    Notes: Abstract Theγ-rays of the decay of Mg27 were investigated with NaJ-crystals, andγ-γ-cascades had been sought for by a fast-slow-coincidence circuit. A method for an exact measuring of the random-coincidence spectrum and a stabilisation method of the multiplier gain is described. The twoγ-lines of 808 keV and 988 keV in coincidence with the 842 keVγ-line and the 817 keVγ-line in coincidence with the 1013 keVγ-line, reported by other authors, could not be observed. If these transitions exist, their intensity would be less than 10−3 per decay.

    Notes: Summary The movement of the rectangular shaped chloroplast ofMesotaenium belongs to the same type as that ofMougeotia. Especially the following facts are in agreement withMougeotia: 1. The low-energy movement (so-called positive phototaxis) is induced by the well known red far-red reaction system. 2. Polarized light induces this movement only when vibrating perpendicularly to the cell axis. 3. Polarized red light vibrating parallel to the cell axis is not only ineffective in inducing the movement but antagonizes the induction achieved by cross vibrating light. Therefore, unpolarized light is less effective than polarized light on a basis of equal energy (this latter effect has not been stated inMougeotia until now). In contrast toMougeotia, inMesotaenium it is impossible to induce movement by single short irradiations of only a few seconds or minutes, but irradiations of about 30 minutes are necessary, and therefore also the reciprocity law does not hold. The long time irradiation, however, can be replaced by 10 to 12 flashes of only a few seconds each with dark periods of 3 to 5 minutes between them. Such a fractionated induction is not only as effective as a continuous one, but now also the reciprocity law is valid for these flashes. The energy requirement is in the same order of magnitude inMesotaenium and inMougeotia. The essential difference betweenMesotaenium andMougeotia, we assume, is concerned with the stability of the induction process, which inMougeotia continues long enough to govern the whole movement, whereas inMesotaenium it is lost after a few minutes and therefore it must be repeated several times until the movement is completed.

    Description / Table of Contents: Summary In some yeast strains, which originally were unable to ferment sucrose, changing-over to sucrose fermenters has been observed. All strains in which changing had occurred belong to the offspring of a strain of Sacch. italicus var. melibiosi, which formerly has been crossed and analyzed in the Carlsberg laboratory. The distribution of changed strains among the offspring strongly points to monogenic behavior of the changing factor. The changing itself seems to be a mutation r→R of the gene responsible for fermentation of raffinose and sucrose; anyone of the M-genes seems not to be involved. In the “changed strains”, not all but only a few cells have been changed. So, with the replica-plate technique, we could separate unchanged and changed substrains from each other in a strain which originated from a single spore and therefore formerly consisted of genetically identical cells.
    Notes: Zusammenfassung Hefestämme, die sich bei der genetisch-biochemischen Analyse als unfähig zur Vergärung von Saccharose erwiesen hatten, zeigten bei erneuter Untersuchung teilweise eindeutige Saccharosegärung. Die Verteilung der jetzt Saccharose-positiven Stämme unter der Nachkommenschaft eines Hybridenstammes deutet auf monogenen Erbgang. Die Saccharose-positiv gewordenen Stämme sind nicht mehr einheitlich; vielmehr konnte mit der “replica-plate”-Methode in einem ursprünglich genetisch reinen Stamm eine Aufspaltung in Saccharosepositive und-negative Subkulturen erzielt werden. Wir halten eine Erhöhung der Mutationsrate r→R bei dem für Saccharosespaltung verantwortlichen Gen für die wahrscheinlichste Erklärung. Diese erhöhte Mutationsrate dürfte abhängig vom genetischen Milieu sein.

    Description / Table of Contents: Summary Among the offspring of Saccharomyces italicus var. melibiosi, crossed with some Carlsberg-Yeasts, we had to our disposal some strains with different numbers of Me-genes, responsible for splitting the disaccharide melibiose. In quantitative fermentation tests with suspensions of equal amounts of cells per volume, we found the adaptation time as well as the rate of melibiose fermentation to be dependent on the number of Me-genes in the genom. Whether different Me-genes are responsible for the synthesis of always identical melibiase molecules or of different types of melibiase, cannot be decided from our experiments.
    Notes: Zusammenfassung An einer Reihe von Hefestämmen, die sich durch die Anzahl von Me-Genen unterscheiden, wurde die Melibiose-Gärung in Abhängigkeit von der Anzahl dieser Me-Gene untersucht. Dabei wurde darauf geachtet, daß die Stämme sich im übrigen genetischen Milieu so wenig wie möglich unterschieden; das war möglich, da die zu vergleichenden Stämme jeweils aus der gleichen Kreuzung stammten. Die CO2-Bildung konnte als Kriterium für die Geschwindigkeit der Melibiosespaltung verwendet werden, da diese den begrenzenden Faktor darstellte. Quantitative Gärungsmessungen zeigten, daß die Adaptationszeit um so kürzer ist, je mehr Me-Gene sich im haploiden Genom befinden. In gleicher Weise ist die Gärgeschwindigkeit von der Anzahl der Me-Gene abhängig. Offen bleibt dabei noch die Frage, ob die verschiedenen Me-Gene für die Synthese verschiedener Melibiasen verantwortlich sind oder ob unter dem Einfluß aller Me-Gene ein und dasselbe Enzym gebildet wird.

    Notes: Summary Mougeotia cells with chloroplasts oriented in profile have been irradiated with small spots of monochromatic red polarized light in order to induce chloroplast movement. In these experiments, four factors have been varied: 1. the orientation of the vibration plane of the light in relation to the cell axis, 2. the localization of the spot, i. e. irradiation of the chloroplast or the cytoplasm, 3. the spot size, and 4. the duration of the irradiation. As a result of our experiments, we conclude that the photoreceptor molecules responsible for the light-induced chloroplast movement are localized in the cytoplasm. As the photoreceptor of this reaction is the well known phytochromesystem, we may assume that also in other plants the phytochrome is localized in the cytoplasm rather than in the chloroplast.

    Notes: Summary By means of partial illumination with small spots of monochromatic light, polarized obliquely to the cell axis, we could demonstrate that the phytochrome molecules inMougeotia are oriented in a spiral pattern around the cell surface. The lines of this spiral form an angle of about 45o with the cell axis. The degree of exactness of this orientation has been discussed.
