    Notes: Abstract. Dorsal fin condition was evaluated in two groups of hatchery-reared steelhead trout, Oncorhynchus mykiss(Walbaum), and compared to wild fish. Hatchery fish were reared either in nursery tanks with baffles or in isolation (1 fish/rearing space). Evaluations were conducted to determine differences in dorsal fin condition between these groups and to monitor the progression and severity of fin damage. Isolated hatchery and wild steelhead trout maintained perfect dorsal fins with characteristic white margins. Hatchery fish reared in nursery tanks with baffles experienced dorsal fin erosion that started when the fish were about 40mm total length (30 days after being on feed). This erosion continued until dorsal fin length averaged only 2·3 mm in fish 161–200 mm total length in contrast to an average dorsal fin length of 20·9mm in wild fish of the same size.

    Description: The percolation of fluids is of utmost relevance for the utilization of underground resources; however, the location and occurrence of fractures are not always known, and important characteristics of faults, such as stress state and permeability, are commonly uncertain. Using a case study at the Brady’s geothermal field in Nevada (USA), we demonstrate how permeable fractures can be identified and assessed by combining fault stress models with measurements of diffuse degassing and emanations at Earth’s surface. Areas of maximum gas emissions and emanations correspond to fault segments with increased slip and dilation tendency, and represent a fingerprint of the geothermal system at depth. Thus, linking gas fluxes with fault stress models serves as a measure of the connectivity between surface and subsurface.

    Description: The sarcoendoplasmic reticulum Ca2+-ATPase (SERCA) is responsible for intracellular Ca2+ homeostasis. SERCA activity in muscle can be regulated by phospholamban (PLB), an affinity modulator, and sarcolipin (SLN), an uncoupler. Although PLB gets dislodged from Ca2+-bound SERCA, SLN continues to bind SERCA throughout its kinetic cycle and promotes uncoupling of Ca2+ transport from ATP hydrolysis. To determine the structural regions of SLN that mediate uncoupling of SERCA, we employed mutagenesis and generated chimeras of PLB and SLN. In this study we demonstrate that deletion of SLN N-terminal residues 2ERSTQ leads to loss of the uncoupling function even though the truncated peptide can target and constitutively bind SERCA. Furthermore, molecular dynamics simulations of SLN and SERCA interaction showed a rearrangement of SERCA residues that is altered when the SLN N terminus is deleted. Interestingly, transfer of the PLB cytosolic domain to the SLN transmembrane (TM) and luminal tail causes the chimeric protein to lose SLN-like function. Further introduction of the PLB TM region into this chimera resulted in conversion to full PLB-like function. We also found that swapping PLB N and C termini with those from SLN caused the resulting chimera to acquire SLN-like function. Swapping the C terminus alone was not sufficient for this conversion. These results suggest that domains can be switched between SLN and PLB without losing the ability to regulate SERCA activity; however, the resulting chimeras acquire functions different from the parent molecules. Importantly, our studies highlight that the N termini of SLN and PLB influence their respective unique functions.

    Description: Respiration from vegetation is a substantial part of the global carbon cycle and the responses of plant respiration to daily and seasonal fluctuations in temperature and light must be incorporated in models of terrestrial respiration to accurately predict these CO 2 fluxes. We investigated how leaf respiration ( R ) responded to changes in leaf temperature ( T leaf ) and irradiance in field-grown saplings of an evergreen tree ( Eucalyptus pauciflora Sieb. ex Spreng). Seasonal shifts in the thermal sensitivity of leaf R in the dark ( R dark ) and in the light ( R light ) were assessed by allowing T leaf to vary over the day in field-grown leaves over a year. The Q 10 of R (i.e., the relative increase in R for a 10 °C increase in T leaf ) was similar for R light and R dark and had a value of ~2.5; there was little seasonal change in the Q 10 of either R light or R dark , indicating that we may be able to use similar functions to model short-term temperature responses of R in the dark and in the light. Overall, rates of R light were lower than those of R dark , and the ratio of R light / R dark tended to increase with rising T leaf , such that light suppression of R was reduced at high T leaf values, in contrast to earlier work with this species. Our results suggest we cannot assume that R light / R dark decreases with increasing T leaf on daily timescales, and highlights the need for a better mechanistic understanding of what regulates light suppression of R in leaves.

    Description: Neisseria is a Gram-negative pathogen with phospholipids composed of straight chain saturated and monounsaturated fatty acids, the ability to incorporate exogenous fatty acids, and lipopolysaccharides that are not essential. The FabI inhibitor, AFN-1252, was deployed as a chemical biology tool to determine whether Neisseria can bypass the inhibition of fatty acid synthesis by incorporating exogenous fatty acids. Neisseria encodes a functional FabI that was potently inhibited by AFN-1252. AFN-1252 caused a dose-dependent inhibition of fatty acid synthesis in growing Neisseria, a delayed inhibition of growth phenotype, and minimal inhibition of DNA, RNA, and protein synthesis, showing that its mode of action is through inhibiting fatty acid synthesis. Isotopic fatty acid labeling experiments showed that Neisseria encodes the ability to incorporate exogenous fatty acids into its phospholipids by an acyl-acyl carrier protein-dependent pathway. However, AFN-1252 remained an effective antibacterial when Neisseria were supplemented with exogenous fatty acids. These results demonstrate that extracellular fatty acids are activated by an acyl-acyl carrier protein synthetase (AasN) and validate type II fatty acid synthesis (FabI) as a therapeutic target against Neisseria.
