    Description: SUMMARY The relation between the complex geological history of the western margin of the North American plate and the processes in the mantle is still not fully documented and understood. Several pre-USArray local seismic studies showed how the characteristics of key geological features such as the Colorado Plateau and the Yellowstone Snake River Plains are linked to their deep mantle structure. Recent body-wave models based on the deployment of the high density, large aperture USArray have provided far more details on the mantle structure while surface-wave tomography (ballistic waves and noise correlations) informs us on the shallow structure. Here we combine constraints from these two data sets to image and study the link between the geology of the western United States, the shallow structure of the Earth and the convective processes in mantle. Our multiphase DNA10-S model provides new constraints on the extent of the Archean lithosphere imaged as a large, deeply rooted fast body that encompasses the stable Great Plains and a large portion of the Northern and Central Rocky Mountains. Widespread slow anomalies are found in the lower crust and upper mantle, suggesting that low-density rocks isostatically sustain part of the high topography of the western United States. The Yellowstone anomaly is imaged as a large slow body rising from the lower mantle, intruding the overlying lithosphere and controlling locally the seismicity and the topography. The large E–W extent of the USArray used in this study allows imaging the ‘slab graveyard’, a sequence of Farallon fragments aligned with the currently subducting Juan de Fuca Slab, north of the Mendocino Triple Junction. The lithospheric root of the Colorado Plateau has apparently been weakened and partly removed through dripping. The distribution of the slower regions around the Colorado Plateau and other rigid blocks follows closely the trend of Cenozoic volcanic fields and ancient lithospheric sutures, suggesting that the later exert a control on the locus of magmato-tectonic activity today.     Description: Studies of the frequency of heterozygous carriers for common inherited diseases of haemoglobin in over 7500 adolescent children in 25 districts in Sri Lanka have disclosed a highly significant variation over very short geographical distances. A further analysis of these findings, including their relationship to the past frequency and distribution of malaria, climatic variation, altitude, ethnic origin and consanguinity rates, have provided evidence regarding the evolutionary basis for the variable distribution of these conditions over short distances. It is likely that the complex interplay between malaria and the environment, together with related ethnic and social issues, exists in many countries across the tropical belt. Hence, these observations emphasise the importance of micromapping heterozygote distributions in high-frequency countries in order to define their true burden and the facilities required for the prevention and management of the homozygous and compound heterozygous disorders that result from their interaction.

    Notes: During wound healing, dermal fibroblasts switch from a migratory, repopulating phenotype to a contractile, matrix-reassembling phenotype. The mechanisms controlling this switch are unknown. A possible explanation is suggested by the finding that chemokines that appear late in wound repair prevent growth factor-induced cell-substratum de-adhesion by blocking calpain activation. In this study, we tested the specific hypothesis that fibroblast contraction of the matrix is promoted by a pro-repair growth factor, epidermal growth factor, and is modulated by calpain-mediated release of adhesions. We employed an isometric force transduction system designed to measure the contraction of a collagen matrix under tension by a population of NR6 fibroblasts transfected with the human epidermal growth factor receptor. By maintaining a fixed level of strain, we could monitor both the initial contraction and subsequent relaxation of the matrix. Epidermal growth factor stimulated a transient, dose-dependent increase in matrix contraction that peaked within 60 minutes and then decayed over the ensuing 3 to 6 hours. Calpain inhibitor I (ALLN) prevented epidermal growth factor-stimulated cell de-adhesion and resulted in a significantly slower decay of matrix contraction, with only a slight decrease of the peak magnitude of contraction. The mitogen-activated protein kinase kinase-1-selective inhibitor PD 98059 that blocks signaling through the extracellular signal-regulated kinase/mitogen-activated protein kinase pathway, required for epidermal growth factor receptor-mediated activation of calpain and de-adhesion, does not significantly affect the magnitude of matrix contraction within minutes of epidermal growth factor addition, but slows the decay similarly to calpain inhibition. Epidermal growth factor receptor signaling thus stimulates the complementary mechanisms of intracellular contractile force generation and calpain-mediated de-adhesion, which are known to coordinately facilitate cell migration. These findings suggest that calpain can act as a functional switch for transmission of intracellular contractile force to the surrounding matrix, with calpain-mediated de-adhesion reducing this transmission and corresponding matrix contraction. Countervailing processes that down-regulate calpain activation can, accordingly, direct the transition of cell function from locomotion to matrix contraction.

Macklin 1952 erstmals untersuchte Familie mit generalisierten oder lokalisierten Formen von Ichthyosis wurde 1970 erneut untersucht. Die Annahme eines autosomal dominanten Erbganges mit wechselnder Expressivität konnte erneut bestätigt werden. Mit einem IBM Computer Modell 1130 wurden marker-Analysen und Kopplungsstudien durchgeführt. Kopplung an den AB0-Locus in einer Entfernung von 10 Rekombinations-Einheiten konnte ausgeschlossen werden. Enge Kopplung wurde außerdem für die marker P, Rh, Duffy, Kidd, Gm, Haptoglobin, Ag und Phosphoglucomutase ausgeschlossen. Nicht für jeden der geprüften marker konnte Kopplung generell ausgeschlossen werden. Die beschriebenen Fälle werden auf Grund der klinischen, histopathologischen und elektronenoptischen Befunde als eigenständige nosologische Entität angesehen und von den übrigen hystrixartigen Ichthyosen abgetrennt.
    Notes: Summary A family examined by Curth and Macklin in 1952 and found to suffer from generalized or localized forms of ichthyosis, was reexamined in 1970. The concept that the disorder had been conditioned by a dominant gene which manifested itself with varying degrees of severity in the various family members, seemed proved again. Analysis for autosomal linkage was done with an IBM model 1130 computer. Linkage to the AB0 locus, as close as 10 recombination units was ruled out. Very close linkage was also excluded for P, Rh, Duffy, Kidd, Gm, haptoglobin, Ag, and phosphoglucomutase. Loose linkage was not ruled out with respect to any marker. Due to clinical, histopathological and ultrastructural findings, our cases are regarded as an independent nosological entity and are therefore to be separated from the other forms of hystrix-like ichthyosis.

    Notes: Abstract Intraesophageal manometry was performed on 14 healthy male volunteers divided into two groups. Group 1, composed of 4 subjects, received first phenylephrine (an alpha adrenergic stimulating drug) and then phentolamine (an alpha adrenergic blocking drug) intravenously. No effect on esophageal motility was observed following administration of either drug; however, adequate alpha stimulation or blockade of esophageal adrenergic receptors was probably not accomplished. Group 2 was composed of 10 subjects who received first isoproterenol (a beta stimulating drug) and then alprenolol (a beta blocking drug) intravenously. A significant decrease in resting pressure of the lower esophageal sphincter (LES) was observed in all of 9 subjects following isoproterenol. No effect was observed in the body of the esophagus. Following beta blockade with alprenolol, changes were observed in the body of the esophagus and in the LES. In the body of the esophagus, a rise in amplitude of contraction occurred in 9 of 10 subjects and prolongation of contraction was noted in 7 of 10 subjects. Changes in the LES were the most striking. Increase in amplitude of sphincteric contraction following deglutition was seen in 9 of 10 subjects. Prolongation of sphincteric contractions was most dramatic and was recognized in all 10 subjects. These data suggest an inhibitory beta receptor in the human distal esophagus. Stimulation of these receptors results in decreased LES resting pressure, while blockade of the receptors produces an increase in smooth muscle contraction—probably by removing inhibitory control.
