    Notes: Based on the substrate specificity for 5-lipoxygenase and the known stereochemical course of the reaction, a hypothetical model of the enzyme active site was developed and used to design 2 types of selective inhibitors of 5-lipoxygenase. Both inhibitor types used aromatic rings in place of (Z)-olefins of the substrate and were designed to mimic the nonpolar end of arachidonic acid. One inhibitor type used a carboxylic-acid interaction with the O-binding centre of the enzyme in analogy with known cyclooxygenase inhibitors, whereas a second type employed a hydroxylamine function to interact with a presumed tyrosine or cysteinyl radical predicted to be in the enzyme active site. Selective 5-lipoxygenase inhibitors were 7-(hexyloxy) naphthalene-2-acetic acid (1) and N-methyl;-N(7-propoxynaphthalene-2-ethyl)hydroxylamine (2). Structure-activity relationships for both types of inhibitors are discussed.

    Notes: The chemistry of the 2-aminospiro[indan-1,3′-pyrrolidine]system2-Aminospir[indan-1,3′-pyrrolidines] 1 are easily synthesized from the α-hydroxyiminoketones 6 and 19 (Schemes 2 and 3, and 6 respectively). Removal of the N-acetyl group in the intermediate 8 of the trans series induces transposition to the 3a-aminomethyl-indano[2,1-b]pyrrolidine system. The configurations of all compounds have been determined by 1H-NMR. spectroscopy.

    Description: The Nankai Trough off southwest Japan has an [~]1300 yr historical record of great earthquakes, including the most recent, the A.D. 1944 Tonankai (M = 8.2) earthquake. Evaluation of the activity of an individual submarine fault is difficult when only onland observations are available. Submarine core records can pinpoint individual fault activity. Here we present Integrated Ocean Drilling Program (IODP) Nankai Trough Seismogenic Zone Experiment data from the shallow portion. IODP Expedition 316 drilled and cored several holes in the shallow portion of the offshore Tonankai earthquake area, including sites in the hanging wall of a margin-dominating splay fault that has previously been interpreted to have ruptured coseismically during megathrust earthquakes. X-ray computed tomography scanning revealed that the uppermost core at one site contains repeated occurrences of mud breccia. Radioisotope dating of the uppermost mud breccia indicates a deposition time consistent with the 1944 Tonankai earthquake, suggesting that the mud-breccia layers result from episodic brecciation caused by seismic shaking. Mud brecciation provides a potential new tool to reconstruct ancient earthquake history in subduction zones.

We present a deep neural network that prospectively predicts lineage choice in differentiating primary hematopoietic progenitors using image patches from brightfield microscopy and cellular movement. Surprisingly, lineage choice can be detected up to three generations before conventional molecular markers are observable. Our approach allows identification of cells with differentially expressed lineage-specifying genes without molecular labeling.

    Description: Seepage of methane-dominated hydrocarbons is heterogeneous in space and time, and trigger mechanisms of episodic seep events are not well constrained. It is generally found that free hydrocarbon gas entering the local gas hydrate stability field in marine sediments is sequestered in gas hydrates. In this manner, gas hydrates can act as a buffer for carbon transport from the sediment into the ocean. However, the efficiency of gas hydrate-bearing sediments for retaining hydrocarbons may be corrupted: Hypothesized mechanisms include critical gas/fluid pressures beneath gas hydrate-bearing sediments, implying that these are susceptible to mechanical failure and subsequent gas release. Although gas hydrates often occur in seismically active regions, e.g., subduction zones, the role of earthquakes as potential triggers of hydrocarbon transport through gas hydrate-bearing sediments has hardly been explored. Based on a recent publication (Fischer et al., 2013), we present geochemical and transport/reaction-modelling data suggesting a substantial increase in upward gas flux and hydrocarbon emission into the water column following a major earthquake that occurred near the study sites in 1945. Calculating the formation time of authigenic barite enrichments identified in two sediment cores obtained from an anticlinal structure called “Nascent Ridge”, we find they formed 38-91 years before sampling, which corresponds well to the time elapsed since the earthquake (62 years). Furthermore, applying a numerical model, we show that the local sulfate/methane transition zone shifted upward by several meters due to the increased methane flux and simulated sulfate profiles very closely match measured ones in a comparable time frame of 50-70 years. We thus propose a causal relation between the earthquake and the amplified gas flux and present reflection seismic data supporting our hypothesis that co-seismic ground shaking induced mechanical fracturing of gas hydrate-bearing sediments creating pathways for free gas to migrate from a shallow reservoir within the gas hydrate stability zone into the water column. Our results imply that free hydrocarbon gas trapped beneath a local gas hydrate seal was mobilized through earthquake-induced mechanical failure and in that way circumvented carbon sequestration within the sediment. These findings lead to conclude that hydrocarbon seepage triggered by earthquakes can play a role for carbon budgets at other seismically active continental margins. The newly identified process presented in our study is conceivable to help interpret data from similar sites.

    Description: Several previous studies have shown that submarine mass-movements can profoundly impact the shape of pore water profiles. Therefore, pore water geochemistry and diffusion models were proposed as tools for identifying and dating recent (max. several thousands of years old) mass-transport deposits (MTDs). In particular, sulfate (SO42−) profiles evidentially indicate transient pore water conditions generated by submarine landslides. After mass-movements thatresult in the deposition of sediment packages with distinct pore water signatures,the SO42− profiles can be kink-shaped and evolve into the concave and linear shape with time due to molecular diffusion. Here we present data from the RV METEOR cruise M78/3 along the continental margin off Uruguay and Argentina. SO42− profiles of 15 gravity cores are compared with the respective acoustic facies recorded by a sediment echosounder system. Our results show that in this very dynamic depositional setting, non-steady state profiles occur often, but are not exclusively associated with mass-movements. Three sites that show acoustic indications for recent MTDs are presented in detail. Where recent MTDs are identified,a geochemical transport/reaction model is used to estimate the time that has elapsed since the perturbation of the pore water system and, thus, the timing of the MTD emplacement. We conclude that geochemical analyses are a powerful complementary tool in the identification of recent MTDs and provide a simple and accurate way of dating such deposits.
