    Notes: Plants regenerated from cell suspension cultures of palmarosa grass, Cymbopogon martinii (Roxb.) Wats, were analysed for somaclonal variation in five clonal generations. A wide range of variation in important quantitative traits, e.g. plant yield, height, tiller number, oil content and qualitative changes in essential oil constituents—geraniol, geranyl acetate, geranyl formate and linalool—were observed among the 120 somaclones screened. Eight somaclones were selected on the basis of high herb and oil yield over the donor line and high geraniol content in the oil. Based on performance in the field trials, three superior lines were selected, and maintained for five clonal generations. The superior lines exhibited a reasonable degree of stability in the traits selected.

    Notes: 1. Polyriboinosinic-polyribocytidylic acid (Poly I:Poly C), an interferon inducer was studied for its effect on gastric ulceration in rats. Polyriboinosinic-polyribocytidylic acid (1,2 and 4 mg/kg, i.m.) showed a dose-dependent inhibition of gastric ulcers induced by aspirin, cold restraint stress and pylorus ligation (Shay's model). Protective dose (PD50)±SEM values of Poly I:Poly C on these models of ulcers were 1.9±0.2, 2.3±0.4 and 2.8±0.4 (mg/kg, i.m.) respectively.2. Polyriboinosinic-polyribocytidylic acid (10-60 μg) produced dose-dependent inhibition of gastric proton pump (H+/K+-ATPase) activity in the gastric parietal microsomal fraction. The concentration of Poly I:Poly C causing a 50% inhibition (IC50)±SEM was found to be 17.6±1.2 μg.3. Polyriboinosinic-polyribocytidylic acid caused a significant decrease in free and total acid and pepsin and an increase in mucin content in Shay (pylorus-ligated) rat.4. Polyriboinosinic-polyribocytidylic acid did not exert a significant influence on isolated tissue preparations for anticholinergic (acetylcholine-induced contraction of guinea-pig ileum) and H2-anti-histaminic (histamine-induced contraction of rat uterus and guinea-pig auricle) activities.5. Thus, the present study indicates that Poly I:Poly C may possess anti-gastric ulcer activity as a result of inhibition of the gastric proton pump.

    Notes: Incubation of excised cucumber cotyledons (Cucumis sativus L.) with NH4Cl solutions exceeding 0.001 M inhibited their greening, fresh weight increases, and incorporation of 14C-leucine into insoluble N compounds. The absorption of 14C-leucine during incubation and retention of moisture by the excised cotyledons after incubation were greatly diminished by the NH4Cl treatments. Treatment with KCl solutions of the same concentrations as the NH4Cl stimulated the greening, fresh weight increases, and the absorption and incorporation of 14C-leucine. Desiccation of cotyledons stored at 5°C for 10 days was inhibited by dilute KCl solutions. The toxicity of NH4Cl was alleviated by KCl solutions at low concentrations.

    Notes: The feasibility of achieving a preassigned stress state by changing the stiffness of determinate and indeterminate pin connected structures is presented. A new force method with all element forces (or stresses or areas) as independent variables is developed. The possibility but futility of getting the element areas by inverting and pre-multiplying the coefficient matrix with the external load vector when areas are chosen as variables is shown. The relationship existing between the compatibility equations and the plastic strength of pin connected structures is illustrated. Possible design methods for trusses under single or multiplicity of load conditions, the design of truss geometry and the relations between fully stressed and minimum weight structures are examined and illustrated through several examples.

    Notes: An integrated force method is developed for the analysis of discrete structures in which all the element forces are taken as the independent variables instead of the conventional way of treating the redundants as the prime unknowns. Computationally the integrated force method is superior to the standard force method. The analogy between Beltrami-Michell formulation for continuous problems and the present formulation for discrete structures is presented. Illustrative examples for the determination of forces and displacements are presented for pin-connected and rigid-connected frame structures for various load conditions.

    Notes: The synthesis of 3-deoxy-D-arabino-heptulosonate-7-phosphate (DAHP) is the first commitment of resources toward aromatics production in Escherichia coli. DAHP is produced during a condensation reaction between phosphoenolpyruvate (PEP) and erythrose 4-phosphate (E4P) catalyzed by DAHP synthases (coded by aroF, aroG, and aroH). Stoichiometric analysis has shown a severe PEP limitation in the theoretical yield of DAHP production from glucose due to the phosphotransferase system (PTS) for sugar uptake. This limitation can be relieved by (i) the recycling of pyruvate from PEP using PEP synthase (Pps) or (ii) use of non-PTS sugars such as xylose. Previous studies have shown the usefulness of overexpressing tktA (encoding transketolase), aroG, and pps (PEP synthase) for DAHP production in an aroB strain unable to utilize DAHP further. In the present study we confirm the predictions of the stoichiometric analysis by introducing pps, tktA, and aroG into vectors under independently controlled promoters. In glucose medium, although TktA has some positive effect on the final DAHP concentration, it has no effect on the yield (percent conversion). With Pps overexpression, the DAHP concentration produced from glucose is increased almost twofold and the yield is approaching the theoretical maximum, as predicted by the stoichiometric analysis. However, this Pps effect is observed only in the presence of both increased AroG and TktA. In xylose mimimal medium, the final DAHP concentration and the yield are completely determined by the AroG activity. TktA and Pps play no or insignificant roles, and the yield can reach the theoretical maximum without overexpression of these two enzymes. The results shown here are important for both rational design of metabolic pathways and industrial production of aromatics such as tryptophan, phenylalanine, indigo, quinic acid, and catechol.

    Notes: The design optimization of air-breathing propulsion engine concepts has been accomplished by soft-coupling the NASA Engine Performance Program (NEPP) analyser with the NASA Lewis multidisciplinary optimization tool COMETBOARDS. Engine problems, with their associated design variables and constraints, were cast as non-linear optimization problems with thrust as the merit function. Because of the large number of mission points in the flight envelope, the diversity of constraint types, and the overall distortion of the design space, the most reliable optimization algorithm available in COMETBOARDS, when used by itself, could not produce satisfactory, feasible, optimum solutions. However, COMETBOARDS' unique features-which include a cascade strategy, variable and constraint formulations, and scaling devised especially for difficult multidisciplinary applications-successfully optimized the performance of subsonic and supersonic engine concepts. Even when started from different design points, the combined COMETBOARDS and NEPP results converged to the same global optimum solution. This reliable and robust design tool eliminates manual intervention in the design of air-breathing propulsion engines and eases the cycle analysis procedures. It is also much easier to use than other codes, which is an added benefit. This paper describes COMETBOARDS and its cascade strategy and illustrates the capabilities of the combined design tool through the optimization of a high-bypass-turbofan wave-rotor-topped subsonic engine and a mixed-flow-turbofan supersonic engine. ©1997 John Wiley & Sons, Ltd.

    Notes: Equimolar alternating copolymers of styrene and methyl methacrylate (prepared with Et1.5AlCl1.5, SnCl4, and ZnCl2) as well as equimolar random copolymer were treated with polyphosphoric acid at 135°C. The extent of cyclization of the alternating copolymers was about 40%, independent of the cotacticity of the copolymer, and there was little or no crosslinking. The random copolymer underwent only 10% cyclization and considerable crosslinking. The extent of cyclization of the alternating copolymer of styrene and methyl acrylate (prepared with Et1.5AlCl1.5) was the same as that of the random copolymer and was lower than that of the corresponding methyl methacrylate copolymer. Both alternating and random copolymers underwent extensive crosslinking.

    Notes: The traditional fully stressed method performs satisfactorily for stress-limited structural design. When this method is extended to include displacement limitations in addition to stress constraints, it is known as the Fully Utilized Design (FUD). Typically, the FUD produces an overdesign, which is the primary limitation of this otherwise elegant method. We have modified FUD in an attempt to alleviate the limitation. This new method, called the Modified Fully Utilized Design (MFUD) method, has been tested successfully on a number of problems that were subjected to multiple loads and had both stress and displacement constraints. The solutions obtained with MFUD compare favourably with the optimum results that can be generated by using non-linear mathematical programming techniques. The MFUD method appears to have alleviated the overdesign condition and offers the simplicity of a direct, fully stressed type of design method that is distinctly different from optimization and optimality criteria formulations. The MFUD method is being developed for practicing engineers who favour traditional design methods rather than methods based on advanced calculus and non-linear mathematical programming techniques. The Integrated Force Method (IFM) was found to be the appropriate analysis tool in the development of the MFUD method.
