    Notes: Abstract There are few descriptions of the magnetic resonance (MR) appearance of hyperfunctioning adrenocortical tumours, particularly those occurring in childhood. We studied five patients, two girls and three boys, aged 6–14.3 years, presenting with clinical syndromes of adrenocortical hyperfunction. The diagnoses were Cushing's syndrome (n = 2) virilisation (n = 2) and Conn's syndrome (n = 1). Bio chemical features suggested an adrenal lesion in each case. MR and ultrasound were performed in all five cases, with CT in four. Each patient had a functional adrenal tumour secreting either cortisol, androgens or aldosterone alone, or a combination of cortisol, androgens and oestradiol. The histological diagnosis was adenoma in four cases and tumour of indeterminate nature in one case. MR clearly showed the tumours (diameter 1.0–7.5 cm), all the lesions being of high signal intensity relative to liver on T2-weighted sequences. CT revealed an adrenal mass in each of the four patients scanned, three of which enhanced after intravenous contrast medium injection. The multiplanar imaging of MR allowed better distinction from adjacent structures and also demonstrated an unenlarged contralateral adrenal gland. In the patient with a 1-cm Conn's adenoma the lesion was more easily seen on MR than CT. Ultrasound showed the four larger tumours but was unable to visualise the contralateral adrenal or the Conn's adenoma. In conclusion, the MR appearances of four adrenocortical adenomas and one indeterminate tumour in children are described. MR has been found to be at least equal to CT in the detection of these tumours, with some possible advantages. Both techniques are superior to ultrasound.

    Notes: Abstract The Sacred Heart Hospice, Sydney, was founded in 1890 and is the largest inpatient palliative-care facility in Australia. Patients with advanced cancer form the predominant patient group, although patients with HIV/AIDS account for approximately 20% of admissions. A community-outreach service, established in 1983, cares for more patients at home than in the Hospice. Recently the Hospice has participated in a number of clinical trials and intends to become a regional centre for palliative-care research, education and training.

Objective and Design: In the present study, we investigated the role of mast cells in a model of polyacrylamide gel (PAG)-induced inflammation in mice.¶Subjects: Balb/c mice and two strains of mast cell deficient mice (WBB6F1/J-W/Wv, WCB6F1/J-S1/S1d).¶Treatment: Various quantities of polyacrylamide gel (Bio-Gel P4) were injected subcutaneously in the backs of mice.¶Methods: Five hours after the injection of PAG the animals were euthanized, the injection sites lavaged and levels of LTB4, PGE2, TNFα and cells were determined.¶Results: Subcutaneous injection of PAG caused a time-dependent response characterized by the accumulation of inflammatory cells peaking at 10 h and the formation of LTB4, PGE2 and TNFα, peaking at 5 h. PAG injection into W/Wv or SL/SLd mice (mice lacking mast cells) resulted in an attenuated response, i.e. LTB4 levels were reduced by 60% and minimal cell influx was seen. The lack of mast cells caused about a 30% reduction in the levels of TNFα found.¶Conclusions: These data suggest that mast cells play a prominent role in the PMN influx, TNFα production and eicosanoid formation in the PAG-induced inflammatory response.

    Notes: Abstract Verrucous carcinoma is a rare squamous cell variant that may arise in the bladder. We present a case of verrucous carcinoma of the bladder with radiologic-pathologic correlations that demonstrate the characteristic frondlike growth pattern. Verrucous carcinoma is an invasive but not a metastizing lesion, and therefore recognizing this lesion may have prognostic and therapeutic implications.

    Notes: Abstract Diagnostic morphological characteristics of copepods of the genus Calanus are restricted largely to minor variations in secondary sex characteristics. This presents a persistent problem in the identification of individuals to species level, especially for immature stages. We have developed a simple molecular technique to distinguish between the North Atlantic Calanus species (C. helgolandicus, C. finmarchicus, C. glacialis and C. hyperboreus) at any life stage. Using the polymerase chain-reaction (PCR), the mitochodrial large subunit (16S) ribosomal RNA (rRNA) gene was amplified from individual copepods preserved in ethanol. Subsequent digestion of the amplified products with the restriction enzymes DdeI and VspI, followed by electrophoretic separation in 2% agarose (Metaphor, FMC Ltd), produced a characteristic pattern for each species. The versatility of the method is demonstrated by the unambiguous identification to species of any life stage, from egg to adult, and of individual body parts.

    Notes: Abstract By applying a microinjection technique previously used in cell biology, a pH-sensitive dye, 2′,7′-bis(2-carboxyethyl)-5,6-carboxyfluorescein) (BCECF), was injected into the gut of the copepod Calanus helgolandicus to determine pH under a range of feeding conditions. The median gut pH of the fore- and hindguts of starved individuals was 6.86 and 7.19, respectively. This was a consistently lower pH than that observed in copepods feeding on either the diatom Thalassiosira weissflogii, the dinoflagellate Prorocentrum micans, or the coccolithophorid Emiliania huxleyi (Strain 92D), all of which had a median gut pH〉7.0. In all treatments, the median pH of the foregut was more acidic than the median of the hindgut, implying that the foregut is the site of acid secretion. The results, which demonstrate pH values down to 6.11, are discussed in terms of coccolith dissolution, pigment degradation and digestive enzyme activity.

    Notes: Abstract Some investigators have proposed the measurement of aspartate transcarbamylase (ATCase) activity as a suitable technique for estimating zooplankton productivity. However, this technique has never been comprehensively evaluated under controlled laboratory conditions nor compared extensively with other productivity techniques in the field. This paper describes such an evaluation, using the reproduction of the calanoid copepod Calanus helgolandicus as an index of germinal growth, an important aspect of zooplankton productivity. ATCase activity and RNA:DNA ratio both increased significantly with increasing gonad maturation. In addition, ATCase activity in females responded immediately to changing food conditions, whilst changes in egg production lagged behind by ≃24 h. Shifting the ATCase data by 24 h revealed a significant and positive correlation of ATCase activity with egg production. However, in well-fed females, ATCase activity assayed at constant temperature was apparently independent of environmental temperature conditions, whilst egg production was temperature-dependent. In the field, ATCase activity was significantly correlated with egg production only in autumn and winter; when a complete seasonal cycle was considered, no correlation was found between the two measurements. These findings suggest that ATCase is involved partially in either germinal growth and/or in other biosynthetic processes of female C. helgolandicus; thus, ATCase activity does not reflect copepod egg-production with any certainty.

    Notes: Abstract We present heteronuclear three-dimensional gradient-NMR techniques for the resonanceassignment of exchangeable (-OH and -NH) protons in uniformly 13C isotopically enrichedoligosaccharides and for the measurement of 1H-1H nuclear Overhauser enhancementsinvolving these protons. These techniques are derived from conventional HOHAHA-HSQCand NOESY(ROESY)-HSQC experiments, and are illustrated in application to a sample ofuniformly 13C-enriched Galβ1-4GlcNAc, and demonstrate that a total of 35 ROEs involvingexchangeable protons can be detected and assigned. We present a quantitative analysis ofthese ROEs that can only be accommodated in a model of the solution behaviour of theoligosaccharide that involves considerable internal motion.
