    Description: Highlights: • Larvae upregulate genes associated with fatty acid and glycogen synthesis under moderate ocean acidification (OA) • Larvae under high levels of OA fail to regulate • Dysfunctional metabolism and stress associated with pathologies in internal organs • Lack of differential gene regulation and stress response in juveniles correspond to a higher resilience to OA stress Elevated environmental carbon dioxide (pCO2) levels have been found to cause organ damage in the early life stages of different commercial fish species, including Atlantic cod (Gadus morhua). To illuminate the underlying mechanisms causing pathologies in the intestines, the kidney, the pancreas and the liver in response to elevated pCO2, we examined related gene expression patterns in Atlantic cod reared for two months under three different pCO2 regimes: 380 μatm (control), 1800 μatm (medium) and 4200 μatm (high). We extracted RNA from whole fish sampled during the larval (32 dph) and early juvenile stage (46 dph) for relative expression analysis of 18 different genes related to essential metabolic pathways. At 32 dph, larvae subjected to the medium treatment displayed an up-regulation of genes mainly associated with fatty acid and glycogen synthesis (GYS2, 6PGL, ACoA, CPTA1, FAS and PPAR1b). Larvae exposed to the high pCO2 treatment upregulated fewer but similar genes (6PGL, ACoA and PPAR1b,). These data suggest stress-induced alterations in the lipid and fatty acid metabolism and a disrupted lipid homeostasis in larvae, providing a mechanistic link to the findings of lipid droplet overload in the liver and organ pathologies. At 46 dph, no significant differences in gene expression were detected, confirming a higher resilience of juveniles in comparison to larvae when exposed to elevated pCO2 up to 4200 μatm.

Abstract: Microplastics are ubiquitous to most marine environments worldwide, and their management has become one of the major challenges facing stakeholders. Here we monitored monthly, between March 2018 and March 2019, the abundance of microplastics (0.3–18.2 mm) at the sea surface within the Kiel Fjord, southwest Baltic Sea. Microplastics were sampled at eight locations, inside and outside the fjord, near potential source of microplastics, such as the outlets of storm drains or the Kiel-Bülk wastewater treatment plant, the Schwentine River mouth and the entrance of the Kiel Canal. Weather (wind, precipitations) and seawater (salinity, temperature) parameters were compared to the spatiotemporal distribution of the microplastics. We found an overall stable, and low (0.04 particles/m3), microplastic load within the Kiel Fjord compared to other urban areas worldwide with comparable population densities. No relationship was found between the microplastic abundance and the environmental factors, but the few samples that yielded unusually high amount of microplastics were all preceded by rainfall and snow/ice melt. During such events, vast amounts of water, potentially contaminated with microplastics, were released into the fjord via the storm drainage system. The microplastic abundances at the wastewater plant outflow were among the lowest of our survey, likely thanks to an efficient filtering system. The results of this study highlight the importance to repeat microplastic samplings over time and space to determine with confidence baseline microplastic abundance and to detect unusual acute contamination, especially during snow and ice melting. Overall, the microplastic abundance within the Kiel Fjord was low, probably thanks to efficient waste management on land. However, improvements are still needed to filter millimetre-sized particles within the storm drainage system, which is likely a major source of microplastics into the marine environment.

    Description: Multiple biochemical measurements were evaluated as an indirect measure of juvenile fish growth rate. Juvenile two-spotted gobies, Gobiusculus flavescens (Fabricius), caught in the Kiel Bight, were incubated in a temperature gradient table at 7 different temperatures ranging from 9 to 22.7°C for up to 28 days and sampled weekly. RNA/DNA ratios (RNA/DNA), protein and lipid amounts were measured in whole fish homogenates and compared with calculated weight-based growth rates of the individuals. RNA/DNA values were not significantly correlated with weight-specific growth rates. Lipid- and protein-based growth rates, on the other hand, were highly correlated with weight-specific growth (R2 of 0.4-0.5) and lipid-based growth rate explained 45.8% variability of weight-based growth in a linear growth model. Weight-based growth rates showed a dome-shaped relationship to temperature with a maximum around 16°C, a trend mirrored in lipid-based growth rates. The results indicate a stage-dependent shift in energy storage and metabolism with a decoupling of RNA/DNA as an index of weight-based growth rate as the juvenile gobies mature and lipids become the main determinant of weight-based growth in these fish.

    Description: A laboratory calibration study was undertaken with juvenile Sepia officinalis (80–85 g initial wet weight) to investigate the effects of different food rations and different starving intervals on RNA/dry weight (DW) ratios and RNA/DNA ratios in cephalopod mantle muscle at two different temperatures. The digestive gland index was also used as an additional indicator of recent growth. High food rations and low temperature went along with high RNA/DW ratios and high RNA/DNA ratios. Starving resulted in a linear decline in growth performance and a concomitant decrease in RNA/DW and RNA/DNA ratio, with RNA/DNA ratios representing the growth data better. RNA/DNA ratios decreased faster at higher temperatures. A fluorimetric assay for nucleic acid analysis was optimized for cephalopod mantle tissues and yielded reproducible RNA/DNA ratios with a relative variance below 10%. Thus, it may be possible to use this estimator of recently encountered feeding regime for the evaluation of mortality rates of early teuthid paralarvae to eventually support stock management. Also, log relative digestive gland weight showed a strong relationship with starving time, but, surprisingly, not with temperature. Data from the two temperatures analyzed could be combined to form a common regression line of relative digestive gland index with starving time. This indicator for recent growth might be especially suitable for large specimens with a well-developed digestive gland.

    Description: Brazilian sardine, the most important resource along the southeastern Brazilian coast, presented great variations and declines in its stocks. The main factors contributing to this are: oceanographic structure changes; recruitment failures; excessive catches of juveniles and increase in fishery effort. In spite of this, no alterations in the density-dependent parameters were detected. Consequently, methods analysing the condition of the larvae coupled with methods determining growth using sagittae otolith increment width were applied to evaluate growth under experimental conditions. The results of the readings on the sagittae were compared with the age of the laboratory-reared sardine larvae and confirmed that increments are formed on a daily basis. Under poor feeding conditions, sardine larvae showed a low growth expressed by dry weight, RNA/DNA ratio and tryptic enzyme activity and by the narrow and low contrast increments in the otoliths. The results of the biochemical indices showed an unexpected decline in the feeding group coupled with a decrease in width of increment numbers 8 and 10. Other factors than food availability were affecting the condition of the larvae and might be indicative of physiological processes and ontogenetic changes occurring in sardine larvae.

    Description: A significant part of the variation in the early life history traits of fish can be ascribed to the parental origin of the individual larvae. The primary source of this parental contribution has been attributed to maternal effects and evidence for paternal effects is equivocal. Maternal effects are a non-genetic contribution of a female to its offspring but most reported maternal effects are products of both genetic and non-genetic contributions, i.e. female effects. In this study, parental effects on traits of larvae of Atlantic herring (Clupea harengus L.) at hatch were investigated at one temperature using a 5 × 3 factorial mating design (North Carolina Design II). This allowed estimation of the true maternal effect and the additive genetic variation (heritability). Furthermore, relationships between individual traits were examined and for the first time nucleic acid content and otolith size at hatch were examined together. A significant correlation between the two was found and it is argued to support the notion that otolith growth is more related to metabolic rate than to somatic growth. Maternal effects were detected in larval weight and yolk-sac volume, while paternal and, hence, genetic effects appeared in larval length, yolk-sac volume, RNA : DNA ratio, and lapillar area. The findings suggest that an increased emphasis should be placed upon the importance of male influence on success of early larval fishes.

This study documents how the abundance of microplastics (〈5 mm) in the Atlantic cod, Gadus morhua, relates to the changes of the fish diet during years with contrasting levels of anoxia for example following years of low or high major Baltic inflows (MBI). A MultiNet Maxi trawl and CTD were deployed annually to collect microplastic samples alongside oxygen, temperature, and salinity conditions. Microplastics were homogenously distributed both within the water column and across years. Gadus morhua diet shifted from dominantly benthic invertebrates (61 %) under oxygenated conditions to dominantly Sprattus sprattus (81 %) under anoxic conditions. The proportion of G. morhua with microplastics in their digestive tract increased when they fed on pelagic fish (38 %) versus on benthic invertebrates (15 %). The proportion of S. sprattus which ingested microplastics (~18 %) did not vary. As anoxia at depth is expected to increase due to climate change, microplastic ingestion by G. morhua will potentially increase.

Western Baltic spring spawning herring (Clupea harengus, L.) is a commercially important fish stock currently suffering a strong decline. Larval survival is essential for stock recruitment and can be substantially decreased by predation. The comb jelly Mnemiopsis leidyi A. Agassiz, 1865, is a lobate ctenophore which is invasive to the Baltic Sea and a known ichthyoplankton predator. However, predation on herring larvae in the Baltic Sea by M. leidyi has not been studied since its initial establishment in 2006. To address this knowledge gap, we conducted feeding experiments to investigate (1) the predation capability of M. leidyi on herring yolk-sac larvae, and (2) the influence of larval age, (3) predator size and (4) the presence of alternative prey on the clearance rate of M. leidyi on herring yolk-sac larvae. Our results showed that M. leidyi exhibited the ability to capture and digest herring larvae. The clearance rates of M. leidyi on herring larvae decreased with larval age and increased with predator size, while the presence of alternative prey had no effect on clearance rates. This finding suggests that M. leidyi can efficiently consume herring yolk-sac larvae under laboratory conditions. However, further investigations using mesocosm or field studies are necessary to evaluate the potential impact of M. leidyi on the mortality and recruitment of herring yolk-sac larvae under Baltic Sea field conditions.
