    Description: We report on the validation of profiles from the MIPAS-Envisat on-line processor of ESA version 4.61 with correlative measurements derived from MIPAS-STR onboard the high-altitude aircraft M55-Geophysica. The validation is made for the July 22, 2002, orbit 2051 in the region of the campaign base in Forli, Italy and for several orbits in February / March 2003 around Kiruna, northern Sweden. This paper includes a careful comparison of the MIPAS-STR data with O3 sondes and with in-situ measurements from the Geophysica obtained during ascent, descent and occasional dives. To obtain accurate N2O and CH4 profiles close to the MIPAS-Envisat tangent points we used the MIPAS-STR measurements of CFC-11 and CFC-12 and correlations obtained by the HAGAR instrument.

    Notes: The viscoelastic behavior of concentrated dispersions of adhesive hard spheres was investigated. By changing the temperature, the interaction potential of the particles was varied from a hard sphere repulsion to a strong attraction. Using torsion resonators and a nickel tube resonator the complex viscosity was measured in the frequency range 70–250 000 Hz. The results were described on the basis of a simple mechanical model in terms of a series of relaxation times and a single relaxation strength. The temperature dependence of the longest relaxation time and the relaxation strength indicated that the system undergoes a kinetic transition with decreasing temperature. The transition could be identified with the percolation threshold. Above the transition temperature where the dispersion is in a stable, fluid state, linear viscoelastic behavior is observed. Below the percolation threshold but still in the one-phase region, nonlinear viscoelasticity was measured, even at the smallest strains.

    Notes: We describe a new counter-rotating Couette apparatus that has been developed for deformation studies of single sub-millimeter sized particles in shear flow. New features are the adaption to the low viscosities of water-based systems and temperature control of the device. The inner to outer radius ratio of the cylinders used is 0.9785 and the height to width aspect ratio of the gap is 4.0, while the inner radius is 100 mm. Because of the limited particle size a high mechanical accuracy of the Couette geometry is necessary. The swing of the inner cylinder is less than 2 μm and that of the outer cylinder less than 4 μm. We achieved this by carefully choosing the design parameters of the aerostatic bearing and the coupling between cylinder and motor unit. Furthermore, special drive units give a shear rate resolution of 0.018 s−1, while the maximum shear rate is 100 s−1. For a liquid viscosity on the order of 1 mPas the effective maximum shear rate is 30 s−1. We have shown that deformations as small as (L−B)/(L+B)(approximate)0.01 of giant bilayer vesicles (typical radius 10 μm) with length L and width B can be observed with our device. © 1998 American Institute of Physics.

    Notes: The microwave spectrum of methyl hydroperoxide which has been investigated in the range of 12–307 GHz is dominated by the internal rotation of the OH group. The molecule exists in a gauche equilibrium configuration. Pure rotational transitions as well as much stronger torsional–rotational transitions have been assigned. A model of the overall rotation for a molecule with two strongly coupled tunneling states has been used for the analysis of all assigned transitions. Quantum-mechanical tunneling between the two equivalent gauche minima splits the ground torsional state by 448.7603(1) GHz. Additional small splittings of the microwave transitions have been attributed to the internal rotation of the CH3 group with a threefold barrier of 1120(10) cm−1. The permanent electric dipole components have been estimated from Stark splittings. Ab initio calculations of the molecular structure and of the potential functions for internal rotations of the OH and CH3 groups complement the experimental study.

    Notes: The genes encoding the α- and β-chains of the human interleukin-2 receptor were expressed in lepidopteran insect cells using the baculovirus expression vector system. The corresponding genes were inserted under the polyhedrin promoter of the Autographa california nuclear polyhedrosis virus and expressed in the Spodoptera frugiperda insect cell line during viral infection. The recombinant receptor proteins were identified in the insect cell lysates by using protein dot blot and ELISA techniques. At 36 h post infection the corresponding proteins were clearly detected using anti-IL-2 α- and β-receptor-specific antibodies. A large amount of the α-chain was also found in the supernatant culture media at 72 h post infection and metabolic labelling with [35S]-methionine indicated that it was proteolytieally cleaved into a 32 kDa soluble form. A similar soluble or secreted form of the β-chain was. however, not observed. Both receptor proteins were expressed on the surface of the insect cells as determined by flow cytometry analysis. Studies performed with the different IL-2 receptor forms (α- and β-chains alone or in combination) in the presence or absence of rIL-2 suggest that the receptor proteins when expressed in infected insect cells are non-functional with respect to tyrosine phosphorylation.

    Notes: Ethylene emission from wild-type Agrobacterium tumefaciens (C58)-induced stem tumours of Ricinus communis was continuously measured with two different methods, process gas chromatography and photo-acoustic spectrometry. Ethylene production was as high as 700 pmol g FW–1 h–1, namely 140 times greater than emitted by non-tumourized control stems. It was highest in 5-week-old tumours, independent of light, depressed by anoxia and, during water deficit it was stimulated by rewatering. A remarkable concomitant CO-production was discovered. Accumulation of 1-aminocyclopropane-1-carboxylic acid (ACC), the substrate of ACC-oxidase, preceded ethylene emission with a maximum 2 weeks after tumour induction. Simultaneously, the xylem in the tumour-adjacent host stem underwent drastic changes: it increased two to three times in thickness, vessel diameters decreased, the rays remained unlignified and became multiseriate. With increasing emission of ethylene aerenchyma developed in the non-transformed, tumour-surrounding tissue that formerly was stem cortex. Cotyledons reacted with epinastic symptoms indicating induction of senescence. The present results reveal an important role of ethylene, in addition to cytokinin and auxin, for the differentiation and physiology of A. tumefaciens-induced tumours.

    Notes: Growth in stagnant, oxygen-deficient nutrient solution increased porosity in adventitious roots of two monocotyledonous (Carex acuta and Juncus effusus) and three dicotyledonous species (Caltha palustris, Ranunculus sceleratus and Rumex palustris) wetland species from 10 to 30% under aerated conditions to 20–45%. The spatial patterns of radial oxygen loss (ROL), determined with root-sleeving oxygen electrodes, indicated a strong constitutive ‘barrier’ to ROL in the basal root zones of the two monocotyledonous species. In contrast, roots of the dicotyledonous species showed no significant ‘barrier’ to ROL when grown in aerated solution, and only a partial ‘barrier’ when grown in stagnant conditions. This partial ‘barrier’ was strongest in C. palustris, so that ROL from basal zones of roots of R. sceleratus and R. palustris was substantial when compared to the monocotyledonous species. ROL from the basal zones would decrease longitudinal diffusion of oxygen to the root apex, and therefore limit the maximum penetration depth of these roots into anaerobic soil. Further studies of a larger number of dicotyledonous wetland species from a range of substrates are required to elucidate the ecophysiological consequences of developing a partial, rather than a strong, ‘barrier’ to ROL.
