    Notes: Generation of pure, metal-vapor plasmas with peak densities above 1019 cm−3 is reported. The plasma production is based on the explosive ablation of electrode material in the capillary μs discharge. The plasma density is controlled by varying the discharge parameters. High purity and density were achieved by optimizing the electrode configuration, the capillary material, and dimensions. © 1999 American Institute of Physics.

    Notes: Abstract. The technique of laser ablation is receiving increasing attention for applications in dentistry, specifically for the treatment of teeth (e.g. drilling of micro-holes and plaque removal). In the process of ablation a luminous micro-plasma is normally generated which may be exploited for elemental analysis. Here we report on quantitative Laser-Induced Breakdown Spectroscopy (LIBS) analysis to study the presence of trace minerals in teeth. A selection of teeth of different age groups has been investigated, ranging from the first teeth of infants, through the second teeth of children, to adults to trace the influence of environmental factors on the accumulation of a number of elements in teeth. We found a close link between elements detected in tooth fillings and toothpastes with those present in teeth.

    Description / Table of Contents: Transforaminal endoscopic spinal disc surgery is one of the newer minimal invasive percutaneous procedures for treatment of lumbar disc hernias. Both non-sequestered and sequestered hernias can be treated with this method. From January 1995 to December 1997 we conducted endoscopic discotomies in levels L3 to S1 in 80 patients. The patients were chosen according to strict indications. Initially local anesthesia was administered; as the learning curve grew spinal anesthesia was used. The results show that this method is effective and pathology targeted. Healthy tissue is conserved and the risk of postoperative fibrosis is low.
    Notes: Die endoskopische Bandscheibenoperation durch das Neuroforamen stellt eine der neueren minimalinvasiven, perkutanen Techniken in der operativen Therapie der lumbalen Bandscheibenvorfälle dar. Sowohl nichtsequestrierte als auch sequestrierte Bandscheibenvorfälle sind mit diesem Verfahren therapierbar. Von Januar 1995– Dezember 1997 wurde eine endoskopische Diskotomie an 80 Patienten in den Etagen L3–S1 ausgeführt. Die Patienten wurden anhand von bestimmten Ein- und Ausschlußkriterien sorgfältig ausgesucht. Die Eingriffe erfolgten anfänglich in Lokalanästhesie, mit zunehmender Erfahrung in Spinalanästhesie. Die bisher vorliegenden Ergebnisse sprechen für eine suffiziente, pathologiegezielte Prolapsbehandlung mit weitgehender Erhaltung der Restbandscheibe und geringerer postoperativer Fibrosierung.

    Notes: Abstract The levels of heavy metals in water and stream sediments in Jamaica are presented and compared to levels in soils and environmental standards. Heavy metals (cadmium, chromium, copper, mercury, nickel, lead and zinc) appear to be less of a problem to Jamaica's freshwater environment than might be expected given the concentrations in soils. This can be explained by taking into account the dynamics of the Jamaican water environment and the relative absence of anthropogenic sources. The dynamics of the freshwater system on the other hand, could pass any environmental threat to the coastal environment. Because the stream sediments are relatively coarse and contain little organic material the bio-availability of micropollutants such as heavy metals from the sediments can be expected to be relatively high.

    Description: Oceans are a net source of molecular hydrogen (H2) to the atmosphere. The production of marine H2 is assumed to be mainly biological by N2 fixation, but photochemical pathways are also discussed. We present measurements of mole fraction and isotopic composition of dissolved and atmospheric H2 from the southern and northern Atlantic between 2008 and 2010. In total almost 400 samples were taken during five cruises along a transect between Punta Arenas (Chile) and Bremerhaven (Germany), as well as at the coast of Mauretania. The isotopic source signatures of dissolved H2 extracted from surface water are highly deuterium-depleted and correlate negatively with temperature, showing δD values of (−629 ± 54) ‰ for water temperatures at (27 ± 3) °C and (−249 ± 88) ‰ below (19 ± 1) °C. The results for warmer water masses are consistent with biological production of H2. This is the first time that marine H2 excess has been directly attributed to biological production by isotope measurements. However, the isotope values obtained in the colder water masses indicate that beside possible biological production a significant different source should be considered. The atmospheric measurements show distinct differences between both hemispheres as well as between seasons. Results from the global chemistry transport model TM5 reproduce the measured H2 mole fractions and isotopic composition well. The climatological global oceanic emissions from the GEMS database are in line with our data and previously published flux calculations. The good agreement between measurements and model results demonstrates that both the magnitude and the isotopic signature of the main components of the marine H2 cycle are in general adequately represented in current atmospheric models despite a proposed source different from biological production or a substantial underestimation of nitrogen fixation by several authors.

    Description: The branched-chain amino acids (BCAAs; Ile, Leu, and Val) not only are important nutrients for the growth of Staphylococcus aureus but also are corepressors for CodY, which regulates virulence gene expression, implicating BCAAs as an important link between the metabolic state of the cell and virulence. BCAAs are either synthesized intracellularly or acquired from the environment. S. aureus encodes three putative BCAA transporters, designated BrnQ1, BrnQ2, and BrnQ3; their functions have not yet been formally tested. In this study, we mutated all three brnQ paralogs so as to characterize their substrate specificities and their roles in growth in vitro and in vivo . We demonstrated that in the community-associated, methicillin-resistant S. aureus (CA-MRSA) strain USA300, BrnQ1 is involved in uptake of all three BCAAs, BrnQ2 transports Ile, and BrnQ3 does not have a significant role in BCAA transport under the conditions tested. Of the three, only BrnQ1 is essential for USA300 to grow in a chemically defined medium that is limited for Leu or Val. Interestingly, we observed that a brnQ2 mutant grew better than USA300 in media limited for Leu and Val, owing to the fact that this mutation leads to overexpression of brnQ1 . In a murine infection model, the brnQ1 mutant was attenuated, but in contrast, brnQ2 mutants had significantly increased virulence compared to that of USA300, a phenotype we suggest is at least partially linked to enhanced in vivo scavenging of Leu and Val through BrnQ1. These data uncover a hitherto-undiscovered connection between nutrient acquisition and virulence in CA-MRSA.

    Description: Hdac3 is a key target for Hdac inhibitors that are efficacious in cutaneous T cell lymphoma. Moreover, the regulation of chromatin structure is critical as thymocytes transition from an immature cell with open chromatin to a mature T cell with tightly condensed chromatin. To define the phenotypes controlled by Hdac3 during T cell development, we conditionally deleted Hdac3 using the Lck-Cre transgene. This strategy inactivated Hdac3 in the double-negative stages of thymocyte development and caused a significant impairment at the CD8 immature single-positive (ISP) stage and the CD4/CD8 double-positive stage, with few mature CD4 + or CD8 + single-positive cells being produced. When Hdac3 –/– mice were crossed with Bcl-xL -, Bcl2 -, or TCR β-expressing transgenic mice, a modest level of complementation was found. However, when the null mice were crossed with mice expressing a fully rearranged T cell receptor αβ transgene, normal levels of CD4 single-positive cells were produced. Thus, Hdac3 is required for the efficient transit from double-negative stage 4 through positive selection.

    Description: A previous study on papillary thyroid carcinomas (PTC) in young patients who were exposed to 131 iodine from the Chernobyl fallout revealed an exclusive gain of chromosomal band 7q11.23 in exposed cases compared to an age-matched control cohort. CLIP2, a gene located within band 7q11.23 was shown to be differentially expressed between exposed and non-exposed cases at messenger RNA and protein level. Therefore, a standardized procedure for CLIP2 typing of PTCs has been developed in a follow-up study. Here we used CLIP2 typing data on 117 post-Chernobyl PTCs from two cohorts of exposed patients with individual dose estimates and 24 non-exposed controls to investigate a possible quantitative dose-response relationship of the CLIP2 marker. The ‘Genrisk-T’ cohort consisted of 45 PTCs and the ‘UkrAm’ cohort of 72 PTCs. Both cohorts differed in mean dose (0.59 Gy Genrisk-T, 1.2 Gy UkrAm) and mean age at exposure (AaE) (2 years Genrisk-T, 8 years UkrAm), whilst the median latency (16 years Genrisk-T, 18 years UkrAm) was comparable. We analyzed the association between the binary CLIP2 typing and continuous thyroid dose with logistic regression. A clear positive dose-response relationship was found for young PTC cases [age at operation (AaO) 〈 20 years, AaE 〈 5 years]. In the elder age group a higher proportion of sporadic tumors is assumed due to a negligible dose response, suggesting different molecular mechanisms in sporadic and radiation-induced cases. This is further supported by the association of elder patients (AaO 〉 20 years) with positivity for BRAF V600E mutation.

    Description: Cranial neural crest cells populate the future facial region and produce ectomesenchyme-derived tissues, such as cartilage, bone, dermis, smooth muscle, adipocytes, and many others. However, the contribution of individual neural crest cells to certain facial locations and the general spatial clonal organization of the ectomesenchyme have not been determined. We investigated how neural crest cells give rise to clonally organized ectomesenchyme and how this early ectomesenchyme behaves during the developmental processes that shape the face. Using a combination of mouse and zebrafish models, we analyzed individual migration, cell crowd movement, oriented cell division, clonal spatial overlapping, and multilineage differentiation. The early face appears to be built from multiple spatially defined overlapping ectomesenchymal clones. During early face development, these clones remain oligopotent and generate various tissues in a given location. By combining clonal analysis, computer simulations, mouse mutants, and live imaging, we show that facial shaping results from an array of local cellular activities in the ectomesenchyme. These activities mostly involve oriented divisions and crowd movements of cells during morphogenetic events. Cellular behavior that can be recognized as individual cell migration is very limited and short-ranged and likely results from cellular mixing due to the proliferation activity of the tissue. These cellular mechanisms resemble the strategy behind limb bud morphogenesis, suggesting the possibility of common principles and deep homology between facial and limb outgrowth.
