    Description: Western boundary currents (WBCs) play an essential role in regulating global climate. In contrast to their widely examined horizontal motions, less attention has been paid to vertical motions associated with WBCs. Here, we examine vertical motions associated with the major WBCs by analyzing vertical velocity from five ocean synthesis products and one eddy-resolving ocean simulation. These data reveal robust and intense subsurface upwelling systems, which are primarily along isopycnal surfaces, in five major subtropical WBC systems. These upwelling systems are part of basin-scale overturning circulations and are likely driven by meridional pressure gradients along the western boundary. Globally, the WBC upwelling contributes significantly to the vertical transport of water mass and ocean properties and is an essential yet overlooked branch of the global ocean circulation. In addition, the WBC upwelling intersects the oceanic euphotic and mixed layers, and thus likely plays an important role in ocean biological and chemical processes by transporting nutrients, carbon and other tracers vertically inside the ocean. This study calls for more research into the dynamics of the WBC upwelling and their role in the ocean and climate systems.

    Description: The Arctic Ocean is experiencing profound environmental changes as the climate warms. Understanding how these changes will affect Arctic biological productivity is key for predicting future Arctic ecosystems and the global CO2 balance. Here we use in situ gas measurements to quantify rates of gross oxygen production (GOP, total photosynthesis) and net community production (NCP, net CO2 drawdown by the biological pump) in the mixed layer in summer or fall from 2011 to 2016 in the Beaufort Gyre. NCP and GOP show spatial and temporal variations with higher values linked with lower concentrations of sea ice and increased upper ocean stratification. Mean rates of GOP range from 8 ± 1 to 54 ± 9 mmol O2·m−2·d−1 with the highest mean rates occurring in summer of 2012. Mean rates of NCP ranged from 1.3 ± 0.2 to 2.9 ± 0.5 mmol O2·m−2·d−1. The mean ratio of NCP/GOP, a measure of how efficiently the ecosystem is recycling its nutrients, ranged from 0.04 to 0.17, similar to ratios observed at lower latitudes. Additionally, a large increase in total photosynthesis that occurred in 2012, a year of historically low sea ice coverage, persisted for many years. Taken together, these data provide one of the most complete characterizations of interannual variations of biological productivity in this climatically important region, can serve as a baseline for future changes in rates of production, and give an intriguing glimpse of how this region of the Arctic may respond to future lack of sea ice.

    Description: Background: DNA methylation is thought to be extensively involved in the pathogenesis of many diseases, including major psychosis. However, most studies focus on DNA methylation alteration at promoters of protein-coding genes, despite the poor correlation between DNA methylation and gene expression. Methods: We analyzed differentially methylated regions and differentially expressed genes in patients with schizophrenia and bipolar disorder and normal subjects. Gene expression and DNA methylation were analyzed with RNA-seq and MeDIP-seq of post-mortem brain tissue (brain region BA9) cohort in five schizophrenia, seven bipolar disorder cases and six controls, respectively. Results: Here, we performed a large-scale integrative analysis using MeDIP-seq, coupled with RNA-seq, on brain samples from major psychotic and normal subjects and observed obvious discrepancy between DNA methylation and gene expression. We found that differentially methylated regions (DMRs) were distributed across different types of genomic elements, especially introns. These intronic DMRs were significantly enriched for diverse regulatory elements, such as enhancers and binding sites of certain transcriptional factors (e.g., Pol3). Notably, we found that parts of intronic DMRs overlapped with some intragenic miRNAs, such as hsa-mir-7-3. These intronic DMR-related miRNAs were found to target many differentially expressed genes. Moreover, functional analysis demonstrated that differential target genes of intronic DMR-related miRNAs were sufficient to capture many important biological processes in major psychosis, such as neurogenesis, suggesting that miRNAs may function as important linkers mediating the relationships between DNA methylation alteration and gene expression changes. Conclusions: Collectively, our study indicated that DNA methylation alteration could induce expression changes indirectly by affecting miRNAs and the exploration of DMR-related miRNAs and their targets enhanced understanding of the molecular mechanisms underlying major psychosis.

    Description: Background: Acute myeloid leukemia (AML) is the second-most common form of leukemia in children. Aberrant DNA methylation patterns are a characteristic feature of AML. GATA4 has been suggested to be a tumor suppressor gene regulated by promoter hypermethylation in various types of human cancers although the expression and promoter methylation of GATA4 in pediatric AML is still unclear. Methods: Transcriptional expression levels of GATA4 were evaluated by semi-quantitative and real-time PCR. Methylation status was investigated by methylation-specific PCR (MSP) and bisulfate genomic sequencing (BGS). The prognostic significance of GATA4 expression and promoter methylation was assessed in 105 cases of Chinese pediatric acute myeloid leukemia patients with clinical follow-up records. Results: MSP and BGS analysis showed that the GATA4 gene promoter is hypermethylated in AML cells, such as the HL-60 and MV4-11 human myeloid leukemia cell lines. 5-Aza treatment significantly upregulated GATA4 expression in HL-60 and MV4-11 cells. Aberrant methylation of GATA4 was observed in 15.0 % (3/20) of the normal bone marrow control samples compared to 56.2 % (59/105) of the pediatric AML samples. GATA4 transcript levels were significantly decreased in AML patients (33.06 ± 70.94; P = 0.011) compared to normal bone marrow/idiopathic thrombocytopenic purpura controls (116.76 ± 105.39). GATA4 promoter methylation was correlated with patient leukocyte counts (WBC, white blood cells) (P = 0.035) and minimal residual disease MRD (P = 0.031). Kaplan-Meier survival analysis revealed significantly shorter overall survival time in patients with GATA4 promoter methylation (P = 0.014). Conclusions: Epigenetic inactivation of GATA4 by promoter hypermethylation was observed in both AML cell lines and pediatric AML samples; our study implicates GATA4 as a putative tumor suppressor gene in pediatric AML. In addition, our findings imply that GATA4 promoter methylation is correlated with WBC and MRD. Kaplan-Meier survival analysis revealed significantly shorter overall survival in pediatric AML with GATA4 promoter methylation but multivariate analysis shows that it is not an independent factor. However, further research focusing on the mechanism of GATA4 in pediatric leukemia is required.

    Description: Background: We investigated the epidemiological and clinical data of all hand, foot, and mouth disease (HFMD) cases in a sentinel hospital of Shenzhen, China from 2009 to 2011. Methods: HFMD cases diagnosed in our institution were assessed from 2009 to 2011. Both epidemiological and clinical features were analyzed retrospectively. All the fatal cases were reported. Results: A total of 12132 patients were diagnosed with HFMD, of which 2944 (24.3%) were hospitalized. Of the 2944 hospitalized patients, the highest proportion of diagnosed cases were admitted in May and July (989/2944, 33.6%). In 2009 all severe HFMD cases were diagnosed with enterovirus 71 (EV71). In 2010 and 2011, some of the severe HFMD were diagnosed with Coxsackievirus A16 (CA16). Incidence was highest in 0-4-year old children, with males being predominant. There were sporadic cases with HFMD the whole year except in February. All cases were cured in 2009. Six deaths were reported during 2010 and 2011. Conclusions: EV71 can cause severe complications and deaths in our region. HFMD is an important public health problem in Shenzhen in spite of stringent measures taken in preschool centers. A high degree of vigilance should be maintained over the disease situation.
