    Notes: Abstract  A set of 187 doubled haploid lines derived from the cross between cvs. Courtot and Chinese Spring was explored for QTLs for three bread-making quality tests: hardness, protein content and strength of the dough (W of alveograph). The scores of the parental lines were quite different except for protein content, and the population showed a wide range of variation. About 350 molecular and biochemical markers were used to establish the genetic map, and technological criteria were evaluated in 1 to 3 years. QTL detection was performed by the ”marker regression” method. The most significant unlinked markers were used in the model as covariates, and the results were tested by bootstrap resampling. For hardness, we confirmed a previously tagged major QTL on chromosome 5DS, and two additional minor QTLs were found on chromosome 1A and 6D, respectively. For protein content two main QTLs were identified on chromosomes 1B and 6A, respectively. For W, three consistent QTLs were detected: two at the same location as those for hardness, on chromosomes 1A and 5D; the third one on chromosome 3B. Therefore, it appeared that except for the Glu-1A locus, storage protein loci were not clearly involved in the genetic control of the criteria studied in the present work. Despite the reasonable size of the population no QTL with interactive effects could be substantially established as measured. All computations were carried out using home-made programmes in Splus language, and these are available upon request.

    Notes: Abstract  Plant height in wheat (Triticum aestivum L. em Thell) is known to be under polygenic control. Crosses involving genes Rht-B1 and Rht-D1, located on chromosomes 4BS and 4DS, respectively, have shown that these genes have major effects. Two RFLP loci were found to be linked to these two genes (Xfba1-4B with Rht-B1 and Xfba211-4D with Rht-D1) by genotyping a population of F1-derived doubled-haploid lines [‘Courtot’ (Rht-B1b+Rht-D1b)בChinese Spring’]. Using a well-covered molecular marker map, we detected three additional regions and one interaction influencing plant height. These regions, located on chromosome arms 4BS (near the locus Xglk556-4B), 7AL (near the locus Xglk478-7A) and 7BL (near the locus XksuD2-7B) explained between 5% and 20% of the variability for this trait in this cross. The influence of 2 loci from chromosome 4B (Xfba1-4B and Xglk556-4B) suggests that there could be a duplication of Rht-B1 on this chromosome originating from Cv ‘Courtot’. Moreover, an interaction effect between loci from chromosome arms 1AS (near the locus Xfba393-1A) and 1BL (near the locus Xcdo1188-1B) was comparable to or even higher than those of the Rht-B1b and Rht-D1b alleles. A model including the main effects of the loci from chromosomes 4B and 4D (Xfba1-4B, Xglk556-4B and Xfba211-4D) and the interaction effect between Xfba393-1A and Xcdo1188-1B is proposed, which explains about 50% of the variation in plant height. The present results are discussed in relation to those obtained using nullisomic or substitution lines.

    Notes: Abstract  An intervarietal molecular-marker map was used for the detection of genomic regions influencing crossability between wheat (Triticum aestivum L. em Thell) and rye (Secale cereale L.). Analysis of deviance and logistic marker-regression methods were conducted on data from doubled haploid lines from a cross between “Courtot” and “Chinese Spring”. A major quantitative trait locus (QTL) involved in crossability, associated with the marker Xfba367-5B, was detected on the short arm of chromosome 5B. An additional locus, Xwg583-5B, was indicated on the long arm of chromosome 5B. This minor QTL might correspond to Kr1 which was presumed to be the major gene controlling crossability. Another locus of the genome, Xtam51-7A on chromosome 7A, was significantly associated with this trait. Alleles of “non-crossability” were contributed by the non-crossable cultivar “Courtot”. The three-marker model explains 65% of the difference in crossability between the two parents. The present results are discussed in relation to those previously carried out to locate the Kr genes by using the telocentric mapping technique.

    Notes: Biosensors with animal and microbial cells immobilized close to the tip of a membrane electrode have been developed for chemical and drug testing. Our experimental results show that biosensors can be used for drug screening and to provide useful information about various cell-chemical interactions. A computer aided analysis (CAA) software package is being developed here using the biosensor for various screening purposes. This software package enables us to use a computer to analyze the biosensor dynamic responses. Computer simulation and parameter estimation techniques are used to select the best model and to describe the biochemical and pharmacologic effects of various chemicals and drugs on different cell lines.

    Notes: The alkaline proteases of B. subtilis NRRL B3411, B. pumilis, and B. licheniformis have been isolated by fractionation followed by ion exchange chromatography and their homogeneity demonstrated. General enzyme properties of the B. sublitis NRRL B3411 alkaline protease have been studied and attempts made to differentiate a group of alkaline proteases. It is clear that the alkaline proteases known as Subtilisins or Subtilopeptidases are not, exclusive to B. subtilis but are common to many Bacilli and therefore the generic name Bacillopeptidases has been proposed. It is clear too that on the basis of the effect of pH on activity, amino acid composition, esterase activity, and immunological cross-reactions the Bacillopeptidases can be divided into two groups or types: (a) Bacillopcptidase A (Subtilisin A or Subtilopeptidase A) which includes Subtilisin Carlsberg, B. licheniformis, and B. pumilis alkaline proteases; (b) Bacillopeptidase B (Subtilisin B or Subtilopeptidase B) which includes B subtilis NRRL B3411, Subtilisin Novo, Subtilisin BPN' (Nagarse), alkaline protease Daiwa Kasei, and (probably) B. subtilis var. amylosacchariticus. At present, no further differentiation is possible and whether or not the enzymes within group A or B are identical remains an open question. Methods for examination of crude enzyme mixtures or fermentation beers are described and from the examination of a number of crude enzymes and fermentation beers it appears that organisms producing Bacillopeptidase A do not produce neutral protease or amylase, while organisms producing Bacillopeptidase B produce a neutral protease and amylase as well.

    Notes: B. subtilis NRRL B3411 neutral protease has been extensively purified by solvent, and salt fractional ion, pigment removal with DEAE-cellulose followed by chromatography on hydroxylapatite, and a final passage through a Sephadex G-100 column. The neutral protease was shown to be homogeneous by disc gel and cellulose acetate electrophoresis, gel filtration chromatography, and ultra-centrifugation. The molecular weight was determined by osmometry and ultracentrifugation to be about 38-42,000 and the amino acid composition and zinc content determined. The general properties of the enzyme, pH-activity relationship, stability, effect of inhibitors, and specificity are discussed. Comparative studies were carried out on the B. subtilis NRRL B3411 and B. subtilis var. amylosacchariticus neutral proteases and these enzymes were found to be indistinguishable by the methods used, but quite distinct from the thermostable enzyme thermolysin from B. thermoprotcolyticus.

    Notes: The stability of the neutral and alkaline proteases in a Bacillus subtilis enzyme mixture was studied in aqueous solutions at room temperature. Stabilization of the proteases in solution for periods up to 25 days was achieved by the addition of various protein preparations including casein and soya protein. The degree of stabilization by casein was concentration dependent to about 2% protein. The instability of the neutral protease in solutions of the B. subtilis enzyme mixture was shown to be due primarily to proteolysis by the alkaline protease since the diisopropylfluorophosphate-treated enzyme was quite stable. Formulation of such enzyme solutions at low pH gave greater stability as did solutions containing an alkaline protease inhibitor from potatoes. A Conceptual approach to the formulation of enzyme solutions containing proteolytic enzyme to ensure maximum stability is proposed.

    Notes: The behavior of the film coefficient of heat transfer for the condensation of organic vapor mixtures was investigated experimentally to establish a satisfactory basis for applying the Nusselt equation to binary systems. Five ideal and nonideal pairs, all of which gave miscible condensates, were studied; the work was carried out under conditions of almost total condensation on a horizontal condensing surface designed to comply as rigidly as possible with the conditions for which Nusselt's equation is valid.The same behavior was observed for all systems and all concentrations studied: the experimental coefficients fell between those for the pure components and followed the behavior pattern for pure components when the temperature difference was taken as that between the bubble point of the condensate and the surface temperature, rather than between the dew point or the measured vapor temperature and the surface temperature, Correlation of the film coefficient showed it to vary approximately linearly with composition if the coefficients were compared at a constant value of the temperature difference, defined as above. This permits determination of the coefficient for a mixture by interpolation between the coefficients for the pure components, which are easily obtained, in preference to making the calculations with the properties of the mixture obtained by laborious and uncertain weighting of the corresponding properties of the pure components.When results are interpreted in the light of the theories of Colburn and Drew, the presence of a vapor-phase resistance to heat and mass transfer, as postulated by them, is indicated.

    Notes: Analytical solutions were obtained for temperature distributions in a single adiabatic fuel cell with heat generation by TΔS, I2R, and polarization. With these results it is possible to characterize heat transfer in a battery with convection in the fuel and air streams, and with conduction of heat in three directions, by means of an average temperature for which an analytical solution was also obtained. Although the temperature can be kept within safe limits in low-power density batteries, the results show that critical dimensions exist beyond which failure by thermal buckling will occur.

    Notes: An antibiotic eluting catheter has been designed to suppress bacteriuria associated with indwelling catheter drainage of the bladder. A coating of hydrophilicgel plastic, Hydron, is used as the vehicle from which the antimicrobial agent is locally released. In 15 dogs, indwelling Hydron-cephalothin catheters provided a high degree of protection against bacteriuria up to 96 hr. After 48 hr, all 12 control animals became infected.In preliminary clinical trials involving 60 definitive cases Hydron-cephalothin (Group II) and plain Hydron (Group III)-coated indwelling catheters prevented the development of bacteriuria to a significant degree compared with uncoated latex controls (Group I). At the end of 5 days, the cumulative per cent infected for Groups I, II, and III was 75, 33, and 4.7%, respectively. Increased lubricity and reduced urethral mocosal reaction at the catheter interface promote drainage and prevent accumulation of detritus at the meatus. An indwelling antibiotic eluting catheter appears to interdict bacterial invasion of the bladder by, through, and around the catheter.
