    Notizen: Laser induced fluorescence probing of the nitric oxide fragment determines the distribution of rotational and vibrational energies of NO produced in the 226 and 280 nm photolysis of nitrobenzene. Combining these results with kinetic energy measurements using vacuum ultraviolet photoionization to detect the fragment gives a detailed view of the energy release in the photolysis. Boltzmann distributions describe the rotational state populations at both photolysis wavelengths. The rotational temperature of NO from the 226 nm photolysis is (3700±350) K, corresponding to an average rotational energy of (0.32±0.03) eV, and that of NO from the 280 nm photolysis is (2400±200) K, corresponding to an average rotational energy of (0.20±0.03) eV. We observe no vibrationally excited NO and place an upper limit of 10% on the fraction of nitric oxide produced in any one vibrationally excited state. Two different limiting models, impulsive energy release and statistical energy redistribution, both correctly predict much more rotational than vibrational excitation, but neither completely describes the observed internal and kinetic energies. The impulsive model finds more NO rotational and translational energy, but much less phenoxy fragment internal energy than we observe. The statistical model does better for the NO rotation and phenoxy fragment internal energy, but underestimates the translational energy substantially. A combination of these two types of behavior provides a physical picture that qualitatively explains our observations. It is likely that statistical energy redistribution occurs during the approach to the transition state for isomerization of nitrobenzene to phenyl nitrite and impulsive energy release dominates during the subsequent rupture of the CO–NO bond.

    Notizen: Helicobacter pylori has been identified as the major aetiological agent in the development of chronic gastritis and duodenal ulcer, and it plays a role in the development of gastric carcinoma. Attachment of H. pylori to gastric epithelial cells leads to nuclear and cytoskeletal responses in host cells. Here, we show that Rho GTPases Rac1 and Cdc42 were activated during infection of gastric epithelial cells with either the wild-type H. pylori or the mutant strain cagA. In contrast, no activation of Rho GTPases was observed when H. pylori mutant strains (virB7 and PAI) were used that lack functional type IV secretion apparatus. We demonstrated that H. pylori-induced activation of Rac1 and Cdc42 led to the activation of p21-activated kinase 1 (PAK1) mediating nuclear responses, whereas the mutant strain PAI had no effect on PAK1 activity. Activation of Rac1, Cdc42 and PAK1 represented a very early event in colonization of gastric epithelial cells by H. pylori. Rac1 and Cdc42 were recruited to the sites of bacterial attachment and are therefore probably involved in the regulation of local and overall cytoskeleton rearrangement in host cells. Finally, actin rearrangement and epithelial cell motility in H. pylori infection depended on the presence of a functional type IV secretion system encoded by the cag pathogenicity island (PAI).

    Beschreibung: Modeling sub-canopy elevation is an important step in the processing of waveform lidar data to measure three dimensional forest structure. Here, we present a methodology based on high resolution discrete-return lidar (DRL) to correct the ground elevation derived from large-footprint Laser Vegetation Imaging Sensor (LVIS) and to improve measurement of forest structure. We use data acquired over Barro Colorado Island, Panama by LVIS large-footprint lidar (LFL) in 1998 and DRL in 2009. The study found an average vertical difference of 28.7 cm between 98,040 LVIS last-return points and the discrete-return lidar ground surface across the island. The majority (82.3%) of all LVIS points matched discrete return elevations to 2 m or less. Using a multi-step process, the LVIS last-return data is filtered using an iterative approach, expanding window filter to identify outlier points which are not part of the ground surface, as well as applying vertical corrections based on terrain slope within the individual LVIS footprints. The results of the experiment demonstrate that LFL ground surfaces can be effectively filtered using methods adapted from discrete-return lidar point filtering, reducing the average vertical error by 15 cm and reducing the variance in LVIS last-return data by 70 cm. The filters also reduced the largest vertical estimations caused by sensor saturation in the upper reaches of the forest canopy by 14.35 m, which improve forest canopy structure measurement by increasing accuracy in the sub-canopy digital elevation model.

However, the evolution of Arctic temperatures during the rest of the Holocene is less clear. Proxy reconstructions, suggest a long-term cooling trend throughout the mid- to late Holocene, whereas climate model simulations show only minor changes or even warming. Here we present a record of the oxygen isotope composition of permafrost ice wedges from the Lena River Delta in the Siberian Arctic. The isotope values, which reflect winter season temperatures, became progressively more enriched over the past 7,000 years, reaching unprecedented levels in the past five decades. This warming trend during the mid- to late Holocene is in opposition to the cooling seen in other proxy records. However, most of these existing proxy records are biased towards summer temperatures. We argue that the opposing trends are related to the seasonally different orbital forcing over this interval. Furthermore, our reconstructed trend as well as the recent maximum are consistent with the greenhouse gas forcing and climate model simulations, thus reconciling differing estimates of Arctic and northern high-latitude temperature evolution during the Holocene.
