    Notizen: Raman spectroscopy is used to investigate strong band bending at the interface in semi-insulating substrates of ZnSe/GaAs heterostructures grown at high epitaxy rates. Direct evidence is given of the enhancement of polar modes strength, on the substrate side, by the electric field of the space-charge zone associated with Fermi-level pinning. The latter is qualitatively analyzed by following band flattening under illumination through the evolution of interfacial coupled LO-phonon–plasmon modes. Corresponding Raman line shapes are discussed within the phenomenological approach of D. H. Hon and W. L. Faust [Appl. Phys. 1, 241 (1973)]. © 1996 American Institute of Physics.

    Notizen: Long wavelength longitudinal optical (LO) and transverse optical (TO) phonons of BeSe and ZnxBe1−xSe layers are identified in a wide composition range by using Raman spectroscopy. A two-mode behavior is clearly evidenced. As predicted by the dielectric model of Hon and Faust, the eigenfrequencies of the BeSe- and ZnSe-like LO modes correspond to the maxima of Im〈−cursive-epsilon(ω, x)−1〉. Excellent agreement is obtained with a model where the calculations are performed by using the equations of motion and polarization derived from the modified random element isodisplacement model. Besides, the TO and LO frequencies for BeSe are determined to be 501 and 579 cm−1, respectively. © 2000 American Institute of Physics.

    Notizen: We compare the sensory profiles of six dark chocolates done by two types of juries: some trained juries and an untrained jury. Six laboratories, each one made up of 10 to 15 judges, are regarded as trained juries since the assessors were well trained before the evaluation of the products. The second type of jury is composed of only one panel of 29 untrained assessors and this jury is named the untrained jury. Fourteen attributes were evaluated and analyses of variance have been carried out by attribute to compare the sensory profiles of the six chocolates done by the trained juries and by the untrained jury. These analyses of variance show that the two types of juries give similar sensory profiles and that the few differences are mainly due to different ways of using the scale.

    Notizen: The growth pattern of the root system of young rubber trees (Hevea brasiliensis) was studied in relation to shoot development over a period of 3 months. Temporal and spatial variations in elongation and branching processes were examined for the different root types, by means of root observation boxes. Shoot growth was typically rhythmic. Root development was periodic and related to leaf expansion. Root elongation was depressed during leaf growth, whereas branching was enhanced. Consequently, highly branched areas with vigorous secondary roots alternated along the taproot with poorly branched areas with shorter roots. Root types were not affected to the same degree by shoot competition: during leaf expansion, taproot growth was just depressed but remained continuous, the emergence and elongation rates of secondary roots were significantly affected and the elongation rates of tertiary roots fell to zero. These results were consistent with the hypothesis that root growth is related to competition for assimilates and to the sink strength of the different root types, whereas root branching appeared to be promoted by leaf development.

    Notizen: We show here that CO2 partial pressure (pCO2) and temperature significantly interact on coral physiology. The effects of increased pCO2 and temperature on photosynthesis, respiration and calcification rates were investigated in the scleractinian coral Stylophora pistillata. Cuttings were exposed to temperatures of 25°C or 28°C and to pCO2 values of ca. 460 or 760 μatm for 5 weeks. The contents of chlorophyll c2 and protein remained constant throughout the experiment, while the chlorophyll a content was significantly affected by temperature, and was higher under the ‘high-temperature–high-pCO2’ condition. The cell-specific density was higher at ‘high pCO2’ than at ‘normal pCO2’ (1.7 vs. 1.4). The net photosynthesis normalized per unit protein was affected by both temperature and pCO2, whereas respiration was not affected by the treatments. Calcification decreased by 50% when temperature and pCO2 were both elevated. Calcification under normal temperature did not change in response to an increased pCO2. This is not in agreement with numerous published papers that describe a negative relationship between marine calcification and CO2. The confounding effect of temperature has the potential to explain a large portion of the variability of the relationship between calcification and pCO2 reported in the literature, and warrants a re-evaluation of the projected decrease of marine calcification by the year 2100.

    Notizen: In Enterobacter aerogenes, multidrug resistance involves a decrease in outer membrane permeability associated with changes in an as yet uncharacterized porin. We purified the major porin from the wild-type strain and a resistant strain. We characterized this porin, which was found to be an OmpC/OmpF-like protein and analysed its pore-forming properties in lipid bilayers. The porin from the resistant strain was compared with the wild-type protein and we observed (i) that its single-channel conductance was 70% lower than that of the wild type; (ii) that it was three times more selective for cations; (iii) a lack of voltage sensitivity. These results indicate that the clinical strain is able to synthesize a modified porin that decreases the permeability of the outer membrane. Mass spectrometry experiments identified a G to D mutation in the putative loop 3 of the porin. Given the known importance of this loop in determining the pore properties of porins, we suggest that this mutation is responsible for the novel resistance mechanism developed by this clinical strain, with changes in porin channel function acting as a new bacterial strategy for controlling β-lactam diffusion via porins.

    Notizen: The crystal structure of the family IIIa cellulose-binding domain (CBD) from the cellulosomal scaffoldin subunit (CipC) of Clostridium cellulolyticum has been determined. The structure reveals a nine-stranded jelly-roll topology which exhibits distinctive structural elements consistent with family III CBDs that bind crystalline cellulose. These include a well conserved calcium-binding site, a putative cellulose-binding surface and a conserved shallow groove of unknown function. The CipC CBD structure is very similar to the previously elucidated family IIIa CBD from the CipA scaffoldin of C. thermocellum, with some minor differences. The CipC CBD structure was also compared with other previously described CBD structures from families IIIc and IV derived from the endoglucanases of Thermomonospora fusca and Cellulomonas fimi, respectively. The possible functional consequences of structural similarities and differences in the shallow groove and cellulose-binding faces among various CBD families and subfamilies are discussed.

    Notizen: Porins form channels allowing the transport of molecules across lipid bilayer membranes. Their structure, location and large number on the bacterial surface lend them multiple functions. Porin loops are potential targets for adhesion to other cells and binding of bactericidal compounds to the surface of Gram-negative bacteria. Variation of the loop structure as a mechanism to escape immune pressure, or modulation of the porin expression in response to the presence of antibiotics, are survival strategies developed by some pathogenic bacteria. Porins may play a significant role as pathogenesis effectors.

    Notizen: The hypothesis that root apical diameter may be used to evaluate root growth potential was tested. Temporal variations in the apical diameter of individual roots of rubber seedlings (Hevea brasiliensis) were studied together with their elongation patterns, using root observation boxes under controlled conditions. This study confirmed the overall positive correlation between apical diameter and growth rale. Moreover, the two parameters, varied in the same way during the life of a given root. For roots with short growth duration, there was a parallel quick decrease in both apical diameter and elongation rate, whereas roots that grew for longer periods showed synchronous fluctuations for both parameters. Since the mean values for the secondary roots within a root system exhibited the same trends, variations in apical diameter and elongation rates should depend on factors influencing the whole root system. When related to shoot rhythmic growth, both apical diameter and elongation rates were depressed during the periods of leaf growth. These effects were enhanced and/or prolonged by shading, hence reinforcing the hypothesis that this development depends on assimilate availability. Such results can be interpreted in terms of a source-sink relationship within the whole plant by considering the apical diameter, representing the size of the meristem related to the number of rneristematic cells, as an indicator of each root's growth potential.
