    Notizen: Abstract  Two species of parrot fish, Scarus frenatus and Chlorurus sordidus, are known to exhibit demographic and life-history differences across the continental shelf of the northern Great Barrier Reef (GBR). DNA sequences from the mitochondrial (mt) control region were analysed to determine whether there were genetic differences between the populations from the mid- and outer-shelf reefs. Analysis of molecular variance (AMOVA) indicated high levels of gene exchange for both species at a local scale between reefs on mid- and outer continental shelf positions (20 km apart) and at a broader scale along the length of the GBR province (〉1000 km apart). There was no evidence to suggest that local differences in scarid life-history characteristics on the northern GBR have a genetic basis. Rather it appears more likely that phenotypically plastic responses to prevailing social and environmental conditions explain differences in the life-history characteristics of both taxa. However, analysis of genetic variation and historical demography revealed striking differences between the two species. S. frenatus haplotypes differed from one another at relatively few nucleotide sites (mean = 3.30), and the pairwise mismatch distribution suggested this species has undergone a population expansion within the limit of the resolution of the marker. C. sordidus haplotypes, however, differed from one another at a number of sites (mean = 7.67). Mismatch distribution analysis suggested that the population size of this species has remained at equilibrium over time. These patterns could also reflect differences in the metapopulation sizes or generation times between taxa. Some of the implications for fisheries management are discussed.

    Notizen: Derivatives of three types of bisorthoesters, hexaalkyl orthoisophthalates, hexaalkyl orthoterephthalates, and hexaethyl orthooxalate, were synthesized. The orthoesters of isophthalic and terephthalic acid were prepared from α, α, α, α′, α′, α′ -hexachloro-m-xylene or hexachloro-p-xylene, respectively. It was found more advantageous to prepare initially the hexamethyl orthoesters by reacting the hexachloro compounds with sodium methoxide in methanol either at the boiling point of methanol or under pressure at temperatures up to 200°C. Transesterification with higher alcohols, for example, ethanol or propanol, with the aid of molecular sieves gave the higher bisorthoesters in high yield and high purity. Attempts to synthesize orthoisophthalates and orthoterephthalates of higher alcohols directly from the hexachloroxylenes and alkoxides of higher alcohols gave only low yields of impure products. The Pinner synthesis of the bisorthoesters from the corresponding dinitriles was also ineffective. The ease of purification of hexa-n-propyl orthoisophthalate and orthoterephthalate makes these orthoesters the favored candidates for use in polymer syntheses. Hexaethyl orthooxalate was prepared in a three-step synthesis from triethyl orthoformate by a convenient, but low yield reaction. All bisorthoesters were purified to give compounds which were monomer grade materials free of normal esters.

    Notizen: Aromatic bisorthoesters were found to be good polymer intermediates and could be condensed with aromatic tetramines under mild conditions, in DMSO at 100°C in a relatively short reaction time to give polybenzimidazoles. The hexapropyl orthoesters of terephthalic and isophthalic acid were the preferred aromatic orthoesters because they were relatively easily purified by vacuum distillation to polymer grade intermediates, since they are liquids. Higher orthoesters distill even under good vacuum near or above the decomposition temperature of the orthoester group. Hexaethyl orthooxalate was also used and is a very useful and stable derivative of oxalic acid, which can be used for condensation reactions. These three orthoesters were used for condensations with 3,3′,4,4′-tetraaminobiphenyl, 1,2,3,4-tetraaminobenzene, 3,3′,4,4′-tetraaminobiphenyl ether, and 3,3′,4,4′-tetraaminobenzophenone. All polybenzimidazoles were obtained in high to quantitative yields and with varying molecular weights (ηinh = 0.1-0.8 dl/g), which in some cases were in the fiber forming range.

    Beschreibung: Germline deletion of the p53 gene in mice gives rise to spontaneous thymic (T-cell) lymphomas. In this study, the p53 knockout mouse was employed as a model to study the mutational evolution of tumorigenesis. The clonality of the T-cell repertoire from p53 knockout and wild-type thymic cells was analyzed at various ages employing TCRβ sequencing. These data demonstrate that p53 knockout thymic lymphomas arose in an oligoclonal fashion, with tumors evolving dominant clones over time. Exon sequencing of tumor DNA revealed that all of the independently derived oligoclonal mouse tumors had a deletion in the Pten gene prior to the formation of the TCRβ rearrangement, produced early in development. This was followed in each independent clone of the thymic lymphoma by the amplification or overexpression of cyclin Ds and Cdk6. Alterations in the expression of Ikaros were common and blocked further development of CD-4/CD-8 T cells. While the frequency of point mutations in the genome of these lymphomas was one per megabase, there were a tremendous number of copy number variations producing the tumors’ driver mutations. The initial inherited loss of p53 functions appeared to delineate an order of genetic alterations selected for during the evolution of these thymic lymphomas.

    Beschreibung: Pancreatic ductal adenocarcinoma (PDAC) is a genomically diverse, prevalent, and almost invariably fatal malignancy. Although conventional genetically engineered mouse models of human PDAC have been instrumental in understanding pancreatic cancer development, these models are much too labor-intensive, expensive, and slow to perform the extensive molecular analyses needed to adequately understand this disease. Here we demonstrate that retrograde pancreatic ductal injection of either adenoviral-Cre or lentiviral-Cre vectors allows titratable initiation of pancreatic neoplasias that progress into invasive and metastatic PDAC. To enable in vivo CRISPR/Cas9-mediated gene inactivation in the pancreas, we generated a Cre-regulated Cas9 allele and lentiviral vectors that express Cre and a single-guide RNA. CRISPR-mediated targeting of Lkb1 in combination with oncogenic Kras expression led to selection for inactivating genomic alterations, absence of Lkb1 protein, and rapid tumor growth that phenocopied Cre-mediated genetic deletion of Lkb1 . This method will transform our ability to rapidly interrogate gene function during the development of this recalcitrant cancer.
