    Notes: Objective:  A possible contact allergy to herbal teas, in particular those derived from the Asteraceae plant family, was investigated in patients allergic to sesquiterpene lactones (SL).Method:  Twenty patients with a contact allergy to SL were recalled and patch-tested with aqueous, ethanol and acetone extracts of 8 different herbal teas based on Asteraceae plants as well as with parthenolide and other SL.Results:  In 18/20 patients with SL allergy there were positive test reactions to the Asteraceae teas, mainly to those based on German chamomile, dandelion and milfoil. Among the SL, parthenolide was the most frequent co-reactor.Conclusion:  Most patients with a contact allergy to SL are allergic to commercial teas derived from the Asteraceae plant family as well.

    Notes: Background:  Hydrochloric acid is formed in water solutions of gold trichloride. Hydrochloric acid in contact with aluminium generates hydrogen gas which can reduce and transform trivalent gold to elemental gold.Objective:  To investigate whether patch testing with aqueous gold trichloride can cause false positive (irritant) reactions in patients without contact allergy to gold and false negative reactions in patients with gold allergy.Methods:  13 patients with and 13 patients without positive patch test reactions to gold sodium thiosulfate were tested with gold trichloride in 2 different vehicles, water and alkaline buffer, using 2 different test techniques, the Finn Chamber technique with aluminium chambers and van der Bend technique with polypropene chambers.Results:  Irritant patch test reactions were obtained with aqueous gold trichloride tested in van der Bend chambers in 10 patients without gold allergy. In gold-allergic patients no positive test reactions were obtained from aqueous gold trichloride in Finn chambers while 2 positive test reactions were obtained from gold trichloride in alkaline buffer tested in van der Bend chambers.Conclusion:  If gold trichloride is patch tested in wrong vehicle and with wrong test technique irritant test reactions may occur which can be misinterpreted as positive allergic reactions in patients without gold allergy as well as negative reactions in patients with gold allergy.

    Description: In 1988 and 1989 the occurrence of external diseases in fish from the German part of the Wadden Sea was quantified. Thirty-two stations, located along seven transects including four estuaries, were sampled monthly or at 3-month intervals with shrimp trawlers: more than 124 000 fish were studied. Six percent of the seven dominant fish species of 〉 12 cm length were diseased. In fish species which complete their life cycle in the Wadden Sea (gobies, hooknose, eelpout, sea scorpion) the total disease incidence was below 0.4%. In whiting, plaice and sole it was between 0.5 and 2.5%, while 6–8% of smelt, cod, eel, dab and flounder suffered from external lesions. The incidence of most diseases increased with increasing fish length. Similar geographical patterns in prevalence were observed for: (1) two types of skeletal deformities, (2) lymphocystis in dab and flounder, (3) papillomatosis in smelt and dab and (4) several infectious ulcerative diseases. Most of these ulcerative diseases of cod and flounder occurred on central estuarine stations, suggesting an impact of the relatively low, but greatly variable salinity on disease development. Buccal granulomatosis of smelt and papillomatosis of eels showed the highest incidence in the Elbe estuary as the most heavily polluted region of the Wadden Sea. A causal relationship between disease development and pollution, however, is not yet clear and requires experimental evidence. The disease types found are ranked into three priority groups regarding their indicator value for a pollution monitoring on the basis of fish diseases. There is evidence that the total disease load of fish in the Wadden Sea is higher than in other shallow coastal regions outside the North Sea. Little is known of the effects of these diseases on single fish and fish populations. “Yellow pest” of cod, which was recorded for the first time during this survey, and which occurred with an incidence of up to 14% in single estuarine samples, causes the most serious lesions and is supposed to be lethal. In total, however, the effects of a number of pathogenic parasites on fish survival is considered to be more serious than that of deformities and infectious diseases.

    Description: Apical membrane targeting of the collecting duct water channel aquaporin-2 (AQP2) is essential for body water balance. As this event is regulated by Gs coupled 7-transmembrane receptors such as the vasopressin type 2 receptor (V2R) and the prostanoid receptors EP2 and EP4, it is believed to be cAMP dependent. However, on the basis of recent reports, it was hypothesized in the current study that increased cAMP levels are not necessary for AQP2 membrane targeting. The role and dynamics of cAMP signaling in AQP2 membrane targeting in Madin-Darby canine kidney and mouse cortical collecting duct (mpkCCD 14 ) cells was examined using selective agonists against the V2R (dDAVP), EP2 (butaprost), and EP4 (CAY10580). During EP2 stimulation, AQP2 membrane targeting continually increased during 80 min of stimulation; whereas cAMP levels reached a plateau after 10 min. EP4 stimulation caused a rapid and transient increase in AQP2 membrane targeting, but did not significantly increase cAMP levels. After washout of the EP2 agonist or dDAVP, AQP2 membrane abundance remained elevated for at least 80 min, whereas cAMP levels rapidly decreased. Similar effects of the EP2 agonist were also observed for AQP2 constitutively nonphosphorylated at ser-269. The adenylyl cyclase inhibitor SQ22536 did not prevent AQP2 targeting during stimulation of each receptor, nor after dDAVP washout. In conclusion, this study demonstrates that although direct stimulation with cAMP causes AQP2 membrane targeting, cAMP is not necessary for receptor-mediated AQP2 membrane targeting and Gs-coupled receptors can also signal through an alternative pathway that increases AQP2 membrane targeting.
