    Notes: Abstract The circular dichroism (CD) spectrum of isolated chlorosomes fromChloroflexus aurantiacus showed a conservative, S-shaped signal with a negative maximum at 723 nm, a positive maximum at 750 nm and a zero-crossing at 740 nm. Proteolytic treatment of chlorosomes with trypsin at 37°C did not change the CD signal or the absorption spectrum in contrast to treatment with proteinase K, where a twofold increase in rotational strength and a slight decrease of the absorption band at 740 nm were observed. Treatment with saturating 1-hexanol concentrations resulted in a blue shift of the absorption band at 740 nm as well as in changes of the CD spectrum. These changes reversed when the sample was diluted to half the saturating 1-hexanol concentration. In contrast to that, we observed an irreversible formation of a giant CD signal using the combination of 1-hexanol and proteinase K treatment. Electron micrographs of chlorosomes treated with both 1-hexanol and proteinase K showed large aggregates of multiple chlorosome size. By comparison of proteinase K induced effects with trypsin effects it appeared that the 5.7 kDa polypeptide has a structural role in the organisation of BChlc in the chlorosome.

    Notes: Capillary electrophoresis (CE) analysis of human serum proteins and apolipoproteins is described and compared with sodium dodecyl sulfate-polyacryl-amide gel electrophoresis and cellulose acetate membrane electrophoresis. Under optimized CE conditions apolipoproteins I could be determined directly in the serum and quantitated with a linear correlation between peak area and concentration up to a concentration of 100 mg/dL.

    Notes: A combination of high performance liquid chromatography (HPLC) and capillary electrophoresis (CE) spiking, Edman degradation, amino acid analysis and matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) was applied for the analysis of the primary structure of β-thymosins. CE has been used to resolve peaks which coelute on HPLC, as well as to help establish the identity of tryptic fragments in the peptide mapping experiments of the two highly homologous β-thymosins.

    Description: Background: The transcriptomes of several cyanobacterial strains have been shown to exhibit diurnal oscillation patterns, reflecting the diurnal phototrophic lifestyle of the organisms.The analysis of such genome-wide transcriptional oscillations is often facilitated by the use of clustering algorithms in conjunction with a number of pre-processing steps. Biological interpretation is usually focussed on the time and phase of expression of the resulting groups of genes.However, the use of microarray technology in such studies requires the normalization of pre-processing data, with unclear impact on the qualitative and quantitative features of the derived information on the number of oscillating transcripts and their respective phases. Results: A microarray based evaluation of diurnal expression in the cyanobacterium Synechocystis sp. PCC 6803 is presented. As expected, the temporal expression patterns reveal strong oscillations in transcript abundance.We compare the Fourier transformation-based expression phase before and after the application of quantile normalization, median polishing, cyclical LOESS, and least oscillating set (LOS) normalization.Whereas LOS normalization mostly preserves the phases of the raw data, the remaining methods introduce systematic biases. In particular, quantile-normalization is found to introduce a phase-shift of 180°, effectively changing night-expressed genes into day-expressed ones. Comparison of a large number of clustering results of differently normalized data shows that the normalization method determines the result. Subsequent steps, such as the choice of data transformation, similarity measure, and clustering algorithm, only play minor roles.We find that the standardization and the DTF transformation are favorable for the clustering of time series in contrast to the 12m transformation. We use the cluster-wise functional enrichment of a clustering derived by LOS normalization, clustering using flowClust, and DFT transformation to derive the diurnal biological program of Synechocystis sp.. Conclusion: Application of quantile normalization, median polishing, and also cyclic LOESS normalization of the presented cyanobacterial dataset lead to increased numbers of oscillating genes and the systematic shift of the expression phase. The LOS normalization minimizes the observed detrimental effects. As previous analyses employed a variety of different normalization methods, a direct comparison of results must be treated with caution.

    Description: During the International Polar Year (IPY) 2007- 2008 education and outreach became more and more important and many different initiatives were started to foster this aim. It got obvious that without a meaningful communication of scientific results to the broader public, including all ages, the very important research outcomes will not be understood and accepted by the public. As this is a key in order to convince stakeholders, decision makers, politicians, but also just normal individuals, that these outcomes are of high importance for everybody’s daily life it and there is a high need to be active and react, it must be the goal of every scientist to make its research as understandable as possible. Maybe even more important is this content is to raise the awareness of the next generation for sensitive issues such as global change, climate warming and the impacts of the different regions of the world. Inspired by the activities of the IPY also the permafrost community started to foster their education and outreach projects. As they recognized its importance the International Permafrost Association (IPA) even launched and Education and Outreach committee in 2010 and started the collaboration with many educational institutions such as the University of the Arctic or APECS. Financially supported by the IPA this committee started its work during the last years and already could achieve some substantial outcomes. For the upcoming International Conference on Permafrost in 2016 (ICOP 2016), that will be held in Potsdam, Germany, it is intended that education and outreach will play an even more important role. For this a special education and outreach conference team was created, that will be responsible for the activities during the conference. They are planning to involve international schools and teachers to the conference activities, which will be a very new and innovative initiative. This presentation will present the proposed actions during, but also before and after, the conference. This could include a video competition for international schools to attend the event, the involvement of one or two local schools and a workshop for international teachers. A special event could be an evening lecture held by a scientist that will be open for the public. It will be a semi-entertaining, semi-scientific presentation that will be easy understandable for non-scientists. One main goal of the planned education and outreach activities will also be the possibility for children and the public to get in touch with ‘real’ scientists, communicate with them and in that way somehow close the gap between science and the daily live. All activities and events will be organized by the education and outreach conference team but mainly supported by the IPA Education and Outreach committee as well as the German ‘coole Klassen’ (cool classes) project, an initiative of the working group ‘Polar Teachers’ of the German Association of Polar Research (DGP).

    Description: Polar research is an interdisciplinary and multi-faceted field of research ranging from history to geology and geophysics to social sciences and education. Thus, several different universities and institutions within Germany participate in polar research. The seminar series POLARSTUNDE, organized by the German Society for Polar Research (Deutsche Gesellschaft für Polarforschung) and the German National Committee of the Association of Polar Early Career Scientists (APECS Germany) regularly features different topics of German polar research. Although initially a "pandemic solution", the seminar series has established itself as a valuable and highly successful part of the German polar research landscape. The seminar series was held in German and was aimed at both scientists and the general public. This article addresses the first season of POLARSTUNDE and provides (1) comprehensive summaries of the talks and (2) insight into the planning and execution from an organizational point of view.
