    Notes: Abstract In 27 inpatients with primary affective disorder the urinary excretion of 3-methoxy-4-hydroxyphenylglycol (MHPG) was measured prior to a 4-week treatment with 150 mg amitriptyline (AT)/day. Ratings according to the Hamilton depression scale were performed before therapy and repeated after 2 and 4 weeks. Plasma levels of AT, nortriptyline (NT), and E-10-hydroxynortriptyline (OHNT) were assayed weekly, and binding of AT to plasma proteins was determined in one sample. Better therapeutic results were obtained at intermediate, as compared to low and high concentrations of AT or AT plus NT. Independent evaluation of AT and metabolite levels revealed that patients with AT of 50–125 mg/ml responded particularly well when NT did not exceed 95 ng/ml or when NT plus OHNT was below 150 ng/ml. Outside this ‘therapeutic window’ the outcome was markedly poorer. Interindividual variation of AT binding was much smaller than variation of total concentrations. Evaluation of free, instead of total levels did not help to clarify the relationship between clinical and pharmacokinetic variables. Plasma levels within the optimal ranges were found in more patients with high than with low MHPG excretion. The frec fraction of OHNT in plasma of healthy subjects was about 35%.

    Notes: Summary Amitriptyline (AT) and the noradrenaline reuptake inhibiting antidepressant oxaprotiline (OT = hydroxymaprotiline) were compared in 59 primary depressive inpatients in a 4-week double blind parallel group design. In the Hamilton Depression Rating Scale and 2 self-rating scales AT proved to be more efficient than OT, mainly with respect to disturbances of appetite and sleep. Agitated patients receiving OT needed more additional tranquilizing medication. The number of side-effects did not differ. Both drugs increased heart rate and skin resistance level (SRL) to about the same degree and did not influence the number of spontaneous fluctuations of SRL, habituation of SRL orienting responses (OR), frequencies of respiration and blinking. Salivation was temporarily more impaired by AT. All physiological variables differed between patients and 30 healthy controls during the whole 4-week trial. Clinical outcome showed a linear relation to OT plasma levels. For AT a therapeutic window was confirmed for concentrations of AT and its metabolite nortriptyline between 125 and 200 ng/ml. Patients whose SRL-OR habituated rapidly had a better outcome than slow habituators. Urinary excretion of 3-methoxy-4-hydroxyphenylglycol was lower in patients than in controls but could not predict outcome with either drug.

    Notes: Abstract. Fluorine-18 fluorodeoxyglucose positron emission tomography (FDG PET) has proven useful in the differentiation of various tumour entities, including breast cancer. In patients with primary breast cancer we performed a 3-h imaging protocol to examine possible improvements in tumour detectability and image contrast. Twenty-nine patients with primary breast cancer with a diameter of ≥2 cm that was demonstrated to be malignant by biopsy or surgery were injected with 370–740 MBq 18F-FDG and scanned in the prone position. Data were acquired 0–40 min, 1.5 h and 3.0 h after injection. After correction for measured attenuation, decay and scatter and iterative reconstruction, standardised uptake values (SUVs) and tumour-to-non-tumour and tumour-to-organ ratios were calculated. Visual analysis was performed using transverse, sagittal and coronal slices as well as 3D reprojection images. Tumour-to-non-tumour and tumour-to-organ ratios were significantly higher for the 3-h images than for the 1.5-h images. SUVs did not increase to the same extent. Lesion detectability was 83% in 1.5-h images compared to 93% in 3-h images. We conclude that tumour contrast in breast cancer is improved by starting the PET acquisition at 3 h p.i. rather than at 1.5 h p.i.

    Description: Permafrost and related landforms (rockglaciers) are widespread phenomena in alpine geosystems. In the context of changing environments due to the significant warming, there is a need for thorough monitoring and analyzing the complex responses of these cryospheric geosystems. Here, the first-time application of wood anatomical methods in this context is presented in order to investigate whether rockglacier movement is reflected in varying cell structures of plants growing on top of the rockglaciers. In order to determine the influence of ground movements (by permafrost creep) and their influence on the conductive elements within roots of plants, wood-samples were taken from active and inactive rockglaciers in the Turtmann Valley, southern Swiss Alps. Since the occurrence of trees is limited altitudinally, the investigation was restricted to Swiss willow shrubs (Salix helvetica) frequently growing in permafrost areas above the timberline in the European Alps. This rather new approach concentrates on general vessel size differences as a result of mechanical stresses. The comparison of vessel sizes in roots of Swiss willow shrubs growing on active and inactive permafrost bodies depicts differences within the roots, which are related to the activity status of the respective rockglacier creep.

    Description: Wounding experiments, imitating the impact of geomorphic processes such as rock fall, were performed on Larix decidua Mill. and Picea abies (L.) Karst., before the beginning and at the end of the growing season. This was done to evaluate the timing and spread of traumatic rows of resin ducts (TDs) as a result of wounding during dormancy, when most mass movements occur. All treatments were successful in inducing TDs. However, a high inter- and intra-species variability concerning the timing and position of the TDs was noted. In larch, TDs were formed as first cells after wounding (immediate reaction) only in the vicinity of the wound. In contrast, none of the spruce trees exhibited immediate reactions. Neither larch nor spruce formed continuous TDs around the stem. TDs in larch were more extensive if, in addition, stems were bent before the growing season. In contrast, spruce showed this reaction when bent at the end of the growing season. The results indicate that the varying lengths of the growing seasons at different geographical locations need to be taken into account before an immediate wound reaction can be clearly defined, if an increase of the reliability and accuracy of process reconstructions is aimed for.
