    Notes: Summary A structural and ultrastructural investigation of epidermis in the esophageal region of Carinoma tremaphoros is correlated with analysis of burrowing locomotion. The pseudostratified epidermis contains multiciliated epidermal cells, gland cells and granule-containing basal or interstitial cells resting on a well-developed basement membrane. Both circular and longitudinal muscles are present within the epidermis, woven between the cells of this layer. These muscles are well-developed along the anterior region of the worm and, like the subepidermal muscles, are a variation of obliquely striated muscle without continuous Z-rods. Epidermal muscles originate as branches from the underlying mesodermal musculature and cross the basement membrane into the epidermis, confirming earlier observations. These muscles are unrelated to myoepithelial epidermal cells described in other metazoans. This condition is considered a secondary derivation consistent with the Gastraea-Enterocoel theory of bilateral metazoan evolution. Carinoma is a peristaltic burrower. Peristaltic waves are limited to the anterior region of the worm where the diameter of the rhynchocoel is maximal and the bilayered epidermal musculature is strongly developed. Following a consideration of burrowing mechanics in annelids, supernumerary epidermal muscles in Carinoma are interpreted as prerequisites to peristaltic burrowing effectiveness in a non-segmented coelomate.

    Notes: Abstract Nemerteans (phylum Nemertea) have been viewed by mostzoologists as descended from, or closely related to,the flatworms. This view is based mainly on theirsupposedly acoelomate body. Their ancestry, however,is a point of controversy and there is evidence for acoelomate, protostomous origin. Notwithstanding thesedifferent views, most zoologists consider nemerteansto be phylogenetically distant from the chordates.Four authors (Hubrecht, Macfarlane, Jensen, Willmer),however, have postulated that nemerteans instead areclosely related to the chordates and that they sharea most recent common ancestor with the vertebrates. We argue that this view is based on a flawed view ofhomology and of seeing evolution as a series ofprogressions, which has no support in modernevolutionary thinking. Since there are nomorphological synapomorphies supporting aChordata-Nemertea clade, these authors instead guesswhat characteristics in extant nemerteans gave rise tocharacters observed in recent chordates. For example,they propose that the nemertean proboscis sheath hasevolved into the notochord. This is mere speculation,lacking testable propositions and is hence void ofinformation, and thus becomes futile in our view. However, the idea of a nemertean-vertebrate sisterrelationship as such is a testable hypotheses, and wetest it by applying the parsimony criterion to a setof morphological characters, and a set of molecular(the 18S rRNA gene) characters. Both tests reject thehypothesis.

    Description: Resolving the deep relationships of ancient animal lineages has proven difficult using standard Sanger-sequencing approaches with a handful of markers. We thus reassess the relatively well-studied phylogeny of the phylum Nemertea (ribbon worms)—for which the targeted gene approaches had resolved many clades but had left key phylogenetic gaps—by using a phylogenomic approach using Illumina-based de novo assembled transcriptomes and automatic orthology prediction methods. The analysis of a concatenated data set of 2,779 genes (411,138 amino acids) with about 78% gene occupancy and a reduced version with 95% gene occupancy, under evolutionary models accounting or not for site-specific amino acid replacement patterns results in a well-supported phylogeny that recovers all major accepted nemertean clades with the monophyly of Heteronemertea, Hoplonemertea, Monostilifera, being well supported. Significantly, all the ambiguous patterns inferred from Sanger-based approaches were resolved, namely the monophyly of Palaeonemertea and Pilidiophora. By testing for possible conflict in the analyzed supermatrix, we observed that concatenation was the best solution, and the results of the analyses should settle prior debates on nemertean phylogeny. The study highlights the importance, feasibility, and completeness of Illumina-based phylogenomic data matrices.
