture medium (66 μL) in 96-well plates overnight and washing away the non-adherent cells before lysing the adherent cells and inoculation of the lysate onto CHSE-214 cells. This highly sensitive method for detecting IPNV-carriers is therefore very suitable for non-destructive sampling of fish in the field.

    Notes: Abstract The role of A-layer (A), protease (P) and haemolysin (H) as virulence factors of Aeromonas salmonicida, the aetiological agent of fish furunculosis, was a investigated using three strains of the bacterium. Strain MT004 lacked the A-Iayer (A−) and produced extracellular caseinase and gelatinase (P+) and haemolysin (T-lysin; H+). Strain MT028 was A−, P− and H−, and strain MT048 was A+, P+ and H−. The pathology and LD50 produced in rainbow trout by cells or extracellular products (ECP) of each strain were determined. The ECP was produced by two different methods where the protease and haemolytic activities differed in relative levels, or when the protease of MT004 ECP was inhibited by the serine protease inhibitor PMSF. The results indicate that the presence of A-layer is not essential, at least for a moderate degree of virulence; that in vitro production of extracellular proteases is not a requisite of virulent strains; that presence of protease and haemolysin in ECP can be correlated with the development of certain lesions and a rapid time to death but cannot be correlated with the lethal toxicity of the ECP. The authors conclude that an as-yet unrecognized component of ECP is responsible for killing fish.

    Notes: Abstract. Ten isolates of Aeromonas hydrophila and two of A. sobria were investigated with regard to elastase and caseinase production, and the pI, heat and EDTA sensitivity of extracellular caseinase enzymes. An extreme degree of heterogeneity amongst the strains was observed. In isoelectricfocusing, the ECP of the various strains possessed between 19 and 31 proteins, and the number of extracellular proteases ranged from one to 12. Similarities between strains using one criterion of comparison (e.g. IEF patterns) did not hold true by other criteria (e.g. heat or EDTA sensitivity of proteases).
