    Description: The ocean is forced by the atmosphere on a range of spatial and temporal scales. In numerical models the atmospheric resolution sets a limit on these scales and for typical climate models mesoscale (〈500 km) atmospheric forcing is absent or misrepresented. Previous studies have demonstrated that mesoscale forcing significantly affects key ocean circulation systems such as the North Atlantic sub-polar gyre (SPG) and the Atlantic meridional overturning circulation (AMOC). Here we present ocean model simulations that demonstrate that the addition of realistic mesoscale atmospheric forcing leads to coherent patterns of change: a cooler sea surface in the tropical and subtropical Atlantic Ocean and deeper mixed layers in the sub-polar North Atlantic in autumn, winter, and spring. These lead to robust statistically significant increases in the volume transport of the North Atlantic SPG by 10% and the AMOC by up to 10%. Our simulations use a novel stochastic parameterisation—based on a cellular automata algorithm—to represent spatially coherent weather systems realistically over a range of scales, including down to the smallest resolvable by the ocean grid (∼10 km). Convection-permitting atmospheric models predict changes in the intensity and frequency of mesoscale weather systems due to climate change, so representing them in coupled climate models would bring higher fidelity to future climate projections.

    Description: The vascular bundle (VB) is a complex structure that resides in the inner stripe of the outer medulla. At present, the tubulovascular spatial organization of the VB, which is crucial for the formation of the osmolarity gradient and for solute transport, is still under debate. In this study, we used computer-assisted digital tracing combined with aquaporin-1 immunohistochemistry to reconstruct all tubules and vessels in the VB of the mouse kidney. We found, first, that the descending and ascending vasa recta travelled exclusively through the VB. The ascending vasa recta received no tributaries (no branches) along their entire path in the medulla and were not connected with the capillary plexus in the interbundle region. Second, a specific group of the descending vasa recta were closely accompanied by the longest ascending vasa recta, which connected only to the capillary plexus at the tip of the papilla. Third, the descending thin limbs of all short-looped nephrons travelled exclusively through the outer part of the VB. The loops of these nephrons (both descending and ascending parts) were distributed in a regular pattern based on their length. Finally, the thick ascending limbs of all long-looped nephrons were located at the margin of the VB (except a few within the VB), which formed a layer separating the VB from the interbundle region. In conclusion, our three-dimensional analysis of the VB strongly suggest a lateral osmolarity heterogeneity across the inner stripe of the outer medulla, which might work as a driving force for water and solute transport.

    Description: The traditional point of view is that in the ocean, the meridional transport of heat is achieved by the wind-driven and meridional overturning circulations. Here we point out the fundamental role played by ocean mixing processes. We argue that mixing (i.e., water mass conversion) associated with eddies, especially in the surface mixed layer, can play an important role in closing the ocean heat budget. Our results argue that the lateral mixing applied at the surface of ocean/climate models should be playing an important role in the heat balance of these models, indicating the need for physically-based parameterizations to represent this mixing.

    Description: Several genome-wide association studies (GWAS) have demonstrated the association between genetic variants in the major histocompatibility complex (MHC) region and chronic hepatitis B (CHB) virus infection, but it is still unknown about the disease-causing loci and potential mechanisms owing to the complicated linkage disequilibrium for this region. To systematically characterize the MHC variations in relation to the CHB infection, we fine mapped the MHC region on our existing GWAS data with SNP2HLA taken the Pan-Asian panel as reference and finally identified four independent associations. The HLA-DPβ1 amino acid positions 84–87, which drove the effect of reported single nucleotide polymorphisms rs9277535 and rs3077, showed the most significant association (OR = 0.65, P = 2.03 x 10 –8 ). The Leu-15 of HLA-C, conferring the effect of rs3130542, increased the risk of CHB infection independently (OR = 1.61, P = 3.42 x 10 –7 ). The HLA-DRβ1*13 , in perfect LD with glutamic at site 71, and rs400488, an expression quantitative trait locus for HLA-J , were newly identified to be associated with CHB infection independently (OR = 1.84, P = 3.84 x 10 –9 ; OR = 0.28, P = 6.27 x 10 –7 , respectively). HLA-DPβ1 positions 84–87 and HLA-DRβ1 position 71 implicated the P1 and P4 in the antigen-binding groove, whereas HLA-C position 15 affected the signal peptide. These four independent loci together can explain ~6% of the phenotypic variance for CHB infection, accounting for 72.94% of that explained by known genetic variations. We fine mapped the MHC region and identified four loci that independently drove the chronic HBV infection. The results provided a deeper understanding of the GWAS signals and identified additional susceptibility loci which were missed in previous association studies.

    Description: U-Pb ages of detrital zircons from Well LS33A at Lingnan Low Uplift of the Qiongdongnan Basin (QDNB) are measured with LA-ICP-MS technique. Th/U ratios of detrital zircons mostly with oscillatory zoning are higher than 0.1, indicating a magmatic origin. Zircon U-Pb data of the Yacheng Formation yield an age range from 95.8 to 387.9 Ma, with two major clusters of 95-106 Ma and 214-237 Ma, representing Yanshanian and Indosinian, respectively. U-Pb age range of the Yinggehai-Ledong Formations varies widely from 87 to 2 490 Ma, exhibiting four remarkable age peaks of 88 Ma, 146 Ma, 208 Ma and 386 Ma on age spectrum. The variation in zircon age assemblage reveals that in provenance.     Description: Background In recent years, there has been increased interest in the use of ultrasound technology in the evaluation of spinal and paraspinal regions. Objective This study aimed to investigate trends in spinal ultrasound research from 1994 to 2015 and compare the contributions of such research from different countries and authors. Study design Bibliometric analysis. Setting Publications related to spinal ultrasound from 1994 to 2015 were retrieved from the Web of Science database. Methods Excel 2013, GraphPad Prism 5, and VOSviewer were used to summarise bibliometric features, including the number of publications, citation frequency, H-index, and country contributions and hotspots (keywords of popular scientific fields). Results A total of 3859 papers were included. The global inflection point (the point in time when the publication growth rate moved from positive to negative) came in 2010. The United States contributed the largest percentage of articles (1041; 26.9%), with the most citations (19 848) and the highest H-index (61). The journals Osteoporosis International and Spine had the highest publication number. The University of Toronto and the University of California, San Francisco were the most contributive institutions. Studies could be divided into three clusters: surgery, osteoporosis, and others. The keywords ‘adolescent idiopathic scoliosis’ and ‘anaesthesia’ were the latest hotspots, appearing around 2012. Conclusion Spinal ultrasound literature has grown continuously over the last 22 years, with the rate slowing down after 2010. The United States was the largest contributor in this field. Recent studies on topics related to ‘adolescent idiopathic scoliosis’ and ‘anaesthesia’ were relatively new and should be closely followed in spinal ultrasound research.

    Description: Osteoporosis is a metabolic bone disease characterized by decreased bone density and strength due to excessive loss of bone protein and mineral content. The imbalance between osteogenesis by osteoblasts and osteoclastogenesis by osteoclasts contributes to the pathogenesis of postmenopausal osteoporosis. Estrogen withdrawal leads to increased levels of proinflammatory cytokines. Overactivated osteoclasts by inflammation play a vital role in the imbalance. Matrine is an alkaloid found in plants from the Sophora genus with various pharmacological effects, including anti-inflammatory activity. Here we demonstrate that matrine significantly prevented ovariectomy-induced bone loss and inhibited osteoclastogenesis in vivo with decreased serum levels of TRAcp5b, TNF-α, and IL-6. In vitro matrine significantly inhibited osteoclast differentiation induced by receptor activator for NF-B ligand (RANKL) and M-CSF in bone marrow monocytes and RAW264.7 cells as demonstrated by tartrate-resistant acid phosphatase (TRAP) staining and actin-ring formation as well as bone resorption through pit formation assays. For molecular mechanisms, matrine abrogated RANKL-induced activation of NF-B, AKT, and MAPK pathways and suppressed osteoclastogenesis-related marker expression, including matrix metalloproteinase 9, NFATc1, TRAP, C-Src, and cathepsin K. Our study demonstrates that matrine inhibits osteoclastogenesis through modulation of multiple pathways and that matrine is a promising agent in the treatment of osteoclast-related diseases such as osteoporosis.—Chen, X., Zhi, X., Pan, P., Cui, J., Cao, L., Weng, W., Zhou, Q., Wang, L., Zhai, X.
