    Notes: Summary A computerized databank of IEF protein patterns for use in identifying flatfish species (in total 17 species, including 15 commercial ones) is presented. The databank includes all species regulated by the Belgian Law (22 May 1996) on the use of official names for fishes and seafood products. It was found that interspecimen similarity of the IEF patterns, as processed by digitization, was always larger than interspecies similarity, which allows for unequivocal authentication of unknown samples, as long as the authentic pattern is available in the databank. The databank was used to authenticate 17 commercial fish fillets.

    Notes: The Comamonas testosteroni strain A20, isolated from activated sludge, displays a phase variation between a mucoid (exopolysaccharide producing) and a non-mucoid colony phenotype. It was found that a population of C. testosteroni cells, subjected to sublethal environmental stress (acute heat shock and reactive oxidative intermediates and visible light), became selectively enriched in non-mucoid colony forming cells (NMCF cells). This observation suggests that NMCF cells are more stress-resistant than MCF cells. The experiments did not allow us to establish whether stress acts as a trigger for phase conversion or as a selector for the more stress-resistant NMCF cells. NMCF cells are more hydrophobic than MCF cells as evidenced by a better attachment to gas bubbles and polystyrene. These observations suggest that the two phenotypic variants might have different ecophysiological functions.

    Notes: The yeast gene YKL741 is situated on the left arm of chromosome XI, 12 kb closer to the centromere with respect to the previously localized PAS1 gene. The new yeast gene codes for a homologue of the human ALD protein (ALD: adrenoleukodystrophy). The similarity between the YKL741 protein and the ALD protein is very high in the C-terminal half, which contains an ATP-binding cassette characteristic of the ABC family of transporters. Additionally the YKL741 protein shows some similarity to the ALD protein in the N-terminal half in three putative transmembrane spanning domains. The sequence has been deposited in the EMBL data library under Accession Number X76133 SC YKL.

    Notes: The product of the cloned GTS1 gene is characterized by structural features found in transcription factors. It contains one Zn-finger motif (CXXCX16CXXC) situated in the N-terminal end with a high degree of homology to the newly identified ‘Glo’ family of Zn-finger proteins (Ireland et al., 1994, EMBO J.13, 3812-3821). The C-terminal end of the protein is characterized by poly (Ala-Gln) and poly-Gln stretches. Poly-Gln are part of trans-acting motifs in known transcription factors. Overexpression of the GTS1 gene results in constitutive flocculation. Whole cell electrophoretic mobility and hydrophobicity of GTS1 overexpressing cells was respectively lower and higher relative to control cells. GTS1-induced flocculation is hardly sensitive to mannose in contrast to FLO1-determined flocculation. Overexpression of the GTS1 gene in a flo1 background does not abolish flocculation, suggesting that the FLO1 gene is not linked with the GTS1 gene in a ‘flocculation pathway’. © 1997 John Wiley & Sons, Ltd.

    Description: The reliable production of marine fish larvae is one of the major bottlenecks in aquaculture due to high mortalities mainly caused by infectious diseases. To evaluate if the compound poly-ß-hydroxybutyrate (PHB) might be a suitable immunoprophylactic measure in fish larviculture, its capacity to improve immunity and performance in European sea bass (Dicentrarchus labrax) yolk-sac larvae was explored. Dietary PHB was applied from mouth opening onwards to stimulate the developing larval immune system at the earliest possible point in time. Larval survival, growth, microbiota composition, gene expression profiles and disease resistance were assessed. PHB administration improved larval survival and, furthermore, altered the larva-associated microbiota composition. The bacterial challenge test using pathogenic Vibrio anguillarum revealed that the larval disease resistance was not influenced by PHB. The expression profiles of 26 genes involved e.g. in the immune response showed that PHB affected the expression of the antimicrobial peptides ferritin (fer) and dicentracin (dic), however, the response to PHB was inconsistent and weaker than previously demonstrated for sea bass post-larvae. Hence, the present study highlights the need for more research focusing on the immunostimulation of different early developmental stages for gaining a more comprehensive picture and advancing a sustainable production of high quality fry.

    Description: The goal of this study was to test whether it is possible to produce early hatching cysts of Artemia franciscana from San Francisco Bay (SFB) by truncation selection. The starting material was an SFB cyst sample, harvested in nature. After selection of early hatching nauplii had taken place, these selected individuals were cultured to maturity, and the hatching rate of their offspring (F1) was compared with that of the parental generation and with the non-selected control F1. The possible differences in hatching rate were used to estimate heritability. Two different selection experiments were run, accompanied with a number of additional tests studying the influence of the hatching set-up, the hatching temperature and diapause termination using hydrogen peroxide, on the hatching rate. Also the influence of different culture salinities on hatching rate and on possible success of selection was studied. The results revealed an influence of all those abiotic factors on the hatching rate, but also a marked interference of the hatching percentage with the hatching rate. Choosing the individuals based on their own phenotypic values (hatching within a certain time span of hatching incubation) and using those to produce the next generation, revealed a positive response. The selection in different salinities showed a different response. Depending on the strength of the selection pressure, the samples were advanced in time, starting with the highest selection pressure, and ending with the control and the parental sample. In spite of the strong interference of environmental factors, our results suggest that selection of early hatching cysts is possible.

    Description: 12 samples (6 original samples and 6 diluted samples) were analysed by 14 WEFTA laboratories for their pH values in an inter-laboratory comparison exercise. As a result it can be stated that the majority of participating laboratories could determine the pH values very exactly. The pH values obtained are ranging only little around the calculated mean (less than 0.1 pH unit). It could also be demonstrated that the participating institutes could analyse both, pH values in fishery products and aqueous salt solutions. However, also in this exercise a number of outliers and deviating values have been detected. Therefore it is of utmost importance to calibrate the pH electrodes in regular intervals and to maintain them carefully. Intra-laboratory comparison measurements are recommended to detect weak points.
