    Notes: Energy dispersive x-ray spectrometry (EDAX system), with scanning electron microscopy was used to detect diffusion of Zn from a source in human dentin and the system was tested for sensitivity and reproducibility. Sections of human teeth in which a ZnO-eugenol cement had been placed in a cavity in the dentin in vivo were prepared and x-ray microanalytical measurements carried out. We found that values of counts for Zn, Ca and P in the same field were linear up to a total counting time of 60 sec. We further found that reproducibility of values between repeat examinations of the same specimens on different days was excellent. The concentration of Zn in the dentin decreased exponentially with distance from source within the distance tested. We conclude that the system can be used to detect trace concentrations of Zn, and that accordingly the diffusion behavior of a variety of ions in dentin could be monitored.

    Notes: As previously reported, young rats on zinc-deficient diets develop a marked epithelial hyperplasia in the mucosa of the cheek, associated with a four-fold increase in the number of mast cells in the lamina propria. We now report the results of a quantitative and qualitative ultrastructural study of these mast cells. In contrast to the cells in mast cell-related disease, the cells were of normal morphology, but of consistently increased size, containing proportionally increased numbers of mitochondria and specific granules. The size distribution of the granules was as in controls. Microvilli were lengthened in proportion to the increase in the cell circumference. The number of nuclear pores was increased but their spacing was as in controls. In all these measurements, the experimental mast cells were a remarkably homogeneous population. We conclude that mast cells are capable of developing into cells of larger size showing evidence of greater activity. It may be that this development occurs in response to the same (unknown) stimulus which causes their number to increase. By virjuc of their increased number and size, mast cells may play an important role in the enhancement of the microcirculation commonly seen in conditions of epithelial hyperplasia.

    Notes: The histopathological and topographical alterations occurring during the early stages of chemical carcinogenesis in the hamster cheek pouch using 7, 12-dimethylbenz (α) anthracene (DMBA) were studied by light and scanning electron microscopy. At the scanning electron microscopic level, cellular overlapping and mild disturbance of intercellular bridges were noticed as early as the second week of DMBA application; these changes progressed with time. Clear epithelial dysplastic changes at the light microscopic level were detected by the sixth week of the experiment. The results of this investigation demonstrate the usefulness of scanning electron microscopy as an adjunct tool to study early alterations in cell morphology which occur in the stratified squamous epithelium of the hamster cheek pounch in response to a chemical carcinogen.
