    Notes: The sequence 5′-GCGATCGC- 3′, designated HIP1, for highly iterated palindrome, was first identified at the borders of a gene-deletion event and subsequently shown to constitute up to 2.5% of the DNA in some cyanobacteria. It is now reported that HIP1 is polyphyletic, occurring in several distinct cyanobacterial lineages and not defining a clade. HIP1 does not introduce gaps into sequence alignments. It aligns with partial HIP1 sites in related sequences showing that it propagates by nucleotide substitutions rather than insertion. Constructs have been created to determine the frequencies at which deletion events occur between palindromes located within the selectable marker neoDeletion between HIP1 sites was more frequent in Synechococcus PCC 7942 than deletion between control palindromes, 5′-CCGATCGG-3′, designated PAL0. However, this is not due to a recombinase that recognises HIP1 and is peculiar to cyanobacteria because similar deletion frequencies were detected in Escherichia coli. Furthermore, the frequency of deletion of DNA flanked asymmetrically by one HIP1 site and one PAL0 site was less than the frequency of deletion of DNA flanked symmetrically by identical copies of either palindrome. This is consistent with deletion by copy-choice.

    Notes: Genomic rearrangements involving amplification of metallothionein (MT) genes have been reported in metal-tolerant eukaryotes. Similarly, we have recently observed amplification and rearrangement of a prokaryotic MT locus, smt, in cells of Synechococcus PCC 6301 selected for Cd tolerance. Following the characterization of this locus, the altered smt region has now been isolated from a Cd-tolerant cell line, C3.2, and its nucleotide sequence determined. This has identified a deletion within smtB, which encodes a trans-acting repressor of smt transcription. Two identical palindromic octanucleotides (5′-GCGATC-GC-3′) traverse both borders of the excised element. This palindromic sequence is highly represented in the smt locus (7 occurrences in 1326 nucleotides) and analysis of the GenBank/EMBL /DDB J DNA Nucleotide Sequence Data Libraries reveals that this is a highly iterated palindrome (HIP1) in other known sequences from Synechococcus strains (estimated to occur at an average frequency of once every c. 664 bp). HIP1 is also abundant in the genomes of other cyanobacteria. The functional significance of smtB deletion and the possible role of HIP1 in genome plasticity and adaptation in cyanobacteria are discussed.

    Notes: In eukaryotes, metallothioneins (MTs) are involved in cellular responses to elevated concentrations of certain metal ions. We report the isolation and analysis of a prokaryotic MT locus from Synechococcus PCC 7942. The MT locus (smt) includes smtA, which encodes a class II MT, and a divergently transcribed gene, smtB. The sites of transcription initiation of both genes have been mapped and features within the smt operator-promoter region identified. Elevated concentrations of the ionic species of Cd, Co, Cr, Cu, Hg, Ni, Pb and Zn elicited an increase in the abundance of smtA transcripts. There was no detectable effect of elevated metal (Cd) on smtA transcript stability. Sequences upstream of smtA, fused to a promoterless lacZ gene, conferred metal-dependent β-galactosidase activity in Synechococcus PCC 7942 (strain R2-PIM8). At maximum permissive concentrations, Zn was the most potent elicitor in vivo, followed by Cu and Cd with slight induction by Co and Ni. The deduced SmtB polypeptide has similarity to the ArsR and CadC proteins involved in resistance to arsenate/arsenite/antimonite and to Cd, contains a predicted helix-turn-helix DNA-binding motif and is shown to be a repressor of transcription from the smtA operator-promoter.

    Notes: Zinc is essential but toxic in excess. Bacterial metallothionein, SmtA from Synechococcus PCC 7942, sequesters and detoxifies four zinc ions per molecule and contains a zinc finger structurally similar to eukaryotic GATA. The dearth of other reported bacterial metallothioneins has been surprising. Here we describe related bacterial metallothioneins (BmtA) from Anabaena PCC 7120, Pseudomonas aeruginosa and Pseudomonas putida that bind multiple zinc ions with high stability towards protons. Thiol modification demonstrates that cysteine coordinates zinc in all of these proteins. Additionally, 111Cd-NMR, and 111Cd-edited 1H-NMR, identified histidine ligands in Anabaena PCC 7120 BmtA, analogous to SmtA. A related Escherichia coli protein bound only a single zinc ion, via four cysteine residues, with low stability towards protons; 111Cd-NMR and 111Cd-edited 1H-NMR confirmed exclusive cysteine-coordination, and these cysteine residues reacted rapidly with 5,5′-dithiobis-(2-nitrobenzoic acid). 1H-NMR of proteins from P. aeruginosa, Anabaena PCC 7120 and E. coli generated fingerprints diagnostic for the GATA-like zinc finger fold of SmtA. These studies reveal first the existence of multiple bacterial metallothioneins, and second proteins with SmtA-like lone zinc fingers, devoid of a cluster, and designated GatA. We have identified 12 smtA-like genes in sequence databases including four of the gatA type.

    Notes: Zn2+-responsive repressor ZiaR and Co2+-responsive activator CoaR modulate production of P1-type Zn2+- (ZiaA) and Co2+- (CoaT) ATPases respectively. What dictates metal selectivity? We show that Δ ziaΔcoa double mutants had similar Zn2+ resistance to Δzia single mutants and similar Co2+ resistance to Δcoa single mutants. Controlling either ziaA or coaT with opposing regulators restored no resistance to metals sensed by the regulators, but coincident replacement of the deduced cytosolic amino-terminal domain CoaTN with ZiaAN (in ziaR-p ziaA-ziaANcoaT) conferred Zn2+ resistance to ΔziaΔcoa, Zn2+ content was lowered and residual Co2+ resistance lost. Metal-dependent molar absorptivity under anaerobic conditions revealed that purified ZiaAN binds Co2+ in a pseudotetrahedral two-thiol site, and Co2+ was displaced by Zn2+. Thus, the amino-terminal domain of ZiaA inverts the metals exported by zinc-regulated CoaT from Co2+ to Zn2+, and this correlates simplistically with metal-binding preferences; KZiaAN Zn2+ tighter than Co2+. However, Zn2+ did not bleach Cu+-ZiaAN, and only Cu+ co-migrated with ZiaAN after competitive binding versus Zn2+. Bacterial two-hybrid assays that detected interaction between the Cu+-metallochaperone Atx1 and the amino-terminal domain of Cu+-transporter PacSN detected no interaction with the analogous, deduced, ferredoxin-fold subdomain of ZiaAN. Provided that there is no freely exchangeable cytosolic Cu+, restricted contact with the Cu+-metallochaperone can impose a barrier impairing the formation of otherwise favoured Cu+–ZiaAN complexes.

    Notes: Homeostatic systems for essential and non-essential metals create the cellular environments in which the correct metals are acquired by metalloproteins while the incorrect ones are somehow avoided. Cyanobacteria have metal requirements often absent from other bacteria; copper in thylakoidal plastocyanin, zinc in carboxysomal carbonic anhydrase, cobalt in cobalamin but magnesium in chlorophyll, molybdenum in heterocystous nitrogenase, manganese in thylakoidal water-splitting oxygen-evolving complex. This article reviews: an intracellular trafficking pathway for inward copper supply, the sequestration of surplus zinc by metallothionein (also present in other bacteria) and the detection and export of excess cobalt. We consider the influence of homeostatic proteins on selective metal availability.

    Notes: Abstract Suspension cultures of Datura innoxia cells were pulse-labeled with [35S]cysteine, then exposed to Cd to determine whether there is a direct precursor-product relationship amongst the different forms of the Cd-induced polypeptides, poly(γ-glutamylcysteinyl)glycines [(γEC)nG, n=2 to 5]. Degradation of the polypeptides and possible regeneration of the [35S]-labeled glutathione and cysteine pools were also examined. After 2 h of exposure to [35S]cysteine, about 70% of the [35S]cysteine in the soluble fraction of the cell was incorporated into [35S]glutathione before exposure of the cells to Cd. One h after Cd exposure, most of the cellular [35S]glutathione was depleted and label was incorporated into (γEC)nG. Analysis of [35S](γEC)nG by reverse phase HPLC showed no direct precursor-product relationship between the synthesis of the shorter and longer chain forms. However, the rate of synthesis of the different polypeptides was linear for 32 h after Cd exposure. There was no evidence of degradation of [35S](γEC)nG nor was it excreted into the medium within this period. From these results it is suggested that in the presence of Cd, a large pool of (γEC)nG is unavailable for elongation to (γEC)n+1G.

    Notes: Summary This paper reports the (de novo) construction of mutans ofSynechococcus PCC 7942 lacking the repressor (SmtB) of the metallothionein gene,smtA. ThesesmtA +/B − cell are more tolerant to elevated [Zn2+] and [Cd2+] than cells containing an intact metallothionein divergon (smt). Previously selected (by step-wise adaptation) Cd2+-tolerant mutants contain additional copies ofsmtA and possibly other undetected mutations. It is now confirmed that these cells also contain a deletion within ‚all’ copies ofsmtB and hence fail to revert to wild type following subculture in medium which has not been supplemented with Cd2+ or Zn2+. Northern analysis showed enhanced accumulation ofsmtA transcripts, even in the absence of added metal ions in these mutants. An increase in the accumulation of Zn2+ is reported in cells containing an intact metallothionein divergon compared to cells deficient in bothsmtA andsmtB. This supports the assumption that SmtA binds Zn2+ within cyanobacterial cells. We also describe the use of the above mentioned mutants to identify additional factors involeved in the regulation of transcription from thesmtA operator-promoter.
